Medial prefrontal cortex supports source memory accuracy for self-referenced items

Previous behavioral work suggests that processing information in relation to the self enhances subsequent item recognition. Neuroimaging evidence further suggests that regions along the cortical midline, particularly those of the medial prefrontal cortex (PFC), underlie this benefit. There has been little work to date, however, on the effects of self-referential encoding on source memory accuracy or whether the medial PFC might contribute to source memory for self-referenced materials. In the current study, we used fMRI to measure neural activity while participants studied and subsequently retrieved pictures of common objects superimposed on one of two background scenes (sources) under either self-reference or self-external encoding instructions. Both item recognition and source recognition were better for objects encoded self-referentially than self-externally. Neural activity predictive of source accuracy was observed in the medial PFC (Brodmann area 10) at the time of study for self-referentially but not self-externally encoded objects. The results of this experiment suggest that processing information in relation to the self leads to a mnemonic benefit for source level features, and that activity in the medial PFC contributes to this source memory benefit. This evidence expands the purported role that the medial PFC plays in self-referencing.

Medial prefrontal cortex supports source memory for self-referenced materials in young and older adults

Behavioral evidence suggests that young and older adults show a benefit in source memory accuracy when processing materials in reference to the self. In the young, activity within the medial prefrontal cortex supports this source memory benefit at study. In this investigation, we examined whether the same neural regions support this memory benefit in both age groups. Using fMRI, we scanned participants while they studied and retrieved pictures of objects paired with one of three scenes (source) under self-reference and other-reference conditions. At the time of study, half of the items were presented once and half twice, allowing us to match behavioral performance between the groups. Both groups showed equivalent source accuracy benefits for objects encoded self-referentially. Activity in the left dorsal medial prefrontal cortex supported subsequent source memory in both age groups for the self-referenced relative to the other-referenced items. At the time of test, source accuracy for both the self- and other-referenced items was supported by a network of regions including the precuneus in both age groups. At both study and test, little in the way of age differences emerged, suggesting that when they are matched on behavioral performance, young and older adults engage similar regions in support of source memory when processing materials in reference to the self; however, when we did not match performance, age differences in functional recruitment were prevalent. These results suggest that by capitalizing on preserved processes (self-referential encoding), older adults can show improvement in memory for source details that they would typically not remember well, relative to the young.

Self-referencing enhances recollection in both young and older adults

Processing information in relation to the self enhances subsequent item recognition in both young and older adults and further enhances recollection at least in the young. Because older adults experience recollection memory deficits, it is unknown whether self-referencing improves recollection in older adults. We examined recollection benefits from self-referential encoding in older and younger adults and further examined the quality and quantity of episodic details facilitated by self-referencing. We further investigated the influence of valence on recollection, given prior findings of age group differences in emotional memory (i.e., “positivity effects”). Across the two experiments, young and older adults processed positive and negative adjectives either for self-relevance or for semantic meaning. We found that self-referencing, relative to semantic encoding, increased recollection memory in both age groups. In Experiment 1, both groups remembered proportionally more negative than positive items when adjectives were processed semantically; however, when adjectives were processed self-referentially, both groups exhibited evidence of better recollection for the positive items, inconsistent with a positivity effect in aging. In Experiment 2, both groups reported more episodic details associated with recollected items, as measured by a memory characteristic questionnaire, for the self-reference relative to the semantic condition. Overall, these data suggest that self-referencing leads to detail-rich memory representations reflected in higher rates of recollection across age.

Reducing perceptual interference improves visual discrimination in mild cognitive impairment: Implications for a model of perirhinal cortex function

Memory loss resulting from damage to the medial temporal lobes (MTL) is traditionally considered to reflect damage to a dedicated, exclusive memory system. Recent work, however, has suggested that damage to one MTL structure, the perirhinal cortex (PRC), compromises complex object representations that are necessary for both memory and perception. These representations are thought to be critical in shielding against the interference caused by a stream of visually similar input. In this study, we administered a complex object discrimination task to two memory‐impaired populations thought to have brain damage that includes the PRC [patients diagnosed with amnestic mild cognitive impairment (MCI), and older adults at risk for MCI], as well as age‐matched controls. Importantly, we carefully manipulated the level of interference: in the High Interference condition, participants completed a block of consecutive perceptually similar complex object discriminations, whereas in the Low Interference condition, we interspersed perceptually dissimilar objects such that there was less buildup of visual interference. We found that both memory‐impaired populations were impaired on the High Interference condition compared with controls, but critically, by reducing the degree of perceptual interference, we were largely able to improve their performance. These findings, when taken together with convergent evidence from animals with selective PRC lesions and amnesic patients with focal damage to the PRC, provide support for a representational‐hierarchical model of PRC function and suggest that memory loss following PRC damage may reflect a heightened vulnerability to perceptual interference.

A single bout of resistance exercise can enhance episodic memory performance

Acute aerobic exercise can be beneficial to episodic memory. This benefit may occur because exercise produces a similar physiological response as physical stressors. When administered during consolidation, acute stress, both physical and psychological, consistently enhances episodic memory, particularly memory for emotional materials. Here we investigated whether a single bout of resistance exercise performed during consolidation can produce episodic memory benefits 48 h later. We used a one-leg knee extension/flexion task for the resistance exercise. To assess the physiological response to the exercise, we measured salivary alpha amylase (a biomarker of central norepinephrine), heart rate, and blood pressure. To test emotional episodic memory, we used a remember-know recognition memory paradigm with equal numbers of positive, negative, and neutral IAPS images as stimuli. The group that performed the exercise, the active group, had higher overall recognition accuracy than the group that did not exercise, the passive group. We found a robust effect of valence across groups, with better performance on emotional items as compared to neutral items and no difference between positive and negative items. This effect changed based on the physiological response to the exercise. Within the active group, participants with a high physiological response to the exercise were impaired for neutral items as compared to participants with a low physiological response to the exercise. Our results demonstrate that a single bout of resistance exercise performed during consolidation can enhance episodic memory and that the effect of valence on memory depends on the physiological response to the exercise.

Aging affects the interaction between attentional control and source memory: An fmri study

Age-related source memory impairments may be due, at least in part, to deficits in executive processes mediated by the PFC at both study and test. Behavioral work suggests that providing environmental support at encoding, such as directing attention toward item–source associations, may improve source memory and reduce age-related deficits in the recruitment of these executive processes. The present fMRI study investigated the effects of directed attention and aging on source memory encoding and retrieval. At study, participants were shown pictures of objects. They were either asked to attend to the objects and their color (source) or to their size. At test, participants determined if objects were seen before, and if so, whether they were the same color as previously. Behavioral results showed that direction of attention improved source memory for both groups; however, age-related deficits persisted. fMRI results revealed that, across groups, direction of attention facilitated medial temporal lobe-mediated contextual binding processes during study and attenuated right PFC postretrieval monitoring effects at test. However, persistent age-related source memory deficits may be related to increased recruitment of medial anterior PFC during encoding, indicative of self-referential processing, as well as underrecruitment of lateral anterior PFC-mediated relational processes. Taken together, this study suggests that, even when supported, older adults may fail to selectively encode goal-relevant contextual details supporting source memory performance.

Task-Selective Memory Effects for Successfully Implemented Encoding Strategies

Previous behavioral evidence suggests that instructed strategy use benefits associative memory formation in paired associate tasks. Two such effective encoding strategies–visual imagery and sentence generation–facilitate memory through the production of different types of mediators (e.g., mental images and sentences). Neuroimaging evidence suggests that regions of the brain support memory reflecting the mental operations engaged at the time of study. That work, however, has not taken into account self-reported encoding task success (i.e., whether participants successfully generated a mediator). It is unknown, therefore, whether task-selective memory effects specific to each strategy might be found when encoding strategies are successfully implemented. In this experiment, participants studied pairs of abstract nouns under either visual imagery or sentence generation encoding instructions. At the time of study, participants reported their success at generating a mediator. Outside of the scanner, participants further reported the quality of the generated mediator (e.g., images, sentences) for each word pair. We observed task-selective memory effects for visual imagery in the left middle occipital gyrus, the left precuneus, and the lingual gyrus. No such task-selective effects were observed for sentence generation. Intriguingly, activity at the time of study in the left precuneus was modulated by the self-reported quality (vividness) of the generated mental images with greater activity for trials given higher ratings of quality. These data suggest that regions of the brain support memory in accord with the encoding operations engaged at the time of study.

The influence of directed attention at encoding on source memory retrieval in the young and old: an ERP study.

Neuroimaging evidence suggests that older adults exhibit deficits in frontally-mediated strategic retrieval processes, such as post-retrieval monitoring. Behavioral research suggests that explicitly directing attention toward source features during encoding may improve source memory for both young and older adults and alleviate age-related source memory impairments, in part, by reducing demands on post-retrieval monitoring. We investigated this hypothesis in the present event-related potential (ERP) study. Young and older adults attended to either objects and their presented color (source) or to the object alone during study and made color source memory decisions at test. We attempted to match performance between groups by halving the memory load for older adults. Behavioral results showed that, while direction of attention to object and color improved source memory for both groups, older adults benefited less than the young. ERPs revealed that demands on late right frontal effects, indicative of post-retrieval monitoring, were similarly reduced by directed attention at encoding for both groups. However, older adults showed reduced ERP correlates of recollection (parietal old-new effect), as well as a sustained widespread negativity, potentially indicative of memory searches for perceptual details in the face of impaired recollection. These results suggest that older adults, like the young, can engage in post-retrieval monitoring when source details are difficult to recover. However, impaired recollection may underlie persistent age-related source memory deficits, even when encoding is supported via directed attention.

Retrospective cues based on object features improve visual working memory performance in older adults.

Abstract Research with younger adults has shown that retrospective cues can be used to orient top-down attention toward relevant items in working memory. We examined whether older adults could take advantage of these cues to improve memory performance. Younger and older adults were presented with visual arrays of five colored shapes; during maintenance, participants were presented either with an informative cue based on an object feature (here, object shape or color) that would be probed, or with an uninformative, neutral cue. Although older adults were less accurate overall, both age groups benefited from the presentation of an informative, feature-based cue relative to a neutral cue. Surprisingly, we also observed differences in the effectiveness of shape versus color cues and their effects upon post-cue memory load. These results suggest that older adults can use top-down attention to remove irrelevant items from visual working memory, provided that task-relevant features function as cues.

Age-related deficits in selective attention during encoding increase demands on episodic reconstruction during context retrieval: An ERP study

Previous event-related potential (ERP) and neuroimaging evidence suggests that directing attention toward single item-context associations compared to intra-item features at encoding improves context memory performance and reduces demands on strategic retrieval operations in young and older adults. In everyday situations, however, there are multiple event features competing for our attention. It is not currently known how selectively attending to one contextual feature while attempting to ignore another influences context memory performance and the processes that support successful retrieval in the young and old. We investigated this issue in the current ERP study. Young and older participants studied pictures of objects in the presence of two contextual features: a color and a scene, and their attention was directed to the objects relationship with one of those contexts. Participants made context memory decisions for both attended and unattended contexts and rated their confidence in those decisions. Behavioral results showed that while both groups were generally successful in applying selective attention during context encoding, older adults were less confident in their context memory decisions for attended features and showed greater dependence in context memory accuracy for attended and unattended contextual features (i.e., hyper-binding). ERP results were largely consistent between age groups but older adults showed a more pronounced late posterior negativity (LPN) implicated in episodic reconstruction processes. We conclude that age-related suppression deficits during encoding result in reduced selectivity in context memory, thereby increasing subsequent demands on episodic reconstruction processes when sought after details are not readily retrieved.
