Effects of supplements of folic acid, vitamin B12, and rumen-protected methionine on whole body metabolism of methionine and glucose in lactating dairy cows.

The present experiment was undertaken to determine the effects of dietary supplements of rumen-protected methionine and intramuscular injections of folic acid and vitamin B(12), given 3 wk before to 16 wk after calving, on glucose and methionine metabolism of lactating dairy cows. Twenty-four multiparous Holstein cows were assigned to 6 blocks of 4 cows each according to their previous milk production. Within each block, 2 cows were fed a diet estimated to supply methionine as 1.83% metabolizable protein, equivalent to 76% of methionine requirement, whereas the 2 other cows were fed the same diet supplemented daily with 18 g of rumen-protected methionine. Within each diet, the cows were administrated either no vitamin supplement or weekly intramuscular injections of 160 mg of folic acid plus 10 mg of vitamin B(12.) To investigate metabolic changes at 12 wk of lactation, glucose and methionine kinetics were measured by isotope dilution using infusions of 3[U-(13)C]glucose, [(13)C]NaHCO(3) and 3[1-(13)C,(2)H(3)] methionine. Milk and plasma concentrations of folic acid and vitamin B(12) increased with vitamin injections. Supplementary B-vitamins increased milk production from 34.7 to 38.9 +/- 1.0 kg/d and increased milk lactose, protein, and total solids yields. Whole-body glucose flux tended to increase with vitamin supplementation with a similar quantitative magnitude as the milk lactose yield increase. Vitamin supplementation increased methionine utilization for protein synthesis through increased protein turnover when methionine was deficient and through decreased methionine oxidation when rumen-protected methionine was fed. Vitamin supplementation decreased plasma concentrations of homocysteine independently of rumen-protected methionine feeding, although no effect of vitamin supplementation was measured on methionine remethylation, but this could be due to the limitation of the technique used. Therefore, the effects of these B-vitamins on lactation performance were not mainly explained by methionine economy because of a more efficient methylneogenesis but were rather related to increased glucose availability and changes in methionine metabolism.

Effect of dry period management on mammary gland function and its endocrine regulation in dairy cows.

The objective of this study was to evaluate the effect of shortening the dry period on the mammary gland and the hormonal regulation of its functions. Holstein cows (n = 18) were assigned to a short dry period (SDP; 35 d; n = 9) or a conventional dry period (CDP; 65 d; n = 9). All cows were fed the same diets, with the exception that, during the dry period, the SDP cows received only the pre-calving diet for 35 d, whereas the CDP cows were fed a high-fiber diet from 65 to 28 d before calving and then received the same pre-calving diet as the SDP cows. Mammary gland functional capacity was evaluated at 70 days in milk, and mammary biopsies were taken in early and midlactation. Dry period length averaged 64.3 ± 1.1 and 31.9 ± 1.0 d for the CDP and SDP cows, respectively. The SDP cows had a lower milk yield and a lower energy-corrected milk yield compared with the CDP cows. The SDP cows also had a lower dry matter intake from wk 5 to 20 of lactation and tended to have lower plasma concentrations of β-hydroxybutyrate from wk 1 to 4. Prepartum serum progesterone and estradiol concentrations were unaffected by the dry period management. Serum growth hormone concentrations and milking-induced prolactin release were similar in both groups. However, during the period when the CDP cows were dry but the SDP cows were still being milked (wk -9 to -6), serum prolactin concentrations were higher in the SDP cows than in the CDP cows. The SDP cows had a lower milk BSA content than the CDP cows after the dry period and similar milk lactose concentrations, suggesting that their mammary tight junctions were closed following parturition and, therefore, that the later stage of their lactogenesis was not impaired by SDP management. In early and midlactation, mammary cell apoptosis and proliferation rates as well as mammary expression of genes involved in the function of this tissue were unaffected by the dry period management strategy. For cows in their second lactation, mammary gland functional capacity at 70 d in milk tended to be lower in the SDP cows. In conclusion, even though SDP management decreased milk production during the subsequent lactation, it did not affect mammary cell activity. Although direct evidence is still lacking, decreased mammary cell growth during the dry period is likely responsible for this negative effect. The higher prolactin concentrations in lactating cows during late gestation could be involved in this effect. More research is needed to test these hypotheses.

Complete-lactation milk and component yields following a short (35-d) or a conventional (60-d) dry period management strategy in commercial Holstein herds

A total of 850 cows distributed among 13 commercial Holstein herds were involved in this study to compare the effects of 2 different dry period (DP) management strategies on milk and component yields as well as body condition score (BCS) over complete lactations. Within each herd and every 2 mo, cows were assigned to a short (35 d dry; SDP) or conventional (60 d dry; CDP) DP management based on previous lactation 305-d milk yield, predicted calving interval, and parity: primiparous (n=414) and multiparous (n=436). Cows assigned to CDP were fed a far-off dry cow ration from dry-off until 21 d prepartum, and were then switched to a precalving ration. Cows assigned to SDP were fed the precalving ration throughout their DP. Rations were different across herds, but the late-lactation, precalving, and early lactation rations were identical for both treatment groups within each herd. Additional milk was obtained at the end of lactation from cows assigned to SDP due to the extended lactation. Average daily milk yield in the following lactation was not different between treatments for third- or greater-lactation cows, but was significantly decreased in second-lactation SDP cows. However, when expressed as energy-corrected milk, this difference was not significant. Although lower for primiparous than multiparous cows, body weight and BCS were not affected by DP management strategy. Milk production and BCS responses to treatments varied among herds. Results from the present study suggest that a short DP management strategy could be more appropriate for todays dairy cows, although not suitable for all cows or all herds.

Influence of methionine supply on the response of lactational performance of dairy cows to supplementary folic acid and vitamin B12.

The present experiment was undertaken to determine if the effects of supplementary folic acid on lactational performance were caused by improved methylneogenesis and if the supply in vitamin B(12) could affect this metabolic pathway. In this eventuality, supplementary Met, a major source of preformed methyl groups, should reduce the requirements for these vitamins. Sixty multiparous Holstein cows were assigned to 10 blocks of 6 cows each according to their previous milk production. Within each block, 3 cows were fed a diet estimated to supply Met as 1.83% metabolizable protein and 3 cows were fed the same diet supplemented with 18 g of rumen-protected methionine (RPM) to supply Met as 2.23% of metabolizable protein. Within each level of Met, cows received no vitamin supplement or weekly intramuscular injections of 160 mg of folic acid alone or combined with 10 mg of vitamin B(12) from 3 wk before to 16 wk after calving. There was no treatment effect on dry matter intake during pre- and postcalving periods: 13.4 +/- 0.4 and 21.8 +/- 0.4 kg/d, respectively. Milk production was not affected by RPM supplementation. Folic acid and vitamin B(12) given together tended to increase milk production during the 16 wk of lactation. This effect was more pronounced during the first 4 wk of lactation: 37.5, 37.7, and 40.3 +/- 0.9 kg/d for cows receiving no vitamin supplement, folic acid alone, or folic acid combined with vitamin B(12), respectively. Milk fat yield was not affected by treatments. Lactose, crude protein, and total solid yields were greater, in early lactation, in cows injected with folic acid and vitamin B(12) together but this effect diminished as lactation progressed. Intramuscular injections of folic acid alone or combined with vitamin B(12) tended to decrease plasma concentrations of homocysteine from 5.51 microM with no vitamin supplement to 4.54 and 4.77 +/- 0.37 microM, respectively. Results of the present experiment suggest that the effects of the combined supplement of folic acid and vitamin B(12) on lactational performance of dairy cows were not due to an improvement in methyl groups supply, because RPM supplement, a source of preformed methyl groups, did not alter the cow responsiveness to vitamin supplements.

Incidence of metabolic disorders and reproductive performance following a short (35-d) or conventional (60-d) dry period management in commercial Holstein herds.

A total of 850 Holstein cows from 13 commercial dairy herds were involved in the present study to compare the effects of 2 different dry period (DP) management strategies on health and reproductive parameters. Cows were assigned to either a short (SDP; 35-d) or a conventional (CDP; 60-d) DP management within each herd, based on previous 305-d milk yield, parity (414 primiparous and 436 multiparous), and estimated calving interval. Cows assigned to CDP were fed a dry cow ration from dry-off until 21 d prepartum, and were then switched to a precalving ration. Cows assigned to SDP were fed the precalving ration throughout their DP. Rations were specific to each herd. A significant treatment × parity interaction was found for culling rate. Dry period management did not affect culling rate for second-lactation cows but a significantly higher culling rate occurred in multiparous CDP cows compared with SDP (42.6 vs. 31.6% ± 3.7 for CDP and SDP, respectively). Management used in the DP did not affect incidence of severe ketosis, displaced abomasum, milk fever, and mastitis, although incidence of these metabolic disorders were lower in second-lactation than third- or greater-lactation cows. The incidence of mild ketosis (evaluated by milk ketone concentration) was lower following SDP, probably as a result of better energy balance. On the other hand, the incidence of retained placenta was higher in multiparous cows assigned to SDP, but the reason for this increase remains unclear. Nevertheless, this did not lead to increased incidence of metritis. Moreover, DP management did not influence reproductive measures, including days in milk at first breeding, number of breedings per conception, as well as conception rates at first and second services. Regarding days open, overall, all 13 herds were not significantly affected by treatment, but 1 herd clearly showed opposite results to the 12 others. Our results indicate that a short DP management strategy could facilitate transition from one lactation to the next by decreasing the incidence of mild ketosis, with no major negative effects on other health parameters and reproduction. The variation in results observed among herds suggests that other management practices influence the response observed following a short or conventional DP, emphasizing the need for other field studies.

CHANGES IN SERUM CONCENTRATIONS OF FOLATES, PYRIDOXAL, PYRIDOXAL-5-PHOSPHATE AND VITAMIN B12 DURING LACTATION OF DAIRY COWS FED DIETARY SUPPLEMENTS OF FOLIC ACID

The present experiment was undertaken to determine the effects of dietary supplements of folic acid (FA) administered from 4 wk prepartum to 305 d of lactation on serum concentrations of folates, vitamin B12, pyridoxal-5-phosphate (P-5-P) and pyridoxal. In addition, concentrations of blood hemoglobin and packed cell volume (PCV) were determined. Sixty-three Holstein cows were assigned to 22 incomplete blocks according to lactation number, milk production and body weight (BW). Within each block, cows received 0, 2 and 4 mg FA d−1 kg−1 BW. Dietary supplements of FA increased serum concentrations of folates (time × FA, P = 0.0001); the highest concentrations were observed during the first two months of lactation in cows fed supplementary FA. The lowest serum concentrations of vitamin B12 were also observed during the first two months of lactation, and the concentrations were lower in primiparous than in multiparous cows (time × parity, P = 0.03). Effects of the forage to concentrate ratio on B-vitamin concentrations in different ruminal fractions of dairy cows

Two studies were undertaken to verify the effect of the forage to concentrate ratio of the diet on B-vitamin concentrations in ruminal contents. In Study 1, eight primiparous and eight multiparous cows were used in a cross-over design, and concentrations of biotin, folates and vitamin B12 were determined in ruminal fluid and plasma of cows fed a high-forage (HF; 58:42 forage to concentrate ratio; DM basis) or a low-forage (LF; 37:63 forage to concentrate ratio; DM basis) diet. In Study 2, six ruminally cannulated lactating cows were used in a cross-over design to evaluate the effects of forage to concentrate ratio (HF = 60:40; LF = 40:60; DM basis) on concentrations of seven B-vitamins in the particle-free fluid and in both liquid- and solid-associated bacteria. Results showed that B-vitamins were present mainly in the bacterial fractions of the ruminal content, while only limited amounts were found in the surrounding fluid. Apparent ruminal synthesis and intestinal disappearance of vitamin B12 and its analogs in dairy cows.

The aim of the project was to calculate the apparent synthesis or destruction of cobalamin (vitamin B(12)) and its analogs in the rumen as well as their apparent intestinal disappearance in dairy cows. Four lactating cows were fed a diet supplemented with cobalt alone (0.76 mg/kg of DM; control) or with cobalt and vitamin B(12) (cyanocobalamin, 500 mg/d; treated). In addition to cobalamin, the only biologically active molecule for the cow, 7 analogs were identified in duodenal and ileal digesta: cobinamide, which lacks the base, ribose, and phosphate groups; and 6 other molecules in which the base, 5,6-dimethylbenzimidazole, is replaced by cresol, 2-CH(3)-adenine, adenine, 2-CH(3)-S-adenine, or 5-OH-benzimidazole, or an unidentified cobamine. Small amounts of cobalamin and cobinamide were detected in the total mixed ration, but apparent synthesis of all forms took place in rumen. During the control period, cobalamin represented 38% of the total amounts of corrinoids produced in rumen. Approximately 11% of the average daily intake of cobalt was used for apparent ruminal synthesis of corrinoids, of which only 4% was incorporated into cobalamin. Only 20% of the supplement of cyanocobalamin was recovered at the duodenal level; cobinamide appeared to be the major product of degradation of supplementary cyanocobalamin in the rumen. During the control and treatment periods, there was an apparent intestinal disappearance of cobalamin and 5-OH-benzimidazole cobamide only; only the apparent intestinal disappearance of cobalamin differed between the 2 periods. Although cobalamin was not the major form synthesized by ruminal microflora and, even if supplementary cyanocobalamin was extensively destroyed by ruminal microflora, based on calculations of apparent intestinal disappearance, cobalamin seems to be the major form absorbed in the small intestine.

Effects of supplementary folic acid and vitamin B12 on hepatic metabolism of dairy cows according to methionine supply1

The present experiment was undertaken to study the interactions between dietary supplements of rumen-protected methionine (RPM) and intramuscular injections of folic acid and vitamin B(12), given from 3 wk before calving to 16 wk of lactation, on hepatic metabolism of lactating dairy cows. Sixty multiparous Holstein cows were assigned to 10 blocks of 6 cows each according to their previous milk production. Within each block, 3 cows were fed a diet calculated to supply Met as 1.83% of metabolizable protein, whereas the 3 other cows were fed the same diet supplemented with 18g of RPM calculated to provide Met as 2.23% of metabolizable protein. Within each level of Met, the cows received no vitamin supplement or weekly intramuscular injections of 160mg of folic acid alone or combined with 10mg of vitamin B(12). Liver biopsies were taken at 2, 4, 8, and 16 wk of lactation. Liver concentrations of folates and vitamin B(12) were increased by their respective supplements but this response to vitamin supplements was altered by methionine supply. Concentrations of total lipids and triglycerides increased in livers of cows fed RPM, whereas concentrations of cholesterol ester, cholesterol, diglycerides, phosphatidylethanolamine, and phosphatidylcholine were not affected. Folic acid, alone or combined with vitamin B(12), tended to increase the ratio of phosphatidylcholine to phosphatidylethanolamine. Gene expression of 5,10-methylene-tetrahydrofolate reductase, microsomal transfer protein, and phosphatidylethanolamine methyltransferase were higher in liver of cows fed RPM supplements. The relative mRNA abundance of 5,10-methylene-tetrahydrofolate reductase and methylmalonyl-CoA mutase were increased by the combined injections of folic acid and vitamin B(12), whereas those of methionine synthase and methionine synthase reductase were not affected by treatments. These results suggest that increasing supply of methyl groups, as preformed labile methyl groups or through methylneogenesis, affected the methylation cycle but had a limited effect on dairy cow performance. The observed effects of the combined supplement of folic acid and vitamin B(12) on lactational performance of dairy cows probably result from an improvement of energy metabolism during early lactation.

The role of folic acid in the nutrition of gestating and lactating primiparous sows

Abstract This experiment aimed to evaluate the folic acid requirements of gestating and lactating gilts. Three groups of gilts received, during gestation, a diet supplemented with either 0, 5 or 15 mg/kg of folic acid. At parturition, each of these groups was subdivided into three subgroups. In each subgroup, the lactation diet was supplemented with 0, 5 or 15 mg/kg of folic acid. The level of folic acid during gestation had no effect on the body weight gain of gilts during gestation but increased linearly ( P ≤ 0.02) the body weight loss during lactation. During gestation and lactation, addition of folic acid increased ( P ≤ 0.05) the concentrations of serum folates. Litter size at parturition and at weaning was not affected by the treatments, although it was numerically higher in sows receiving 15 mg/kg of folic acid during gestation. Milk yield and milk fat, protein and lactose were not influenced by treatments. Concentration of milk folates measured at 1 and 3 weeks of lactation increased ( P ≤ 0.001) with the level of folic acid in the lactation diet; an effect of the gestation treatments was observed at 1 week ( P ≤ 0.001) but had disappeared by the third week of lactation. In piglets, the concentration of serum folates tended to follow the changes observed on milk folates. Growth of piglets and total litter weight from birth to 8 weeks of age increased linearly (P ≤ 0.05) with the level of folic acid in the gestation diet. Therefore, a dietary supplement of 15 mg/kg of folic acid was necessary to avoid the drop in concentrations of serum folates of sows in gestation and seemed to influence some traits of the reproductive performance.
