Effect of dimethyl phthalate on kidney and reproductive system in rats

 
 

Abstract


Objective \nTo study the effect of dimethyl phthalate on kidney and reproductive system in rats. \n \n \nMethods \nSixty Wistar clean grade male rats were randomly divided into low dose group, middle dose group, high dose group and control group with 15 rats in each group. Serum creatinine (Cr), urea nitrogen bun (BUN), luteinizing hormone (LH), follicle stimulating hormone (FSH), testosterone (T), cystatin C (cys-C), β-2 microglobulin (β-Mg) were detected in the drug groups after 6 weeks of treatment.The changes of renal histomorphology were observed by HE staining. Single factor analysis of variance was used to compare the two groups. \n \n \nResults \nSerum creatinine and BUN increased significantly in high dose group (F=11.70, 4.16; P<0.05), while Cystatin C (Cys-C) and β-2-microglobulin (β-Mg) increased significantly in low, medium and high dose groups. (F=31.16, 8.05; P<0.05). Serum testosterone level decreased in high dose group, LH level increased in low dose group, but decreased in high dose group and middle dose group, FSH level increased in low, medium and high dose group (F=4.82, 59.34, 6.91; P<0.05). There was no obvious change in renal tissue structure in the control group and low dose group, while the glomerular atrophy occurred in the kidney in the middle dose group, and the glomerular atrophy, degeneration and necrosis occurred in the high dose group. \n \n \nConclusion \nDimethyl phthalate can induce the changes of reproductive endocrine hormone level and glomerular lesion in rats. \n \n \nKey words: \nDimethyl phthalate;\xa0Urinary system;\xa0Toxicity;\xa0Rat
