Solid support synthesis of 2,4-disubstituted thiazoles and aminothiazoles

Abstract 3-Iodobenzoic acid was loaded on Rink amide resin. Pd(0) coupling reaction with tributyl(1-ethoxyvinyl)tin followed by bromination using NBS gave an α-bromoketone bound to a solid support. Condensation of thioamide or thiourea, followed by TFA cleavage from the resin, gave 2,4-disubstituted thiazoles or 2-aminothiazoles, respectively. The primary amine of aminothiazole was treated with various acyl or sulfonyl chlorides to provide 2-substituted thiazole analogs.

Solid-phase synthesis of imidazo[1,2-a]pyridines and imidazo[1,2-a]pyrimidines ☆

The synthesis of imidazo[1,2-a]pyridine and imidazo[1,2-a]pyrimidine derivatives by condensation between an α-bromoketone bound to solid support and various 2-aminopyridine or 2-aminopyrimidine derivatives was described. Either an acid labile linker or a base labile linker was used in this study.

Synthesis of benzo[5′,6′]cyclohepta[4,5]pyrrolo[2,3-b]pyridin-12-one

Abstract The synthesis of a 7-azaindole derivative 8 with potential antitumoral activity is described starting from pyrrolo[2,3- b ]pyridine; regioselective lithiation/methylation of alkyl 7-azaindole-3-car☐ylates 4 afforded the 2-methyl derivatives 5 . Demethylation of ester 6a using BBr 3 afforded the acid 7 , which is cyclised into the tetracyclic derivative 8 .
