Determining physiological potential in black oats seeds shows some difficulties due to the lack of standardized tests. On the other hand, the measuring the electric conductibility of the seed immersion solution can be a fast method, that can be applied to oats seeds, because it indicates the integrity of cellular membranes, and gives us, indirectly, the information about the physiological quality of the seeds. The objective of this study was to evaluate the effect of the number of seeds and the imbibition time over the electrical conductivy of the solution, in the evaluation of the strength of oats seeds with and without shell. The experiment was conducted using three lots of common oats seeds. The test was made at 25o C, imbibition testing times of 4, 8 and 16 hours, with 25 and 50 seeds. The treatments constituted a factorial, using a complete randomized design with four repetitions. The use of 25 black oat seeds without shell in 75mL distilled water, with an imbibition time of eight hours, was enough to stratify the lots in different strength levels.

This research was aimed at determining the exposition period at low temperature in the modified cold germination test, to evaluate physiological potential of squash seed lots. Six seed lots of squash, variety Menina Brasileira (Cucurbita moschata Duch.), produced in 2003/2004, were submitted at different exposition times for the development of the cold test. The modified cold gernation test was carried out at 10°C of constant temperature, during exposition time of three, five, seven and nine days. A completely randomized desing was used to the cold test, 0 constituting a factorial 6x4, corresponding to the six seed lots and four exposition time (3, 5, 7, and 9 days), with four repetitions. The averages were compared by the Tukey test, at 5% of probability. The correlation test of Person (r) was used too. For the modified cold germination test, the period of three days was the most indicated to evaluate the physiological potential of squash seeds.

Relative expression of CYP and Arabinogalactan genes in soybean seed coats

Carlos André Bahry; Eduardo Venske; Anelise Tessari Perboni; Paulo Dejalma Zimmer

The aim of this study was to evaluate the relative expression of two candidate genes, CYP (cyclophilin) and Arabinogalactan, possibly involved with seed quality, in contrasting soybean seed coats. The following genotypes were used: BMX Potencia RR and CD 202, both with yellow coats, and TP and IAC, with black coats. The relative expression of the genes was evaluated by qPCR in seven development phases of the coats of the four genotypes at 25; 30; 35; 40; 45; 50 and 55 days after anthesis. The design was completely randomized, with three replicates. The data were submitted to analysis of variance and the means were compared via the Scott Knott test, at 5% probability. The CYP gene showed greater expression in the intermediate stages of development of the coats of the genotypes CD 202 and IAC, yellow coat and black coat, respectively. The A rabinogalactan gene presented greater expression in the final stages of coats development of BMX Potencia RR.

The attributes that confer seed quality can largely be considered dependent on the coat. The aim of this study was to evaluate the quality and hydration pattern of the seeds of four contrasting soybean genotypes for the coats characteristics; BMX Potencia RR and CD 202 with yellow coats and IAC and TP with black coats. The first stage of the study consisted of seed multiplication in the 2012/2013 harvest. After storage for five months at a temperature of 17 oC and a relative humidity of 50%, the seeds were subjected to the following tests: germination, first count and seedling height, with and without immersion of the seeds, emergence in soil, emergence speed and germination index; electrical conductivity; thousand weight seeds and imbibitions of seeds. The design was completely randomized, with five replicates. The data was subjected to variance analysis and the means compared using the Tukey test at a 5% level of probability. Regression analysis was used for the seeds imbibitions test. Genotypes with black coats (especially IAC) presented better physiological quality in relation to genotypes with yellow coats. The latter had greater thousand weight seeds. BMX Potencia RR presented greater hydration than the other genotypes in the first hours of imbibitions and TP, the least. Genotypes with black coats emitted a radicle faster than the yellow coats genotypes.

Phosphorus can contribute to increase seed quality and grain yield. The aim of this study was to evaluate the effect of phosphorus fertilization on quality, chemical composition and yield of soybean seeds and yield of crops established from these seeds. Assay 1 evaluated the following treatments: T1 - control, sowing without P2O5; T2: 50% less than the recommended P2O5 rate based on the soil analysis; T3: 100% of the recommended P2O5 rate; T4: 50% more than the recommended P2O5 rate; and T5: 100% more than the recommended P2O5 rate. After harvest, the number of seeds and pods per plant, yield, germination, viability and vigor by tetrazolium test, chemical composition and thousand-seed weight were determined. Assay 2 was conducted with the seeds harvested in Assay 1 to evaluate the number of pods per plant, number of seeds per plant and yield. The experiment was arranged in randomized blocks with five replicates. The phosphorus fertilization provided greater vigor in soybean seeds, besides contributing to increased phosphorus, iron and zinc in seeds produced. Seeds with higher phosphorus concentration contribute to the increase in soybean yield.
