Inhibition of mouse and rat lymphoproliferation by gangliosides

Our previous study have demonstrated that Glycosphingolipids (GSLs) have an immunosuppressive effect on murine lymphoproliferation and IL-2 production. In the present study we examined the effect of a pool of Gangliosides (Gang) on spleen lymphocyte proliferation from either isogeneic strains of Wistar rats or BALB/c mice. Two hundred-fifty grams adult female isogeneic Wistar rats and 8-week-old BALB/c mice were used. The animals were sacrificed and the spleen harvested aseptically for cellular assays. Spleen cells suspensions were obtained by homogenization in RPMI 1640 with a loose tissue grinder. After washing, the cells were suspended in RPMI 1640 supplemented. Cell viability was measured by Trypan blue exclusion. Cells were cultured in triplicate using increasing concentrations of Gang (1; 2; 5; 10; 15; 20 mg/well) and in the presence of Concanavalin A. The cells were incubated for 48 hours and were pulsed with [3H] thymidine 18 hours prior to harvesting on glass fiber paper for counting in a b-counter. Data were presented as rate of inhibition, as previously described. At concentrations 1 and 2 mg/well, Gang stimulated lymphoproliferation (30% and 50%, rats and mice respectively), while at concentration from 5 to 20 mg/well an increasing inhibition was observed for spleen cells from both mouse and rat (from 40% up to 80%). In preliminary studies we observed inhibition of mixed lymphocyte reaction on spleen cells from rats treated with Gang for 10 days (data not shown). Our data suggest that Gang may be investigated as a immunosuppressive drug in organ transplantation.

Efeitos do pneumoperitônio no rato

Effects of pneumoperitoneum in rats. The effects of different pneumoperitoneal pressures on kidney and liver functions and on arterial gasometry in rats are provided. A sample of twenty male rats weighing from 350 to 400 grams and distributed into two groups of ten animals was used. Each animal served as experimental control from which blood samples were collected before introducing CO2 into the peritoneal cavity. Samples underwent pressures of 4 and 6 mm Hg during one hour. Results show that increase in intra-abdominal pressure by CO2 in rats causes changes in their arterial gasometry, liver and kidney. However, no liver or kidney lesions during the above mentioned time were reported.

Estudo da transmigração bacteriana na apendicite experimental em coelhos

The present study aims to evaluate the occurrence of the bacterial transmigration in induced ezxperimental appendicitis in rabbitis through complete oclusion of the vermiform appendix. 24 New Zealand white rabbits with a body weight ranging from 2500 to 3000 grams were used. They were divided into 2 groups. The animals from group I were submitted to a median laparotomy and fragments from the spleen, left kidney, right lung, abdominal limphonode, light content from the vermiform appendix and blood from the portal vein were withdrawn for culture. The animals from group 2 were submitted to median laparotomy and complete occlusion of the vermiform appendix in order to induce appendicitis and, after 24 hours, material was collected following the steps of group 1. It was not observed an increase in bacteria in the samples analysed in the Control Group Group 1). The increase of Escherichia coli was observed in all the animals from the Experiment Group (Group 2). The bacterial transmigration occured after 24-hour observation in rabbits with experimental appendicitis.

Anastomose arterial com fio de polidioxanona e fio de polipropileno. Estudo comparativo em cães

Arterial anastomose with polydioxanone and polypropilene suture. Comparative study in dogs. The objetive of this study was to compare the effects of the absorbable polidioxanone suture to the nonabsorbable polypropylene suture, in end-to-end anastomoses, using dogs femoral arteries. Twenty dogs were separated in two groups for observation on the 7 th and 30 th post-operatory day. Each dog had its femoral arteries seccioned and then aproximated one side with separate stitches of polidioxanone 6-0 suture, and the other side with polypropylene 6-0 suture. For the statistical studies Fisher, Mac nemar, Wilsoxon, Mann-Witney and T of Student tests (α ≤ 0,05) were carried out. No significant differences were observed between the sutures employed in clinical evolution. The results obtained in the arteriographic and macroscopic avaliation were similar, as well as the morphological histologic analysis. In the morfometric histologic analysis the polypropylene suture showed a significantly higher number of foreign body giant cells reaction, comparing to the polydioxanone suture on the 7 th or 30 th post-operatory day. The results allow us to conclude that, in spit of the higher reaction of foreign body observed with the polypropilene suture, the polydioxanone suture showed similar results in dogs arterial anastomoses.

Gastrostomia percutânea endoscópica em suínos realizada com instrumentos de uso rotineiro em hospital geral

ABSTRACTPURPOSE: To perform a endoscopic gastrostomy by the introducer method with routine instruments used ina general hospital, without special instruments or special kits. METHODS: This procedure was performed in pigs ( Sus scrofa domesticus ) under observation for sevendays and then submitted to euthanasia.The technique was evaluated for macroscopic and histologic parameters. RESULTS: All animals had a good evolution without major complications. Some minor complications occurred like a rupture of Foley catheter balloon and subcutaneous space abscess. CONCLUSION: The percutaneous gastrostomy with routine general hospital instruments is successfulperformed, is safe,cheap and must be performed by skilled endoscopists. KEY WORDS: Surgery. Endoscopy.
