Mast cells populations fluctuate along the spinal dura mater of the developing rat

The present study reveals developmental changes in the number, the phenotype and the distribution pattern of mast cells (MCs) along the cervical, the thoracic and the lumbar parts of the spinal dura mater. Postnatal infiltration of spinal dura by MCs does not appear to follow a sequential developmental pattern and meningeal MCs are unevenly distributed along the various parts of the examined dura. At each spinal level, areas most densely populated by MCs are the dorsal dura and the dural sleeves of the dorsal (sensory) spinal roots The developmental time course of the total MCs number is characterized by significant fluctuations in all three parts examined, with notable increases at P1, P4, P21 and P60 (peak value) for the cervical part, at P1 (peak value), P7 and P21 for the thoracic part and at P1, P7 (peak value) and P30 for the lumbar part. At P180, MCs number declines to 56%, 33% and 13% of the peak values for the cervical, the thoracic and the lumbar part, respectively. However, a different developmental pattern is followed by each subpopulation of MCs identified on the basis of their staining characteristics, namely connective tissue type mast cells (CTMCs), mucosal type or cells with characteristics of immature mast cells (MTMCs) and mixed type MCs, in each part examined. The findings may be of importance in elucidating physiological and pathological processes in the dura mater and the vertebral column.

Parallel development of blood vessels and mast cells in the lateral geniculate nuclei

The present study examined quantitatively developmental changes of the vasculature in the dorsal (dLGN) and the ventral (vLGN) lateral geniculate nuclei together with concomitant changes in the number of mast cells (MCs), known for their role in angiogenesis. Vascular network, marked after transcardial perfusion of India ink, and MCs detected with conventional histochemical techniques were examined at postnatal days (P) 1, 8, 14, 21, 31, 90 and 300 of Wistar rats. Quantitative analysis by means of an image analysis system showed age-dependent changes in both vascular parameters [vascular area and relative frequency (%) of capillaries and medium- and large-diameter vessels] and mast cells number in the developing dLGN and vLGN. Despite quantitative differences in the vascularization and MC infiltration between the two nuclei at some age points, MC number, vascular area and the percentage frequency of capillaries exhibited similar developmental time courses, especially up to the end of the first postnatal month. Both MC number and the capillary frequency reached maximal levels at P31 and declined thereafter, following a massive or a partial, respectively, decrease up to P300.

The role of an attenuated anticoccidial vaccine on the intestinal ecosystem and on the pathogenesis of experimental necrotic enteritis in broiler chickens

The objective of the present study was to investigate the effect of an attenuated anticoccidial vaccination on the intestinal ecosystem and on the pathogenesis of experimental necrotic enteritis (NE) in broiler chickens. Two hundred and forty 1-day-old Cobb 500 broiler chickens were randomly allocated to four treatment groups according to the following experimental design: control Group N; Group PN, where birds were vaccinated with anticoccidial vaccine; Group M, where birds were challenged with Clostridium perfringens and with Eimeria maxima; and Group PM, where birds were both vaccinated and challenged. From each bird, the intestine, gizzard and liver were scored for gross NE lesions. Intestinal digesta were collected for pH and viscosity determination. Samples from the gastrointestinal tract and liver were taken for microbiological analysis. Evaluation of the experimental data revealed that Group M had significantly higher overall mean NE intestinal lesions compared with Group PM. Viscosity values of jejunum digesta as well as pH values of the duodenum, jejunum and ileum digesta in Group M were significantly lower compared with Group PM. C. perfringens counts in the caeca of Group PM were significantly lower compared with Group M. The milder decrease of pH and viscosity values of intestinal content and the reduction of C. perfringens counts in the caeca in challenged and vaccinated birds may explain the lower score of NE gross intestinal lesions and may suggest a positive effect on intestinal ecosystem and a significant protective effect of attenuated anticoccidial vaccination against NE in a subclinical experimental model.

Developmental changes of mast cell populations in the cerebral meninges of the rat

It is known that both the dura and the pia mater attract and support the differentiation of mast cells. The present study shows that unevenly distributed mast cells in the cerebral meninges of the rat can be found in perivascular sites and vessel ramification points, but can also be unrelated to the meningeal vasculature. It also documents changes in the number, localization and staining preferences of the mast cells in the two meninges of the developing and mature rat brain. Quantitative examination of all types of histochemically differentiated meningeal mast cells reveals no major (although some exist) differences between right and left side subpopulations, but strongly suggests a different origin and fate of the dural and the pial mast cells. The number of dural mast cells, already high from postnatal day 0, although declining from postnatal day 21 onwards, remains conspicuous up to postnatal day 180. In contrast, pial mast cells are comparatively very few in the first day of the postnatal life, and despite a transient significant increase in the following two weeks, they reach almost zero levels from postnatal day 21.

High stocking density as a predisposing factor for necrotic enteritis in broiler chicks

Stocking density is a management factor which has critical implications for the poultry industry. The aim of the present study was to investigate the effect of high stocking density as a predisposing factor in an experimental model of necrotic enteritis in broiler chicks. The experimental challenge model included an oral inoculation with 10-fold dose of attenuated anticoccidial vaccine and multiple oral inoculations with a specific strain of Clostridium perfringens. Two hundred and forty as hatched day-old broiler chicks were randomly allocated to four treatment groups according to the following experimental design: group N, with normal stocking density (15 birds/m2) and no challenge; group D, with high stocking density (30 birds/m2) and no challenge; group P, with normal stocking density and positive challenge; and group DP, with high stocking density and positive challenge. From each bird, the intestine, gizzard and liver were collected and scored for gross lesions. The intestinal digesta was collected for pH and viscosity determination. One caecum from each bird was taken for microbiological analysis. The statistical analysis and evaluation of the experimental data revealed significant interaction effects between “stocking density” and “challenge”, regarding gross lesion scores in intestine and liver, pH values in jejunum, ileum and caeca as well as C. perfringens counts in the caeca (P ≤ 0.05). High stocking density in challenged birds increased the gross lesion score in the intestine (P ≤ 0.05), contrary to unchallenged birds. It can be concluded that high stocking density affects unfavourably the welfare and gut health of broiler chicks, predisposes to necrotic enteritis in a subclinical experimental model and increases further its importance as a management factor for the poultry industry.

Developmental changes in the vascular network of the rat visual areas 17, 18 and 18a

The present study examines quantitatively the areal and the laminar fluctuations of the vascular network in the visual areas 17, 18 and 18a of the rat cerebral cortex, from postnatal day (P) 1 to P60. For this purpose, the detailed vascular networks of the visual areas, marked after transcardial perfusion of India ink, are analyzed with the use of an image analysis system in order to measure the total vascular density (VD) and the relative density of capillaries (CD), of medium (MD)- and large (LD)-sized vessels in combination with changes in the mean diameter of all three types of vessels. Comparative quantitative microscopy showed that both VD and CD do not exhibit significant interareal differences in the adult rat brain. However, while VD reaches adult values much earlier in area 18a (P21) than in areas 17 and 18 (P60), CD obtains adult values at P31 in areas 17 and 18a, but later (P60) in area 18. Maturation process of laminar VD, CD, MD and LD was not found to follow a simple (i.e. inside-out or mediolateral) sequence, and, in each cortical area, laminar fluctuations of vessels density revealed a complicated developmental pattern, which might be attributed to their changing structural and functional status. Developmental changes in the diameter of capillaries, examined in conjunction with concomitant changes of vascular and capillary density in each area, suggest the existence of angiogenesis in all three visual areas during the third postnatal week of age.

Analysing consumers’ perceived differences in wild and farmed fish

Purpose – The purpose of this paper is to investigate consumers’ attitudes and behaviours towards wild and farmed fish, in order to identify possible distinct consumer groups, and to examine potential linkages between characteristics of the consumers’ demographic and socio-economic status and marketing aspects in wild and farmed fish. Design/methodology/approach – Using data from an in-person field survey, a TwoStep cluster analysis was employed in order to detect perceived differences among consumers with different profiles. Findings – The analysis identified two distinct consumer groups differentiated primarily by income: the low-potential aquaculture consumers and the high-potential aquaculture consumers, representing 67 and 33 per cent of the total sample, respectively. The study provides evidence that there is a lesser preference towards farmed fish. The effect of cold stress on the pathogenesis of necrotic enteritis in broiler chicks

Cold stress is a physical environmental stressor with significant effect on the poultry industry. The aim of the present study was to investigate the effect of cold stress as a predisposing factor for necrotic enteritis in broiler chicks. The experimental challenge model included an oral inoculation with 10-fold dose of attenuated anticoccidial vaccine and multiple oral inoculations with a specific strain of Clostridium perfringens. Birds were either challenged or not as described above, and either exposed or not to repeated cold stress (15°C for 12 h/day for 4 days). From each bird, intestinal gross lesions were scored and intestinal digesta pH and viscosity were measured. C. perfringens was counted in the caecum. The statistical analysis and evaluation of the experimental data revealed that the cold stress in challenged birds significantly increased the incidence and the severity of necrotic enteritis lesions (Ρ ≤ 0.05), while causing no lesions in unchallenged birds. Moreover, the cold stress caused a significant increase (Ρ ≤ 0.05) in the pH and C. perfringens counts in the caeca. The study provides evidence that cold stress increased the susceptibility to necrotic enteritis in a subclinical experimental model and thus should be regarded as a physical environmental stressor that could significantly affect the welfare, health and intestinal ecosystem of broiler chicks.

Methodologies for solving the integration problem of agricultural enterprise applications: Agents, web services and ontologies

The great expansion of Information Technology (IT) creates many new vital challenges for the future of the enterprises in the agri-food sector. In fact, many members in the agri-food sector have realised the challenges and that adoption of IT is crucial for their survivor. For this reason, different types of Agricultural Information Systems (AISs) have been developed. Unfortunately, one issue which seems to attract less attention is the integration of all these AIS. We believe that integration of individual AISs is important because in today’s globalised environment interoperability is essential in creating “complete” and effective solutions based on AISs. In this paper we argue that the issue of integration should be of greater focus in the development of AISs in the future. Furthermore, we discuss existing methodologies to approach the issue of integration such as agents, ontologies and web services.

Consulting for Sustainable Development, Information Technologies Adoption, Marketing and Entrepreneurship Issues in Livestock Farms☆

Consulting agricultural services constitute a major key for implementing successfully sustainable development EU goals in rural areas. Today, in the context of the Rural Development Programme 2007-2013, there are initiatives for funding crop and livestock farms and entrepreneurship. Presently, there are bureaus offering consulting services and reliable information to t he Greek farmers regarding Information Technologies adoption and innovative e-tools, marketing and processing of agricultural products, certificated agro-products (ISO), modernization of agricultural holdings, etc. Farmers have the opportunity to participate in EU funded measures that support the development of new activities, such as innovative actions, marketing services, new information technologies software and web tools and promote production innovations through specific investments. This paper aims to study and evaluate the adoption of consulting from livestock farms and discuss future opportunities. The study is based on primary data collected from the prefecture of Rodopi, in Thrace/Northeastern Greece, through a farm management survey which was carried out during September and December 2011. A sample of 120 livestock farmers between the ages of 20 and 70 was surveyed for the empirical application of this study using a well-structured questionnaire. The survey took place in private consulting bureaus and locally at farms. The collected data were coded and statistically analyzed using SPSS. Data was classified with cluster analysis in order to identify and describe groups with common attitudes among farmers. Findings show that 54% of the farmers in the sample are interested in consulting and adopt information technology solutions, marketing and entrepreneurship issues, aiming to sustainability in the context of EU goals. The results also indicate that 63% of the farme rs have already participated in a program, while 37% have participated in the schemes promoting the establishment of young farmers. Moreover, 55% of them would prefer to be consulted by private consulting bureaus rather than government consulting divisions. Private consulting bureaus satisfy farmers who consider them as a significant support in their attempt to achieve the contemporary EU goals for sustainable development through the adoption of information technologies tools and innovative trade and marketing techniques. Farmers can be further classified in two groups that share similar either innovative or traditional aspects about Greek agriculture trends, in relation to their social characteristics.
