Effect of Electrolyzed Water Combined with Ultrasound and Organic Acids to Reduce Salmonella Typhimurium, Staphylococcus aureus, Bacillus cereus on Perilla Leaves

This study was performed to compared the effectiveness of individual treatments (electrolyzed water: EW, organic acid, and ultrasound) and their combination on reducing foodborne pathogens from perilla leaves. Perilla leaves were innoculated with a cocktail of Salmonella Typhimurium, Staphylococcus aureus, Bacillus cereus. Inoculated perilla leaves were treated with EW combined with different concentration of acetic acid (0.5%, 1.0%, 1.5%, 2.0%) for 1 min at room temperature. Treatment of 3 pathogens on perilla leaves with electrolyzed water com- bined with ultrasound (25 kHz) and 0.5% acetic acid was also performed for 1 min. While the numbers of S. Typhimu- rium and B. cereus showed reduced with increasing acetic acid concentration, there is no difference in the number of S. aureus treated with EW containing 0.5% to 1.5% acetic acid. Discoloration was observed the perilla leaves treated with EW combined with more than 1.0% acetic acid. For all three pathogens, the combined treatment of EW and ultra- sound resulted in additional 0.42 to 0.72 log 10 CFU/g. The maxium reductions of S. Typhimurium and B. cereus were 0.95, 1.23 log 10 CFU/g after treatment with EW combined with 0.5% acetic acid and ultrasound simultaneously. The results suggest that the treatment of EW combined with 0.5% acetic acid and ultrasound increased pathogens reduc- tion compared to individual treatment.

Analysis of Yield of Eleutherosides B and E in Acanthopanax divaricatus and A. koreanum Grown with Varying Cultivation Methods

An analysis of the yield of eleutherosides B and E in Acanthopanax divaricatus and A. koreanum was performed using high performance liquid chromatography to evaluate production by different cultivation methods. In A. divaricatus and A. koreanum, the total content of eleutherosides B and E was 2.466–7.360 mg/g varying by plant section, 3.886–11.506 mg/g by pinching site, 3.655–10.083 mg/g by planting time, and 3.652–10.108 mg/g by fertilizer ratio. Thus the total content of eleutherosides B and E in A. divaricatus and A. koreanum differed depending on cultivation methods. These results present useful information for high eleutheroside content applications in A. divaricatus and A. koreanum. This information can affect selection of plant section and cultivation methods for nutraceutical, pharmaceutical, and cosmeceutical material development.

Perilla transformation using selection markers containing antibiotics and basta

선발마커로 두 종류의 항생제 (hpt와 nptII)와 제초제(bar) 유전자를 사용하여 아그로박테리움을 이용한 수정된 들깨 형질전환방법을 개발하였다. 들깨 배축 절편을 pMOG6-Bar 운반체 혹은 pCK-Bar 운반체를 가진 아그로박테리움 EHA 105와 각각 3일간 공동배양 하였다. 1차로 형성된 신초는 하이그로마이신(15mg/L)이나 카나마이신(125 mg/L)을 사용하여 선발하였고 재분화 된 신초들은 확실한 형질전환체를 얻기 위해서 포스피노트리신(1.2 mg/L)에서 한번 더 선발하였다. 뿌리는 호르몬이 없는 MS배지에서 재분화 신초로 부터 유도하였으며 80개의 재분화개체를 획득하였다. 들깨 지놈으로 형질전환유전자의 삽입은 서던 블럿으로 확인하였고 유전자의 발현은 노던 블럿으로 분석하였다. To 식물체로부터 유도된 T1들깨 종자들은 후대로의 안정된 유전자 전이를 확인하기 위하여 0.3% 바스타 제초제 살포로 확인하였다. 【A modified method of Agrobacterium-mediated perilla transformation was developed using two selection markers of an antibiotics (either hpt or nptll) and an herbicidal (bar) gene. Perilla hypocotyl explants were cocultured with Agrobacterium tumefaciens EHA 105 strain harboring plasmid vector (either pMOG6-Bar or pCK-Bar) for three days, respectively. Primary shoots were selected with antibiotics of hygromycin (15 mg/L) or kanamycin (125 mg/L) and regenerated shoots were further selected with herbicide phosphinothricin (ppt,1.2 mg/L) to obtain authentic transformants. Roots were induced for the regenerated shoots on the MS medium without hormone and 80 putative transgenic plants were obtained. Transgene integration into perilla genome was confirmed by Southern blot and their expression was analyzed by Northern blot. T1 perilla seeds drived from To plants were tested 0.3% basta spray for identification of stable gene delivery to next generation.】

Identification of Phenolic Compounds and Antioxidant Effects from the Exudate of Germinating Peanut(Arachis hypogaea)

A new soybean variety, ‘Joongmo 3009’ (Milyang 222) was developed at the National Institute of Crop Science (NICS) in 2012. ‘Joongmo 3009’ was released by pedigree selection from the cross between ‘Cheongja 2(Milyang 121)’ and ‘Daemangkong’. It has determinate growth habit, white flower, brown pubescence, brown pod color, green seed coat, green cotyledon, spherical seed shape, oval leaf shape and large seed size (29.3 grams per 100 seeds). It was late 16 days in maturing date than the check cultivar ‘Cheongjakong’. The average yield of ‘Joongmo 3009’ was 2.91 ton per hectare, which was higher 36 percentage than the check variety, in the regional yield trials carried out in three adaptable locations of Korea from 2010 to 2012. The number of breeder’s right is ‘5474’

Optimization of Extraction Conditions and Comparison of Rosmarinic and Caffeic Acids from Leaves of Perilla frutescens Varieties

Hairy vetch can fix nitrogen from the atmosphere as a leguminous cover crop. This research was carried out to determine optimum seeding method of hairy vetch and application effect in paddy. Field experiment was conducted at Sinheung series (fine loamy, mixed, nonacid, mesic family of Fluvaquentic Endoaquepts) from Sep. 2011 to Oct. 2012 at the National Institute of Crop Science (NICS), RDA, Suwon, Gyeonggi province, Korea. Hairy vetch used in the study was ‘Cheongpyungbora’, developed by National Institute of Crop Science. Seeding methods of hairy vetch consisted of Broadcasting Before Rice Harvesting (BBRH), Partial Tillage Seeding (PTS), Minimum Tillage Seeding (MTS), No Tillage Seeding (NTS), and Drill Seeding (DS). Both MTS and NTS showed the highest biomass among the seeding methods. The rice yield of MTS and NTS significantly increased compared to conventional fertilization (CF). Also soil properties including organic matter and bulk density were improved by incorporation of hairy vetch. Therefore, we suggested that MTS and NTS could be used to produce hairy vetch and rice in paddy.
