O acido desoxiribunocleico ribossomal (rDNA) e utilizado como uma ferramenta importante para caracterizar o polimorfismo entre os fungos. Existem muitas copias de rDNA as quais sao arranjadas por espacos nao codificados. Essas copias sao altamente conservadas entre especies de fungos. O objetivo deste trabalho foi estudar a regiao do Espaco Interno Transcrito (ITS) e analisar as diferencas no polimorfismo da sequencia dessa regiao no fungo Scleroderma UFSMSc1 com sequencias dos isolados de Scleroderma e Pisolithus do banco de dados GenBank. O DNA do isolado de Scleroderma UFSMSc1 foi extraido por meio da solucao de extracao a base de CTAB. A partir do DNA, foram feitas reacoes de PCR com os oligonucleotideos iniciadores universais ITS1 e ITS4, cujo produto amplificado foi purificado e sequenciado. A regiao do ITS do fungo mostrou uma banda simples de aproximadamente 650 pares de base. Na analise da sequencia dessa regiao em comparacao com algumas depositadas no GenBank, observou-se a formacao de agrupamento com especies de Scleroderma. Os resultados mostraram que essa tecnica favorece a identificacao de especies de Scleroderma, visto que tais fungos sao dificeis de ser identificados apenas por seus caracteres morfologicos.

Family farmers in the southeastern region of Pará come over time discussing new production systems that are more appropriate to their reality for environmental conservation and sustainability. Thus, arbuscular mycorrhizal fungi can be used as a strategy to promote environmental conservation. The objective of this work was to evaluate the development and degree of mycorrhizal dependence of jatobá (Hymenaea courbaril L.) seedlings. The work was carried out in Vegetation House from nov / 2009 to Feb / 2010. The jatobá seeds were collected in the municipality of Marabá. Before planting these seeds were immersed in water for 20 minutes to remove the floury pulp and to avoid the appearance of fungi and bacteria that would harm the germination. The experimental design was completely randomized in a factorial scheme of 1 x 3 x 2 x 10, as a forest species (jatobá), three treatments of FMA inoculation (G. margarita inoculation with S. heterogama and without inoculation). Two substrates (Red Argissolo gully soil and gully soil mixed with bovine manure and vermiculite) and 10 replicates. Morphological growth parameters (height, neck diameter and number of leaves) were evaluated at 30 and 60 days after germination. While the degree of mycorrhizal dependence and sporulation were evaluated at 60 days after germination. The degree of mycorrhizal dependence of jatobá was classified as a facultative plant and the fungus with the highest sporulation rate was the species Gigaspora margarita.
