Statistical characterisation of heavy metal contents in Paracentrotus lividus from Mediterranean Sea

This work focuses on the estimation of Hg, As, Cr, Ni, Cu, V, Cd and Pb by inductively coupled plasma mass spectrometry in 135 adult specimens of Paracentrotus lividus collected in different coastal areas of Sicily (Gela, Punta Secca, Ragusa (RG), Siracusa, Priolo, Catania, Messina, Milazzo, Brolo and Filicudi), in order to monitor the Mediterranean marine ecosystem by use of sea urchin as bioindicator. Moreover, the paper deals with the statistical classification of the tested samples according to the sampling area based on metal concentrations. The descriptive statistics findings were obtained and, a starting multivariate matrix was built. Data-sets were subjected to Kruskal–Wallis test to verify the significance of differences in metal concentrations, and then a factor analysis with principal components extraction was performed to try to differentiate urchin samples collected in different areas. The results showed that P. lividus is a suitable organism to be used as bioindicator.

Toxic and essential metals determination in commercial seafood: Paracentrotus lividus by ICP-MS

Toxic and essential metals content was determined by inductively coupled plasma mass spectrometry in commercial sea urchins samples from the Mediterranean Sea and Pacific Ocean. Results show that Chilean samples have the highest values of Cd, As, Ni, Cr and V; Spanish samples have the maximum Hg content while Sicilian samples have the lowest content of toxic metals. The toxic metal traces were compared with the limits of European Community (EC) No. 1881/2006 for bivalve molluscs. All samples exceed Pb and Cd limit levels; regarding Hg levels, only Sicilian and Sardinian samples have Hg content below the EC limits. The dietary intake of toxic and essential elements was evaluated for an adult. Furthermore, our study highlights a correlation between the toxic elements content and the marine environmental conditions of the places of origin even though only the generic FAO fishing area is specified.

Extracts deriving from olive mill waste water and their effects on the liver of the goldfish Carassius auratus fed with hypercholesterolemic diet.

The present research aims to evaluate the beneficial effects of polyphenols derived from waste water from a olive mill, obtained by non-plastic molecular imprinting device, in a hypercholesterolemic diet on Carassius auratus, commonly known as goldfish that was selected as experimental model. The study was conducted with morphological and histochemical analyses and also the data were supported by immunohistochemical analysis. Results show the beneficial activity of polyphenols with a reduction of the damage in the steatotic group, confirming that they may be suggested in the treatment of diseases by lipid accumulation, and used as any addition in feed for farmed fish, in order to improve the organoleptic and nutritional quality. The beneficial effects of waste oil extract should be suggested in the contexts of research programmes focused on the products to the health system. Furthermore, the olive mill waste water polyphenols free can be used as natural fertilizers.

Determination of 1,2/1,3-diglycerides in Sicilian extra-virgin olive oils by (1)H-NMR over a one-year storage period.

Abstract This study is aimed to monitor by 1H NMR spectroscopy the effect of a 12-month storage period on the 1,2-diglycerides over 1,3-diglycerides ratio for five mono-cultivar ‘extra virgin olive oils’ (EVOO) (Arbequina, Arbosana, Cerasuola, Nocellara and FS17) and one blend of two different cultivars (Nocellara + Biancolilla) preserved in the dark and at room temperature. These quantifications, at 500 MHz, are readily extracted through a specific and original integration difference method. Albeit it was known that the isomerisation rate is affected by the free acidity, we here demonstrate that it also depends on the presence of specific macromolecules (lipases), indeed, different EVOO cultivars with similar free acidity, show different isomerisation rate. Our results are consistent with similar diglyceride monitoring performed on Greek and Spanish EVOOs by 31P NMR.

Metabolite and mineral profiling of “Violetto di Niscemi” and “Spinoso di Menfi” globe artichokes by 1H-NMR and ICP-MS

Abstract Globe artichoke has been long considered a nutraceutical food for its valuable content of bioactive compounds. However, beside a well-known polyphenol profile, poor information is available about its metabolite and mineral composition. The aim of this study was to investigate edible parts of Sicilian artichokes, ‘Spinoso di Menfi’ and ‘Violetto di Niscemi’, by 1H NMR and ICP-MS for elucidating these compositional aspects. Although bracts and hearts of both artichokes shared a very similar metabolite pattern, ‘Spinoso di Menfi’ showed a higher number of metabolites, such as amino acids and polyphenols, than ‘Violetto di Niscemi’. ‘Spinoso di Menfi’ was also marked by higher levels of macro- and microelements when compared to ‘Violetto di Niscemi’. Also, artichoke heart demonstrated to accumulate higher mineral levels than bracts. 1H NMR and ICP-MS successfully profiled metabolites and metals in such plant food, partially covering the lack of literature data about ‘Spinoso di Menfi’ and ‘Violetto di Niscemi’ artichokes.

Determination of trace elements in goat and ovine milk from Calabria (Italy) by ICP-AES.

There are many sources of contamination to which milk could be exposed: grazing animals can ingest contaminants present in nature, such as lead in soil, or that have been deposited on grass, resulting from industrial emissions. Another possible route of contamination is represented by feed for animals, which may contain heavy and essential metals. The potential of high-resolution inductively coupled plasma spectrometry was evaluated to quantify reliably various toxic and essential elements (Fe, Zn, Pb, Cr, Ni, Cu, As, Se, Cd) in 47 samples of goat and ovine milk from various farms in Calabria (southern Italy). The results showed that concentrations of cadmium were below the limit of detection. Lead levels were below the maximum limits as set by the EC in almost all samples tested. The highest values were those of Zn followed by Fe, Cu and Se.

Statistical Analysis of Mineral Concentration for the Geographic Identification of Garlic Samples from Sicily (Italy), Tunisia and Spain

We performed a statistical analysis of the concentration of mineral elements, by means of inductively coupled plasma mass spectrometry (ICP-MS), in different varieties of garlic from Spain, Tunisia, and Italy. Nubia Red Garlic (Sicily) is one of the most known Italian varieties that belongs to traditional Italian food products (P.A.T.) of the Ministry of Agriculture, Food, and Forestry. The obtained results suggest that the concentrations of the considered elements may serve as geographical indicators for the discrimination of the origin of the different samples. In particular, we found a relatively high content of Selenium in the garlic variety known as Nubia red garlic, and, indeed, it could be used as an anticarcinogenic agent.

Interdonato lemon from Nizza di Sicilia (Italy): chemical composition of hexane extract of lemon peel and histochemical investigation

Abstract Considering that the determination of authenticity and of the geographical origin of food is a very challenging issue, in this study we studied by means of histological and histochemical analyses the famous Sicilian lemon known as ‘Interdonato Lemon of Messina PGI’. Since the protected geographical indication Interdonato lemon of Messina possesses high organoleptic properties, the composition of the hexane extract of lemon peel was determined by HRGC and HRGC–MS analyses and compared with that of lemon of different cultivars. The results obtained are informative of the oil’s quality and explain the variation of the lemon essential oil composition. Given the fundamental economic implications of any fraud, the aim of this study was to determine a fingerprint able to evaluate the authentication of the geographic origin in such way to prevent frauds in national and international markets. Fast UPLC/PDA determination of squalene in Sicilian P.D.O. pistachio from Bronte: optimization of oil extraction method and analytical characterization

A fast reversed-phase UPLC method was developed for squalene determination in Sicilian pistachio samples that entry in the European register of the products with P.D.O. In the present study the SPE procedure was optimized for the squalene extraction prior to the UPLC/PDA analysis. The precision of the full analytical procedure was satisfactory and the mean recoveries were 92.8±0.3% and 96.6±0.1% for 25 and 50mgL-1 level of addition, respectively. Selected chromatographic conditions allowed a very fast squalene determination; in fact it was well separated in ∼0.54min with good resolution. Squalene was detected in all the pistachio samples analyzed and the levels ranged from 55.45-226.34mgkg-1. Comparing our results with those of other studies it emerges that squalene contents in P.D.O. Sicilian pistachio samples, generally, were higher than those measured for other samples of different geographic origins.

NMR analysis of aldehydes in Sicilian extra-virgin olive oils by DPFGSE techniques

The DPFGSE NMR sequences open new perspectives in the volatile compounds analysis of food matrices. Many fresh extra-virgin Sicilian olive oils, analyzed by this technique, show two main resonances in the aldehydic spectral region (9–10 ppm), at 9.18 and 9.58 ppm. The former was never reported so far, the latter was sometime highlighted as a minor aldehydic component signal of spectra showing stronger resonances at 9.45 and 9.70 ppm. Thermal treatment at 220°C of extra virgin olive oil samples lead to the complete transformation of the resonances at 9.18 and 9.58 ppm into those at 9.45 and 9.70 ppm in 50 minutes. Analogous transformation takes place place in CDCl 3 at rt in several weeks. These results suggest the transformation of relatively unstable compounds into thermodynamically more stable products whose resonances are commonly reported in the literature. Even though these chemical changes involve minimal amount of product, they are of crucial importance to define: i) organoleptic extra virgin olive oil properties; ii) fraudulent chemical or thermal treatment detection; iii) extra virgin oil ageing.
