The palladium-catalyzed direct arylation of imidazolone N-oxides with aryl bromides to afford the corresponding 4-aryl imidazolone N-oxides is described. This method has been successfully used for the synthesis of GSK2137305.

Design and synthesis of aryl ether and sulfone hydroxamic acids as potent histone deacetylase (HDAC) inhibitors.

A series of novel hydroxamic acid based histone deacetylases (HDAC) inhibitors with aryl ether and aryl sulfone residues at the terminus of a substituted, unsaturated 5-carbon spacer moiety have been synthesized for the first time and evaluated. Compounds with meta- and para-substitution on the aryl ring of ether hydroxamic acids 19c, 20c, 19e, 19f and 19g are potent HDAC inhibitors with activities at low nanomolar levels.

Synthesis of Heterocycles by Palladium-Catalyzed Intramolecular Heteroarylation

Abstract Synthesis of heterocycles by intramolecular C N bond formation through a palladium-catalyzed Hartwig–Buchwald reaction is described. The review covers the results reported for the synthesis of five- and six-membered rings, as well as medium-size and macro-heterocycles over the past 15 years. The review is organized by the size of the rings formed with further partition into subsections based on number of heteroatoms on the ring or fused ring systems. A separate section deals with tandem C C/C N palladium-catalyzed annulation sequences and cascades.

Improved Synthesis of Dihydrothebainone and Its 14 β-Epimer

Abstract An improved synthesis for the preparation of diastereomerically pure dihydrothebainone (1) from thebaine (3) is reported. A 41% overall yield was realized over three steps via direct transformation of dihydrothebaine-O (4) to thebainone-A (6) with 6N HCl.
