Changes in Glycoproteins and Glycolipids of the Ghosh-Lai Rat Stomach Following Perfusions with Ethanol

Publisher Summary The chapter describes the acute effect of ethanol, at a concentration of 10% or more, on gastric mucosa and secretion of HCL. The quantitative aspects of serum protein leakage into the stomach following ethanol instillation and its relation to the movements of gastric glycoproteins through gastric mucosa have not been studied through different methods. In a study described in the chapter, isolated Ghosh-Lai stomachs were prepared in rats under urethane anesthesia. Perfusion of isolated Ghosh-Lai rat stomachs with 10%, 15%, or 20% ethanol caused an increase in the concentration of total proteins, immunoreactive serum albumin, and protein- and lipid-bound carbohydrates inthe perfusates, as compared to saline controls. The presence of immunoreactive serum albumin and mannose, in a relatively large proportion, indicated considerable leakage of serum proteins into the gastric content following ethanol perfusion. As no significant admixture of hemoglobin was found in ethanol perfusates (at most, 1 part of blood per 30 000 vol.), it was concluded that no breaking of vascular membrane occurred in Ghosh-rat stomach following ethanol perfusion in concentrations used. The high amount of protein-bound fucose indicated the elution of the preformed surface epithelial mucus by ethanol perfusions, perhaps with stimulation of its secretion.
