Cellulase enzyme is one of the highest demand enzyme by various industrial fields. Cellulase enzyme produced by various types of microorganisms, including fungi. The production of cellulase enzyme by fungi requires optimum conditions. Thus, an optimization step is needed to increase cellulase enzyme production. This study aimed to determine the optimum incubation time, temperature, and pH for Thielaviopsis ethacetica SLL10 to produce cellulase, and to determine the specific activity of cellulase enzymes in optimum conditions. T. ethacetica SLL10 was grown in potato dextrose agar (PDA) medium and it’s used as a culture stock. Spores were harvested from a 5-day-old culture stock and used as an inoculum. Inoculum containing 5,9x106 spores/mL were inoculated into production medium to determine the growth of fungi and optimum incubation time for cellulase enzyme production. Inoculum was inoculated too into the production medium for optimization in temperature and pH variation during fermentation. The crude extract obtained from each treatment was calculated for enzyme activity so that the optimum incubation time, temperature and pH were known. Enzyme-specific activity was calculated after cellulase was reproduced in conditions of optimum incubation time, temperature and pH. T. ethacetica SLL 10 produced cellulase enzyme maximally at 40°C and pH 5.5 for 10 days of incubation. The specific activity of cellulase enzyme from T. ethacetica SLL 10 reached 3,1578 U/mg under optimum conditions.
