The purpose of this study was the comparison of the diagnostic sensitivity between buffy coat (BC), peripheral blood (PB), lymph node (LN), bone marrow (BM) and short-term culture (P-D) cytology that has been based on the detection of Ehrlichia canis morulae, in the acute phase of canine monocytic ehrlichiosis (CME). Their cellular localization, total numbers and microscopic differentials were also investigated. The highest sensitivities were achieved after evaluating 1000 oil immersion fields (OIFs) in BC (66%) and an equal number in LN (60.9%) smears, separately or together (74%). The morulae were more often detected into lymphocytes than monocytes. The highest total number of morulae (n=143) were found in P-D smears. Finally, to avoid false positive diagnoses, platelets, lymphocytic azurophilic granules, lymphoglandular bodies and phagocytosed nuclear material should not be confused with the morulae.

Twenty domestic shorthaired (DSH) and 20 Siamese (S) kittens were allocated into 4 breed-specific groups, of 10 kittens each, that were fed exclusively cooked sardines (F groups) or commercial feline canned food based on oily fish (C groups) for a 4-month period. Clinical signs were scored every 15 d along with body weight recording and blood sampling for the measurement of alpha-tocopherol and selenium (Se) concentrations and glutathione peroxidase (GSH-Px) activity. Subcutaneous adipose tissue samples were obtained per month to determine its fatty acid composition. Steatitis, reproduced in all 20 F-group kittens, was accompanied by systemic signs in 5 DSH and 6 S animals. The severity of the disease reached its zenith at the second week in the DSH-F-group kittens and the fourth and sixth week in the S-F-group kittens. alpha-Tocopherol plasma level was significantly lower in F groups compared to their corresponding controls, whereas the opposite was true for Se and red blood cell GSH-Px activity. In conclusion, the results of this study have shown that although the morbidity rate is not different between the two breeds, the delay of Siamese cats to develop symptomatic steatitis is presumably attributed to an inherent resistance as a result of the long-standing evolution of more efficient antioxidant mechanisms. BACKGROUND Excess reactive oxygen species due to oxidative stress and the ensuing lipid peroxidation are believed to be involved in mammary gland tumor (MGT) pathogenesis. OBJECTIVE The purpose of this study was to evaluate the extent of lipid peroxidation as evidenced by thiobarbituric acid-reactive substances (TBARS) formation, and the concentration of α-tocopherol as an inhibitor of lipid peroxidation, in blood and neoplastic tissue of dogs with malignant MGT. The correlation between inflammatory cell infiltration score and TBARS or α-tocopherol in MGT was also evaluated. METHODS Sixteen intact female dogs with malignant MGT and 12 clinically healthy and age/weight-matched controls were included in the study. In all dogs, serum TBARS, α-tocopherol, total cholesterol, and triglycerides were measured. Tissue TBARS and α-tocopherol levels were determined in 1 cm(3) sized tissue samples collected from MGT and adjacent, ipsilateral, normal mammary gland tissue from the 16 affected dogs. The degree of inflammatory cell tumor infiltration was evaluated histologically. RESULTS There were no statistically significant differences in serum levels of TBARS, α-tocopherol, total cholesterol, and triglycerides between dogs with and without malignant MGT. TBARS were significantly higher, whereas α-tocopherol was lower in neoplastic tissue when compared with normal mammary gland tissue. There was no correlation between TBARS or α-tocopherol concentration and the inflammatory cell infiltration score in neoplastic tissue. CONCLUSION The increased level of TBARS suggests oxidative stress induction in canine malignant MGT. The origin of this phenomenon is not clear, as a potential oxidative burst could not be attributed to inflammatory cells infiltrating the tumors.
