Abstract The stereoisomerism of some spiro-1,3-dioxanes obtained from chiral 1,3-diols or chiral substituted cyclohexanones is discussed on the basis of the helical chirality of the 1,5-dioxaspiro[5.5]undecane skeleton and of the data of conformational analysis. The influence of the flexibility of the rings on the representative number of isomers and on their NMR spectra is commented. The compositions of the stereoisomers (diastereoisomers and enantiomers) of some representative compounds have been determined by gas chromatography using chiral columns.

Synthesis and stereochemistry of some new brominated spiro 1,3-dioxanes

New dibrominated spiro 1,3-dioxanes have been obtained by a regio- and diastereo-selective reaction between six-membered ring spiro compounds and bromine. Studies on the stereochemistry of these brominated compounds, exhibiting a 1,5-dioxaspiro[5.5]undecane skeleton, were carried out by NMR methods in solution and by the solid-state molecular structure of four compounds established by single-crystal X-ray diffractometry. The diasteroselectivity of the bromination was explained by the asymmetric induction of chiral carbon atoms and the chiral spiro skeleton.

A main target in the treatment of hypertension is the angiotensin-converting enzyme (ACE). This enzyme is responsible for producing angiotensin II, a potent vasoconstrictor. Therefore, one of the targets in the treatment of hypertension is to inhibit ACE activity. Hence, this study’s aim is to use computational studies to demonstrate that the proposed heterocyclic compounds have a molecular affinity for ACE and that, furthermore, these heterocyclic compounds are capable of inhibiting ACE activity, thus avoiding the production of the vasopressor Angiotensin II. All this using computer-aided drug design, and studying the systems, with the proposed compounds, through molecular recognition process and compared with the compounds already on the market for hypertension.
