This work aimed to study the possible relationships among the magnitude of abomasal worm burden and the proliferation of globular leucocytes and mucosal mast cells in the abomasal mucosa, and the white blood cell count. Eighteen Suffolk × Greyface lambs were infected with Haemonchus contortus, and 12 were kept free of nematodes. Blood samples were collected on days 0, 30, and 57 post-infection (p.i.) for leucogram determination. At day 62, all animals were euthanized to count the total number of nematodes recovered in the abomasum and to count the number of mucosal mast cells and globular leucocytes. On day 57, higher levels of parasitism corresponded to lower leucocyte counts. The infected groups had lower lymphocyte counts throughout the experimental period. Animals with higher numbers of parasites had lower neutrophil and eosinophil counts on day 57. The lower the worm burden, the greater the number of mucosal mast cells (r=-0.85; p<0.01) and globular leucocytes (r=-0.87, p<0.01) observed. The sheep most resistant to haemochosis had greater peripheral and tissue cellular immune responses.

The aim of this study was to evaluate the nitric oxide (NO) levels, and oxidative and antioxidant markers of lambs experimentally and naturally infected by Haemonchus contortus, and its relation to lesions in the abomasum. For experimental study, a total of 14 healthy lambs were divided into two groups with seven animals each. Group A represented the uninfected animals (control), and Group B was formed by infected animals with 15,000 larvae of H. contortus. Blood was collected on days 15, 45, and 75 post-infection (PI) to obtain serum for biochemical analysis: lactate dehydrogenase (LDH), nitrite/nitrate (NOx), advanced oxidation protein products (AOPP), and ferric reducing ability of plasma (FRAP). Parasitological stool examination (eggs per gram of feces--EPG) was performed on days 15, 45, and 75 PI to verify the evolution of the infection. On day 15 PI EPG was negative, but on days 45 and 75 PI the EPG was positive for animals from Group B. In the three periods evaluated it was observed an increase of LDH levels in serum of lambs infected with gastrointestinal nematodes, but on the other hand NOx levels were reduced on the same periods in infected animals. The AOPP and FRAP levels did not differ between groups on days 15 and 45 PI, but increased significantly on day 75 PI in infected lambs. The same variables were studied in 10 lambs naturally infected with helminths, where more than 97% corresponded to H. contortus (hematocrit and EPG values were 18.8 ± 2.5% and 7120 ± 2940, respectively). Similar to the experimental study, the levels of NOx reduced, and the levels of LDH, FRAP, and AOPP increased in serum of this animal associated inflammatory infiltrate in the mucosa of the abomasum. Therefore, during the infection by H. contortus it was observed alterations in oxidative markers, indicators of cell lesion confirmed by histological examination of the abomasum, and consequently there were changes in antioxidant levels, with the purpose of cell protection. We also conclude that helminth infection interferes with the nitric oxide metabolism.

The aim of this study was to evaluate the purine levels of lambs experimentally infected with Haemonchus contortus. A total of 12 healthy lambs were divided into two groups, composed of 6 animals each: Group A represented the healthy animals (uninfected), while in Group B the animals were infected with 15 000 larvae of H. contortus. Blood was drawn on days 15, 45 and 75 post-infection (PI) in order to perform the purine analysis (ATP, ADP, AMP, adenosine, inosine, hypoxanthine, xanthine and uric acid) by high pressure liquid chromatography (HPLC) in serum. On day 15 PI a significant (P<0·05) increase in the levels of ATP and inosine was observed in the infected animals, unlike the levels of ADP, adenosine, xanthine and uric acid which were reduced. On day 45 PI a significant (P<0·05) increase in the ATP and xanthine levels in infected animals was observed, contrasting with reduced levels of ADP and uric acid. Finally, on day 75 PI an increase occurred in the levels of ATP, adenosine and hypoxanthine in infected lambs, concomitant with a reduction in the levels of ADP and uric acid (P<0·05). These changes in purine levels may influence the inflammatory process and the pathological events.

The aim of this study was to evaluate the ecto-adenosine deaminase (E-ADA) activity in erythrocytes of lambs experimentally infected with Haemonchus contortus, correlating it with the degrees of anemia of the experimental animals. A total of 14 healthy lambs, with negative fecal exam for parasites, were to carry out the present study. They were divided into two groups, composed by seven animals: Group A represented the healthy animals (uninfected), while in Group B the animals were infected with 15,000 larvae of H. contortus. Blood was drawn on the days 15, 45 and 75 post-infection (PI) in order to perform the hematological analysis, as well as the mensuration of E-ADA activity in erythrocytes. Parasitological stool exam were performed on the same days mentioned above to follow up the evolution of the infection, as well to determine the number of eggs per gram of feces (EPG). On day 15PI, the animals presented negative EPG and there was not significant (P>0.05) difference between groups in relation to E-ADA activity and hematologic parameters. Animals in Group B had positive EPG for helminths on days 45 and 75 PI, accompanied by varying degrees of anemia, when compared to Group A. At the same periods E-ADA activity was significantly (P<0.05) increased in the erythrocytes of animals of Group B when compared with the not-infected ones. Statistically, there was a negative correlation (P<0.01) between activity E-ADA in erythrocytes and hematocrit on days 45 (r = -0.76) and 75 (r = -0.85)PI. Based on these results and in the scientific literature, it is possible to conclude that the E-ADA may participate on mechanisms related with the pathogenesis and host response against anemia caused by H. contortus.

Haemonchus contortus (order Strongylida) is a common parasitic nematode infecting small ruminants and causing significant economic losses worldwide. It induces innate and adaptive immune responses, which are essential for the clearance of this nematode from the host. Ecto-adenosine deaminase (E-ADA) is an enzyme that plays an important role in the immune system, while Zinc (Zn) has been found playing a critical role in E-ADA catalysis. Therefore, the aim of this study was to assess the effect of Zn supplementation on E-ADA activity in serum of lambs experimentally infected with H.contortus. To reach this purpose 28 male lambs (in average 25 kg) were used. The animals were divided into four groups: A and B composed of healthy animals (uninfected); C and D, infected with H.contortus. Groups B and D were supplemented with Zn Edetate, subcutaneously with 3 mg kg of live weight, on days 11 and 25 post-infection (PI). Blood and fecal samples were collected on the days 11, 25 and 39 PI, in order to assess hematocrit, seric E-ADA, and eggs per gram (EPG) counting, respectively. The animals of groups C and D showed severe hematocrit reduction (days 25 and 39 PI) and were EPG positive (days 11, 25 and 39 PI). On day 41 PI, three animals each group were subjected to necropsy. This procedure showed that animals of groups A and B did not have helminths in abomasum and intestines, while H.contortus were observed in groups C (5782.5 ± 810.9) and D (6185.0 ± 150.0). Infected and untreated animals (group C) showed a reduction in E-ADA activity, but this was not observed when the animals were supplemented with Zn (Group D). Therefore, based on our results, it was possible to observe that Zn supplementation exercised a positive effect on E-ADA activity in lambs infected with H.contortus, and did not allow a reduction in E-ADA activity, as occurred in the group infected and without supplementation. However, Zn supplementation was not able to prevent the worm burden.

The aim of this study was to evaluate the effects of selenium and copper on oxidative stress and its performance in lambs experimentally infected with Haemonchus contortus. Twenty-eight five-months old lambs were experimentally infected by the oral route with 5000 third-stage infective larvae and allocated into four groups, i.e., untreated animals, animals treated intramuscularly with sodium selenite (0.2 mg kg(-1)), animals treated subcutaneously with copper (3.5 mg kg(-1)), and animals treated with sodium selenite (IM; 0.2 mg kg(-1)) and copper (SC; 3.5 mg kg(-1)). These animals received oat hay (Avena sativa) and commercial concentrate, totaling 15% of crude protein, 30% being derived from oat hay and 70% of the concentrate. Lipid peroxidation, antioxidant enzymes, eggs per gram of feces (EPG) and body weight were assessed on the day of infection and after 20, 40, 60 and 80 days post-infection. The number of H. contortus adults was assessed at the end of the experiment. The selenium associated or not with copper reduced the effects of oxidative stress caused by infection. The groups supplemented with copper had increased body weight, and the combination of these two minerals reduced the EPG and number of H. contortus adults in lambs. The use of selenium associated with copper may help the control of infection by H. contortus.

The aim of the present study was to assess the effects of zinc edetate on the oxidative stress of lambs infected by Haemonchus contortus. Twenty-four lambs were allocated into four groups: Group I--uninfected animals; Group II--uninfected animals treated subcutaneously with zinc edetate; Group III--animals infected by H. contortus and Group IV--animals infected and treated. The oxidative stress index (OSI) and the eggs per gram of feces (EPG) were assessed after 10, 17, 24, 31 and 38 days post-infection. Based on the EPG and the quantity of adult H. contortus, the infection did not differ between groups III and IV. Zinc edetate reduced the OSI in Group IV in relation to Group I after 24 days post-infection, and in relation to group III after 31 days post-infection. Treatment with zinc edetate could help reduce the oxidative stress induced by H. contortus in lambs.

The aim of this study was to assess the effects of iron supplementation on oxidative stress and on the activity of the adenosine deaminase (ADA) in rats experimentally infected by Trypanosoma evansi. For this purpose, 20 rats were divided into four experimental groups with five animals each as follows: groups A and B were composed by healthy animals, while animals from groups C and D were infected by T. evansi. Additionally, groups B and D received two subcutaneous doses of iron (60 mg kg(-1)) within an interval of 5 days. Blood samples were drawn on day 8 post infection in order to assess hematological and biochemical variables. Among the main results are: (1) animals from group C showed reduced erythrogram (with tendency to anemia); however the same results were not observed for group D; this might be a direct effect of free iron on trypanosomes which helped to reduce the parasitemia and the damage to erythrocytes caused by the infection; (2) iron supplementation was able to reduce NOx levels by inhibiting iNOS, and thus, providing an antioxidant action and, indirectly, reducing the ALT levels in groups Band D; (3) increase FRAP levels in group D; (4) reduce ADA activity in serum and erythrocytes in group C; however, this supplementation (5) increased the protein oxidation in groups B and D, as well as group C (positive control). Therefore, iron showed antioxidant and oxidant effects on animals that received supplementation; and it maintained the activity of E-ADA stable in infected/supplemented animals.

The aim of this study was to evaluate the acetylcholinesterase (AChE) activity in lymphocytes of lambs experimentally infected by Haemonchus contortus. A total of 14 healthy lambs were used, divided into two groups of seven animals each. Group A (negative control) represented the uninfected animals, and Group B (positive control) was formed by animals infected with 15,000 larvae of H. contortus. Blood was drawn on the days 15, 45 and 75 post-infection (PI) in order to perform the white blood cells (WBC) count, as well as the evaluation of AChE activity in lymphocytes. Parasitological stool exam (eggs per gram of feces - EPG) was performed on the same days to follow up the evolution of the infection. On day 15 PI it was verified negative EPG; however, on days 45 and 75 PI it was observed positive EPG only in the animals of group B. In the three evaluated periods was observed a lower number of leukocytes, associated with decreased lymphocytes and neutrophils in lambs infected by this gastrointestinal nematodes. Lambs infected with H. contortus showed significant (P<0.01) lower AChE activity in lymphocytes compared uninfected. Statistically, there was a positive correlation (P<0.05) between AChE activity in lymphocytes and number of lymphocytes (r=0.69). The lymphocytes are cells with direct participation in the cholinergic system; therefore, based on these results, it can be concluded that the experimental infection with H. contortus influences the number of lymphocytes, and consequently the AChE activity in these cells.

Two experiments were performed in order to determine the toxic effects of varying doses of aflatoxins in calves. Clinical, productive and pathologic aspects of affected calves were considered. In the first experiment, nine 2 to 4-month-old calves Holstein Friesian calves were fed, for two months, daily amounts corresponding to 1.5% of their body weight of a ration containing 500±100 ppb of aflatoxins. Three calves of similar age and weight were used as controls and, except for being a ration free of aflatoxins, were kept in the same condition as the treated calves. In the second experiment, three 4-5-month old Holstein Friesian calves, were orally fed daily small parcels of a concentrate of aflatoxins diluted in 500 ml of water corresponding to 1,250, 2,500 e 5,000 ppb of B1 aflatoxin (AFB1). A male 4-month-old Holstein Friesian calf was used as control. During all the experimental period of the first experiment, the weight gain of the calves receiving AFB1 was equivalent to that of the control group. In the first experiment no differences were observed between treated and control calves when the values of serum activity of aspartate transaminase (AST), serum albumin (SA), total serum protein (TP), and PVC, determined weekly, were compared. However there was a significant difference between treated and control groups in the serum activities of alkaline phosphatase (AP) and gamma glutamyl transferase when the serum sampled on the 63th day of the experiment was considered. During the whole experimental period and up to three weeks after the final of the experiment, no clinical signs or histopathological changes associated with the consumption of aflatoxins were observed in any of the calves of the first experiment. In the second experiment, clinical signs observed in three treated calves included loss of appetite, decrease in weight gain, and loss of weight. Jaundice, intermittent diarrhea, tenesmus and apathy were only observed in the calf receiving 5,000 ppb of AFB1. Due to these clinical signs the calf was euthanized. Increased activity of AF and GGT were observed in all the calves of the treated group during most part of the experimental period. A marked drop in the serum levels of SA was observed in the serum sampled on the 49o day of the experiment in the calf receiving the largest dose of aflatoxin. No changes were observed regarding PCV, TP, total bilirubin, direct bilirubin and in the serum activity of AST in any of the calves of the second experiment. Histopathological changes in intoxicated calves included bile duct proliferation, cytoplasmic vacuolar hepatocelular degeneration consistent with hepatocelular deposit of lipids, periportal to bridging fibrosis, megalocytosis, subendothelial edema and fibrosis in terminal hepatic veins. Necropsy findings in the euthanatized calf which receive de largest doses of AFB1 included slight enlargement of the liver which was firm and diffusely light-yellow, mild ascites, and edema of the mesentery and of abomasal folds. Data stemmed from these two experiments allow to conclude that AFB1 doses of 500±100 in the ration do not cause pathologic changes or decrease in productivity in calves kept in experimental conditions, but can be associated to minimal serum biochemistry; while AFB1 doses of 1.250, 2.500 e 5.000 ppb in the ration cause chronic hepatic disease in calves in kept in experimental conditions.
