Polyploidy, B chromosomes, and heterochromatin characterization of Mimosa caesalpiniifolia Benth. (Fabaceae-Mimosoideae)

Saulo Marçal de Sousa; Aryane Campos Reis; Lyderson Facio Viccini

Eight populations of Mimosa caesalpiniifolia Benth. were investigated using a cytogenetic approach. Here, we describe for the first time details of the karyotype including chromosome morphology, physical mapping of chromomycin A3 (CMA) 4′,6-diamidino-2-phenylindole (DAPI) and silver staining of nucleolar organizer regions (Ag-NOR banding), as well as 45S rDNA sites. All populations studied showed karyotypes with 2n = 2x = 26 small metacentric and submetacentric chromosomes, although some individuals exhibited 2n = 4x = 52 chromosomes. Moreover, we observed putative additional B chromosomes in some populations. The CMA banding and fluorescent in situ hybridization techniques revealed NOR heteromorphism on the unique pair containing 45 rDNA site (chromosome 12) while the Ag-NOR banding indicated NORs on both cytotypes. Up to two and four nucleoli were observed, respectively, on individuals with 2n = 2x = 26 and 2n = 4x = 52 chromosomes and the differences in nucleolar size seems to be directly related to NOR heteromorphism in some individuals. The data present new and important information to understand karyotypic evolution of Mimosa and Fabaceae.

The genus Heteropterys is one of the major genera in Malpighiaceae. However, few cytological and palynological studies were reported. The present work described for the first time the chromosome number, heterochromatin pattern, meiotic behavior, pollen viability and palynological aspects of Heteropterys ubellata, a very spread species. One large Brazilian population was evaluated using conventional techniques for meiotic studies and acetolyse to access the pollen morphology. The species showed 2n = 20 chromosomes, normal meiotic development and viable pollens. Great blocks of heterochromatin were observed around the centromers. DAPI staining was positive for centroeric heterochroatin, hile CMA3 ark as observed just at terinal regions of one pair of hoologues chromosomes. This result and the presence of one chromosome pair attached to the nucleoli during the pachytene and diakinesis suggested the presence of only one pair of NORs. Palynological analysis revealed that pollen grains are apolar, 6 porate and with colpoids associated to all pores. The pollen content was positive for the starch test, and the exine was rugulate with little psilate regions.

Contributions to cytogenetics of Plectranthusbarbatus Andr. (Lamiaceae): a medicinal plant.

Aryane Campos Reis; Lyderson Facio Viccini; Saulo Marçal de Sousa

Abstract Accessions of Plectranthus barbatus (Lamiaceae), a medicinal plant, were investigated using a cytogenetic approach and flow cytometry (FCM). Here, we describe for the first time details of the karyotype including chromosome morphology, physical mapping of GC rich bands (CMA3 banding), as well as the mapping of 45S and 5S rDNA sites. All accessions studied showed karyotypes with 2n = 30 small metacentric and submetacentric chromosomes. The CMA3 banding and fluorescent in situ hybridization techniques revealed coincidence between CMA3 bands and 45S rDNA sites (6 terminal marks) while for the 5S rDNA were observed 4 subterminal marks no coincident with CMA3 marks. For nuclear genome size measurement, the FCM procedure provided histograms with G0/G1 peaks exhibiting CV between 2.0–4.9 and the mean values obtained for the species was 2C = 2.78 pg, with AT% = 61.08 and GC% = 38.92. The cytogenetic data obtained here present new and important information which enables the characterization of Plectranthus barbatus.

An increment in the Duranta repens L. (Verbenaceae) knowledge: DNA content, karyology, meiosis and palynology

Saulo M. Sousa; Aryane Campos Reis; Pâmela S. Silva; Lyderson Facio Viccini

Abstract Meiotic behavior, pollen morphology, interphasic nucleus pattern and karyotype description for Duranta repens (Verbenaceae) are presented. The species had semi-reticulate interphasic nucleus and in mitotic metaphase, chromosome number was 2n = 34 (32 m + 2 sm), with secondary constriction in one chromosome pair. Additionally to the A complement, we observed B chromosomes, varying to 0–3 in some cells of several individuals. Meiotic behavior was regular and pollen grains were isopolar and tricolporate, being classified as prolate-spheroidal with psilate-perfurate exine ornamentation.
