Effect of divergence in residual feed intake on feeding behavior, blood metabolic variables, and body composition traits in growing beef heifers

This study examined the relationship of feed efficiency and performance with feeding behavior, blood metabolic variables, and various body composition measurements in growing beef heifers. Individual DMI and growth were measured in yearling Limousin x Holstein-Friesian heifers [n = 86; initial BW = 191.8 (SD = 37) kg] fed a TMR diet comprising 70:30 concentrate:corn silage on a DM basis (ME of 2.65 Mcal/kg of DM; DM of 580 g/kg) for 82 d. Meal duration (min/d) and meal frequency (events/d) were calculated for each animal on a daily basis using an Insentec computerized feeding system. Physical measurements as well as ultrasonic fat and muscle depths were recorded on 3 equally spaced occasions during the experimental period. Blood samples were collected by jugular venipuncture on 4 equally spaced occasions and analyzed for plasma concentrations of IGF-I, insulin, leptin, and various metabolites. Phenotypic residual feed intake (RFI) was calculated for all animals as the residuals from a multiple regression model regressing DMI on ADG and midtest BW(0.75). Overall, ADG, DMI, feed conversion ratio (FCR), and RFI were 1.51 (SD = 0.13), 6.74 (SD = 0.99), 4.48 (SD = 0.65), and 0.00 (SD = 0.48) kg/d, respectively. Residual feed intake was positively correlated with DMI (r = 0.47) and FCR (r = 0.46), but not with ADG or midtest BW. Positive correlations (ranging from r = 0.27 to r = 0.63) were estimated between ultrasonic measures of final lumbar fat and lumbar fat accretion over the test period and DMI, FCR, and RFI. The inclusion of gain in lumbar fat to the base RFI model increased R(2) (0.77 vs. 0.80) value for the degree of variation in DMI not explained by midtest BW and ADG alone. The Pearson rank correlation between RFI and carcass-adjusted RFI (RFI(c)) was high (r = 0.93). From the plasma analytes measured, NEFA (r = -0.21; P < 0.05) and beta-hydroxybutyrate (r = 0.37; P < 0.05) concentrations were correlated with RFI. Plasma leptin (r = 0.48), glucose:insulin (r = -0.23), NEFA (r = -0.32), and beta-hydroxybutyrate (r = 0.25) were associated with FCR. However, systemic IGF-I and insulin were unrelated (P > 0.05) to any measure of feed efficiency. The feeding behavior traits of eating rate, daily feeding events, and nonfeeding events were positively correlated (P < 0.05) with RFI and RFI(c). This multifactorial study provides new information on some of the biological processes responsible for variation in feed efficiency in beef cattle.

The effect of elevated progesterone and pregnancy status on mRNA expression and localisation of progesterone and oestrogen receptors in the bovine uterus

To investigate the effects of pregnancy or post-ovulatory progesterone (P(4)) supplementation on the expression of oestrogen and P(4) receptors (ESRs and PGRs) in the bovine uterus, heifers (n=263) were randomly assigned to the following treatments: i) cyclic, normal P(4); ii) cyclic, high P(4); iii) pregnant, normal P(4); and iv) pregnant, high P(4) on days 5, 7, 13 and 16 of pregnancy/oestrous cycle. Elevated P(4) was achieved through P(4)-releasing intravaginal device insertion on day 3 after oestrus, resulting in increased concentrations from day 3.5 to 8 (P<0.05) in the high groups than in the normal groups. Irrespective of treatment, PGR and ESR1 mRNA expressions were highest on days 5 and 7 and decreased on day 13 (P<0.05), while ESR2 mRNA expression increased on day 7 (P<0.05) and similar levels were maintained within the normal P(4) groups subsequently. Expression in the high P(4) groups decreased on day 13 (P<0.05). PGR-AB and PGR-B protein expressions were high in the luminal and superficial glands on days 5 and 7, but by day 13, expression had declined to very low or undetectable levels and high P(4) concentration tended to decrease or decreased significantly (P<0.05) the expression in these regions on days 5 and 7. ESR1 protein expression was high, with no treatment effect. ESR2 protein was also highly expressed, with no clear effect of treatment. In conclusion, early post-ovulatory P(4) supplementation advances the disappearance of PGR protein from the luminal epithelium on days 5 and 7, and decreases ESR2 mRNA expression during the mid-luteal phase, but has no effect on PGR or ESR1 mRNA expression.

Genome-wide associations for milk production and somatic cell score in Holstein-Friesian cattle in Ireland

BackgroundContemporary dairy breeding goals have broadened to include, along with milk production traits, a number of non-production-related traits in an effort to improve the overall functionality of the dairy cow. Increased indirect selection for resistance to mastitis, one of the most important production-related diseases in the dairy sector, via selection for reduced somatic cell count has been part of these broadened goals. A number of genome-wide association studies have identified genetic variants associated with milk production traits and mastitis resistance, however the majority of these studies have been based on animals which were predominantly kept in confinement and fed a concentrate-based diet (i.e. high-input production systems). This genome-wide association study aims to detect associations using genotypic and phenotypic data from Irish Holstein-Friesian cattle fed predominantly grazed grass in a pasture-based production system (low-input).ResultsSignificant associations were detected for milk yield, fat yield, protein yield, fat percentage, protein percentage and somatic cell score using separate single-locus, frequentist and multi-locus, Bayesian approaches. These associations were detected using two separate populations of Holstein-Friesian sires and cows. In total, 1,529 and 37 associations were detected in the sires using a single SNP regression and a Bayesian method, respectively. There were 103 associations in common between the sires and cows across all the traits. As well as detecting associations within known QTL regions, a number of novel associations were detected; the most notable of these was a region of chromosome 13 associated with milk yield in the population of Holstein-Friesian sires.ConclusionsA total of 276 of novel SNPs were detected in the sires using a single SNP regression approach. Although obvious candidate genes may not be initially forthcoming, this study provides a preliminary framework upon which to identify the causal mechanisms underlying the various milk production traits and somatic cell score. Consequently this will deepen our understanding of how these traits are expressed.

Methane emissions, body composition, and rumen fermentation traits of beef heifers differing in residual feed intake.

This study examined the relationship of residual feed intake (RFI) and performance with methane emissions, rumen fermentation, and digestion in beef heifers. Individual DMI and growth performance were measured for 22 Simmental heifers (mean initial BW 449 kg, SD = 46.2 kg) offered grass silage ad libitum for 120 d. Ultrasonically scanned muscle and fat depth, BCS, muscularity score, skeletal measurements, blood variables, rumen fermentation (via stomach tube), and total tract digestibility (indigestible marker) were measured. Methane production was estimated using the sulfur hexafluoride tracer gas technique over two 5-d periods beginning on d 20 and 75 of the RFI measurement period. Phenotypic RFI was calculated as actual DMI minus expected DMI. The residuals of the regression of DMI on ADG and midtest metabolic body weight, using all heifers, were used to compute individual RFI coefficients. Heifers were ranked by RFI and assigned to low (efficient), medium, or high (inefficient) groupings. Overall ADG and DMI were 0.58 kg (SD = 0.18) and 7.40 kg (SD = 0.72), respectively. High-RFI heifers consumed 9 and 15% more (P < 0.05) than medium- and low-RFI groups, respectively. Body weight, growth, skeletal, and composition traits did not differ (P > 0.05) between low- and high-RFI groups. High-RFI heifers had higher concentrations of plasma glucose (6%) and urea (13%) and lower concentrations of plasma creatinine (9%) than low-RFI heifers (P < 0.05). Rumen pH and apparent in vivo digestibility did not differ (P > 0.05) between RFI groups, although acetate:propionate ratio was lowest (P = 0.07) for low-RFI (3.5) and highest for high-RFI (4.6) heifers. Methane production expressed as grams per day or grams per kilogram metabolic body weight was greater (P < 0.05) for high (297 g/d and 2.9 g/kg BW0.75) compared with low (260 g/d and 2.5 g/kg BW0.75) RFI heifers, with medium (275 g/d and 2.7 g/kg BW0.75) RFI heifers being intermediate. Regression analysis indicated that a 1 kg DM/d increase in RFI was associated with a 23 g/d increase (P = 0.09) in methane emissions. Results suggest that improved RFI will reduce methane emissions without affecting productivity of growing beef cattle.

Predictive value of bovine follicular components as markers of oocyte developmental potential

The follicle is a unique micro-environment within which the oocyte can develop and mature to a fertilisable gamete. The aim of this study was to investigate the ability of a panel of follicular parameters, including intrafollicular steroid and metabolomic profiles and theca, granulosa and cumulus cell candidate gene mRNA abundance, to predict the potential of bovine oocytes to develop to the blastocyst stage in vitro. Individual follicles were dissected from abattoir ovaries, carefully ruptured under a stereomicroscope and the oocyte was recovered and individually processed through in vitro maturation, fertilisation and culture. The mean (±s.e.m.) follicular concentrations of testosterone (62.8±4.8 ngmL(-1)), progesterone (616.8±31.9 ngmL(-1)) and oestradiol (14.4±2.4 ngmL(-1)) were not different (P>0.05) between oocytes that formed (competent) or failed to form (incompetent) blastocysts. Principal-component analysis of the quantified aqueous metabolites in follicular fluid showed differences between oocytes that formed blastocysts and oocytes that degenerated; l-alanine, glycine and l-glutamate were positively correlated and urea was negatively correlated with blastocyst formation. Follicular fluid associated with competent oocytes was significantly lower in palmitic acid (P=0.023) and total fatty acids (P=0.031) and significantly higher in linolenic acid (P=0.036) than follicular fluid from incompetent oocytes. Significantly higher (P<0.05) transcript abundance of LHCGR in granulosa cells, ESR1 and VCAN in thecal cells and TNFAIP6 in cumulus cells was associated with competent compared with incompetent oocytes.

Pivotal Role for Monocytes/Macrophages and Dendritic Cells in Maternal Immune Response to the Developing Embryo in Cattle

ABSTRACT In mammals, successful pregnancy is dependent in part on the adaptation or regulation of the maternal immune system to prevent the rejection of the embryonic semiallograft. A modification in Th cell function and secretion is a requirement for the establishment and maintenance of pregnancy. Although there is strong evidence from studies in humans and mice linking successful pregnancy with the predominance of Th2-type immunity, the situation in cattle remains unclear. This study describes the characterization of the immune response of the bovine maternal endometrium to the presence of a developing embryo, with specific emphasis on the macrophage and dendritic cell populations and associated factors, using quantitative real-time PCR, in situ hybridization, and immunohistochemistry. Furthermore, in vivo and in vitro models were developed to investigate the potential role of progesterone and interferon-tau (IFNT) in the regulation of these immune factors. There was a marked increase in the population of CD14+ cells and CD172a-CD11c+ cells in the endometrium in response to pregnancy, which was paralleled by increased mRNA expression of a number of non-Th-associated factors, including IL12B and IL15, and downregulation of IL18. In addition, we identified several novel IFNT- and progesterone-regulated factors, including IL12B, MCP1, MCP2, PTX3, RSAD2, and TNFA, whose regulation may be critical to pregnancy outcome. Our findings give center stage to non-Th cells, such as monocytes/macrophages and dendritic cells, in the bovine immune response to the semiallogenic embryo. In conclusion, we propose that in cattle, successful pregnancy establishment is associated with a dramatic regulation of the cytokine network, primarily by endometrial monocytes/macrophages and dendritic cells.

The genetic origin and history of speed in the Thoroughbred racehorse

Selective breeding for speed in the racehorse has resulted in an unusually high frequency of the C-variant (g.66493737C/T) at the myostatin gene (MSTN) in cohorts of the Thoroughbred horse population that are best suited to sprint racing. Here we show using a combination of molecular- and pedigree-based approaches in 593 horses from 22 Eurasian and North-American horse populations, museum specimens from 12 historically important Thoroughbred stallions (b.1764-1930), 330 elite-performing modern Thoroughbreds and 42 samples from three other equid species that the T-allele was ancestral and there was a single introduction of the C-allele at the foundation stages of the Thoroughbred from a British-native mare. Furthermore, we show that although the C-allele was rare among the celebrated racehorses of the 18th and 19th centuries, it has proliferated recently in the population via the stallion Nearctic (b.1954), the sire of the most influential stallion of modern time, Northern Dancer (b.1961).

Genetic parameters for cattle price and body weight from routinely collected data at livestock auctions and commercial farms

Beef outputs from dairy farms make an important contribution to overall profitability in Irish dairy herds and are the sole source of revenue in many beef herds. The aim of this study was to estimate genetic parameters for animal BW and price across different stages of maturity. Data originated from 2 main sources: price and BW from livestock auctions and BW from on-farm weighings between 2000 and 2008. The data were divided into 4 distinct maturity categories: calves (n = 24,513), weanlings (n = 27,877), postweanlings (n = 23,279), and cows (n = 4,894). A univariate animal model used to estimate variance components was progressively built up to include a maternal genetic effect and a permanent environmental maternal effect. Bivariate analyses were used to estimate genetic covariances between BW and price per animal within and across maturity category. Direct heritability estimates for price per animal were 0.34 ± 0.03, 0.31 ± 0.05, 0.19 ± 0.04, and 0.10 ± 0.04 for calves, weanling, postweanlings, and cows, respectively. Direct heritability estimates for BW were 0.26 ± 0.03 for weanlings, 0.25 ± 0.04 for postweanlings, and 0.24 ± 0.06 for cows; no BW data were available on calves. Significant maternal genetic and maternal permanent environmental effects were observed for weanling BW only. The genetic correlation between price per animal and BW within each maturity group varied from 0.55 ± 0.06 (postweanling price and BW) to 0.91 ± 0.04 (cow price and BW). The availability of routinely collected data, along with the existence of ample genetic variation for animal BW and price per animal, facilitates their inclusion in Irish dairy and beef breeding objectives to better reflect the profitability of both enterprises.

Longitudinal study of the effect of rubber slat mats on locomotory ability, body, limb and claw lesions, and dirtiness of group housed sows

This study evaluated the influence of floor type on sow welfare with particular focus on lameness, claw lesions (CL), and injuries. The study used 164 gilts housed in groups of 8 from AI to 110 d of pregnancy in pens with concrete (n = 84) slatted floor left uncovered or covered by 10-mm rubber slat mats (n = 80) through 2 parities. Lameness (0 = normal to 5 = severe), limb (0 = normal to 6 = severe) and body (0 = normal to 5 = severe) lesions, and manure on the body (MOB; score 0 to 2) were recorded at AI, 24 to 72 h postmixing, between 50 and 70 d of pregnancy, and 2 wk before farrowing. Claw lesions (score 0 = normal to 3 = severe) were recorded at AI and between 50 and 70 d of pregnancy. The dirtiness and wetness of the floors was scored weekly (score 0 = clean to 4 = >75% of the pen soiled/wet). Data from the first and second parities were analyzed separately. Sows were categorized as nonlame (score ≤ 1) or lame (score ≥ 2). Median (M(e)) scores were calculated for CL and body and limb lesions and were classified as less than or equal to the median or greater than the median lesion scores. Sows on rubber slat mats had a reduced risk of lameness during both parities (P < 0.01) compared with sows on concrete. They also had an increased risk of scores greater than the median for toe overgrowth (M(e) = 2 and M(e) = 3 in the first and second parity, respectively) and heel sole crack (HSC; M(e) = 3) during both parities (P < 0.01) and for cracks in the wall (CW; M(e) = 4) and white line damage (WL; M(e) = 4; P < 0.01) in the first and second parity, respectively. There was a reduced risk of lameness in sows with scores greater than the median for HSC (P = 0.05) in the first parity and WL (M(e) = 3; P < 0.01) and CW (M(e) = 3; P < 0.05) in the second parity. Wounds (M(e) = 3) and severe lesions (M(e) = 0) on the limbs with scores greater than the median were associated with an increased risk of lameness (P < 0.01) in the first and second parity, respectively. Sows on rubber slat mats had a reduced risk of scores greater than the median for swellings (M(e) = 4) and wounds (P < 0.01) during both parities. Pens with rubber slat mats were dirtier than uncovered pens (P < 0.01); however, there was no association between MOB and flooring type. There was also no association between body lesion score and flooring type. In this study, CL were not associated with an increased risk of lameness. Therefore, even though rubber slat mats were associated with an increased risk of CL, they improved the welfare of group housed sows by reducing the risk of lameness and limb lesions.

A genome-wide association study for somatic cell score using the Illumina high-density bovine beadchip identifies several novel QTL potentially related to mastitis susceptibility

Mastitis is an inflammation-driven disease of the bovine mammary gland that occurs in response to physical damage or infection and is one of the most costly production-related diseases in the dairy industry worldwide. We performed a genome-wide association study (GWAS) to identify genetic loci associated with somatic cell score (SCS), an indicator trait of mammary gland inflammation. A total of 702 Holstein-Friesian bulls were genotyped for 777,962 single nucleotide polymorphisms (SNPs) and associated with SCS phenotypes. The SCS phenotypes were expressed as daughter yield deviations (DYD) based on a large number of progeny performance records. A total of 138 SNPs on 15 different chromosomes reached genome-wide significance (corrected p-value ≤ 0.05) for association with SCS (after correction for multiple testing). We defined 28 distinct QTL regions and a number of candidate genes located in these QTL regions were identified. The most significant association (p-value = 1.70 × 10−7) was observed on chromosome 6. This QTL had no known genes annotated within it, however, the Ensembl Genome Browser predicted the presence of a small non-coding RNA (a Y RNA gene) in this genomic region. This Y RNA gene was 99% identical to human RNY4. Y RNAs are a rare type of non-coding RNA that were originally discovered due to their association with the autoimmune disease, systemic lupus erythematosus. Examining small-RNA sequencing (RNAseq) data being generated by us in multiple different mastitis-pathogen challenged cell-types has revealed that this Y RNA is expressed (but not differentially expressed) in these cells. Other QTL regions identified in this study also encoded strong candidate genes for mastitis susceptibility. A QTL region on chromosome 13, for example, was found to contain a cluster of β-defensin genes, a gene family with known roles in innate immunity. Due to the increased SNP density, this study also refined the boundaries for several known QTL for SCS and mastitis.
