OXYGEN AND LACTATE MONITORING IN 3D BREAST CANCER ORGANOID CULTURE WITH SENSOR-INTEGRATED MICROFLUIDIC PLATFORM
Abstract
Heterogeneous breast cancer models are essential tools for the development of personalized chemotherapy. Here, we present a platform for 3D cultivation of breast cancer spheroids with embedded electrochemical sensors for highly long-term stable measurement of the metabolic parameters oxygen and lactate in real-time. During the monitored culture of over one week, tumor organoids develop from patient-derived, triple-negative breast cancer stem cells within an extracellular matrix. The transparent, gas-tight platform combines matrix-based 3D cell culture, optical analysis capability and microfluidics. The control of culture conditions and continuous electrochemical metabolic monitoring enhance the information content and quality during drug response experiments and beyond.
