Exploration of the hypothalamic–pituitary–adrenal function as a tool to evaluate animal welfare

Measuring HPA axis activity is the standard approach to the study of stress and welfare in farm animals. Although the reference technique is the use of blood plasma to measure glucocorticoid hormones (cortisol or corticosterone), several alternative methods such as the measurement of corticosteroids in saliva, urine or faeces have been developed to overcome the stress induced by blood sampling itself. In chronic stress situations, as is frequently the case in studies about farm animal welfare, hormonal secretions are usually unchanged but dynamic testing allows the demonstration of functional changes at several levels of the system, including the sensitization of the adrenal cortex to ACTH and the resistance of the axis to feedback inhibition by corticosteroids (dexamethasone suppression test). Beyond these procedural aspects, the main pitfall in the use of HPA axis activity is in the interpretation of experimental data. The large variability of the system has to be taken into consideration, since corticosteroid hormone secretion is usually pulsatile, follows diurnal and seasonal rhythms, is influenced by feed intake and environmental factors such as temperature and humidity, age and physiological state, just to cite the main sources of variation. The corresponding changes reflect the important role of glucocorticoid hormones in a number of basic physiological processes such as energy metabolism and central nervous system functioning. Furthermore, large differences have been found across species, breeds and individuals, which reflect the contribution of genetic factors and environmental influences, especially during development, in HPA axis functioning. Usually, these results will be integrated with data from behavioral observation, production and pathology records in a comprehensive approach of farm animal welfare.

The LayWel project: welfare implications of changes in production systems for laying hens

The conditions under which laying hens are kept remain a major animal welfare concern. It is one of the most intensive forms of animal production and the number of animals involved is very high. Widespread public debate has stimulated the call for more animal friendly, alternative systems to barren conventional cages. Directive 1999/74/EC has encouraged technical changes in current systems. Not only have traditional cages been modified (so-called ‘enriched cages’), but also new alternative systems (e.g. aviaries) have been developed. There is an ongoing need to evaluate the actual welfare status of hens in these novel systems including those on commercial farms. The LayWel project, was funded via the European Commissions Sixth Framework Programme and national funding from several EU countries. Its general objective was to produce an evaluation of the welfare of laying hens in various systems, with special focus on enriched cages, and to disseminate the information in all member states of the EU and associated countries. The project took into account pathological, zootechnical, physiological and ethological aspects. A major achievement of the LayWel project was the compilation of a database collecting data from different housing systems and thus enabling data comparison. The project partners recommend that support is given to maintaining the database in the future so that data can be more reliably modelled. As the type of data collected did not often allow a formal statistical analysis the evaluation of welfare was a presentation of risk factors and advantages and disadvantages of various housing systems. Conclusions are that, with the exception of conventional cages, all systems have the potential to provide satisfactory welfare for laying hens. However this potential is not always realised in practice. Among the numerous explanations are management, climate, design, different responses by different genotypes and interacting effects. A second major achievement of the project was the development of feather scoring and integument (skin, head and feet) scoring systems together with comprehensive sets of photographs. It is recommended that the integument scoring systems are widely adopted and used in on-going research. Farms should also routinely and frequently carry out integument scoring to assist in the detection of damaging pecking, which is currently a widespread welfare problem. Within LayWel an on-farm auditing procedure was developed in the form of a manual for self-assessment. The manual first explains what is meant by welfare and outlines the relevance of welfare assessment. It also summarises risks to welfare in the main categories of housing system. The second part contains recording forms, with guidance for assessing hen welfare. These enable regular checks of a range of indicators of laying hen welfare to be carried out systematically. The indicators were chosen to be relevant to hen welfare as well as feasible and reliable to apply in practice. A series of conclusions and recommendations were made on various aspects of housing systems, behaviour, health and mortality and other matters in relation to bird welfare. Behavioural and physiological responses of three chicken breeds to pre-slaughter shackling and acute heat stress.

1. The aim of this study was to compare the behavioural and physiological responses to hanging and acute heat stress in three different chicken breeds. Chicks were obtained from a slow-growing French ‘Label Rouge’ line (SGL), a fast-growing standard line (FGL) and a heavy line (HL). The SGL, FGL and HL birds were slaughtered at their respective market ages of 12, 6 and 6 weeks, in an attempt to achieve similar body weights. Before stunning, birds were either shackled by their legs on the moving line for 2 min (shackling stress: SH) or placed in a room at 35°C and 60% of humidity for 3·5 h and then shackled for 2 min (acute heat stress plus shackling: H + SH) or subjected to minimal stress by shackling for 10 s before stunning (control group: C). 2. Bird physiological responses to the three pre-slaughter treatments were estimated by measuring blood corticosterone, glycaemia, creatine kinase activity, acid–base status and electrolyte concentration as well as lactate content and glycolytic potential in the breast (Pectoralis major) and thigh (Ilio tibialis) muscles. Behavioural responses to shackling stress were evaluated by measuring wing flapping duration, straightening up attempts and vocalisations. 3. Blood corticosterone was higher in SH and H + SH groups than in the C group, regardless of genotype. The struggling activity on the shackle line differed among chicken breeds. It was more intense and occurred more rapidly after hanging in the SGL birds than in both other breeds. Furthermore, SGL struggling activity was not affected by hanging duration while it increased with hanging duration in FGL and HL birds. 4. Wing flapping duration was negatively correlated with blood pH, bicarbonate concentration and positively correlated with breast muscle lactate content, indicating that struggling stimulated ante-mortem glycolysis activity in breast muscle. Acute heat stress affected blood Ca2+ and Na+ concentration and increased glycaemia and glycolytic potential of thigh muscle. 5. Both acute heat stress and shackling before slaughter were experienced as stressful events by all types of birds.

Welfare of ducks in European duck husbandry systems

European duck meat production is based on the use of Pekin, Muscovy and Mule duck genotypes that vary in their behavioural and physiological characteristics. Furthermore, large differences exist in their housing and management conditions. The aim of this review is to discuss the welfare of these different genotypes in typical husbandry systems, focusing on ducks kept for meat production. Firstly, factors that can affect duck welfare, such as stocking density and group size, access to straw, an outdoor run, or open water, are described. Secondly, welfare problems such as feather pecking, fear and stress, and health problems are assessed. Thirdly, the various systems used in Europe are described for these aspects. Giving ducks access to straw, an outdoor run, or open water increases the behavioural opportunities of the ducks (foraging, preening, bathing, and swimming), but can also lead to poor hygiene and increased health- and food safety risks. Therefore, practical solutions that allow expression of natural behaviour, but do not lead to hygiene or health problems have to be found and some practical suggestions are provided.

Factorial correspondence analysis of fear-related behaviour traits in Japanese quail

Factorial correspondence analysis was performed on 341 quails from a F2 cross between two lines divergently selected on the duration of tonic immobility over 29 generations. Several fear- or stress-related traits were recorded, i.e. tonic immobility duration, number of inductions needed to induce tonic immobility, open-field behaviour (time spent walking, latency before first movement and number of defecations), asymmetry of tibia lengths and corticosterone concentration after restraint stress. Variables were categorised in classes and analysed by factorial correspondence analysis. The first axis was mostly described by open-field behaviour, and the second by tonic immobility traits (duration of tonic immobility and number of inductions), which showed that these behaviours were almost independent. No relationship was found between axes of the factorial correspondence analysis and corticosterone concentration or asymmetry of tibia lengths, showing that these variables reflected other characteristics of stress susceptibility than those described by tonic immobility and open-field behaviour. These results show that reaction to stress of quails is a multidimensional trait and cannot be summarised by one trait.

Major welfare issues in broiler breeders

Under current practices, broiler parent stock (broiler breeders) encounter several welfare problems, such as feed restriction and injury during mating. Intensive selection for production traits, especially growth rate, is associated with increased nutritious requirement and thus feed consumption, but also reproductive dysfunctions and decreased sexual activity in broiler breeders. A resulting serious welfare problem is the subsequent severe feed restriction which is applied during rearing, in order to prevent health problems and to achieve better egg production. This severe feed restriction has negative effects on bird welfare as it causes chronic stress resulting from hunger. Recent research has focused on management practices to alleviate the negative effect of this feed restriction. Using both a more fibrous feed and an appetite suppressant seems most successful. There are welfare concerns involved in the mating of broiler breeders. In many countries mutilations (beak trimming, despurring and toe clipping) are carried out in male broiler breeders as a standard procedure to prevent excessive damage to the hens during mating. It has been reported that broiler breeder males show low libido but, at the same time, may be aggressive and show rough behaviour during mating, which can cause severe injuries to the hens, although some injuries may be caused by over-mating hens. Current studies aim to develop management practices to reduce hen injury due to over-mating and improve the sexual behaviour in both males and females. In the last part of the review, an examination of how breeding companies are taking welfare into consideration in their commercial selection scheme is made. In practice, although for example reduced mortality and some health criteria are included in selection programs and some alternative breeds as used, other welfare issues remain, such as feed restriction during rearing.

Influence of Stimuli from Chicks on Behavior and Concentrations of Plasma Prolactin and Luteinizing Hormone in Incubating Hens

Removal of eggs from the nests of incubating birds or substitution of eggs for chicks disrupts incubation behavior and induces changes in the secretion of prolactin and luteinizing hormone (LH). The aim of the present study was to determine how different stimuli, such as physical contact with eggs and tactile, visual, and/or auditory cues from chicks, interact to control the transition between incubation and brooding and to induce changes in prolactin and LH plasma concentrations. Physical contact with chicks, in the presence or absence of eggs, induced brooding behavior and an immediate fall in plasma prolactin concentration and a gradual increase in LH concentration. Vocalizations, particularly clucking and food calls, increased rapidly while incubation and nest attachment disappeared slowly. No change in plasma prolactin or LH concentration was observed in incubating hens which could hear and see or only hear chicks. These incubating hens showed no interest in chicks and continued to incubate persistently. To conclude, tactile stimuli, alone or in combination with visual and/or auditory stimuli from newly hatched chicks, are the only cues that induce the transition from incubation to brooding and the associated decrease in prolactin secretion and increase in LH secretion.

The past, present and future of force- feeding and "foie gras" production

Force-feeding is a very old practice, first recorded in ancient Egypt, but until the 1950′s foie gras production remained somewhat limited in volume. Foie gras is currently produced in various countries but approximately 80% of world production and consumption takes place in France. Geese, which were the most common specie been force-fed until recently, now account for less than 10% of the total world foie gras production. Ducks such as the Muscovy duck (Cairina moschata) account for less than 5%, and mule ducks for the rest. Thus, over 35 million mule ducks were force-fed in France in 2001, accounting for nearly 95% of the domestic foie gras production. This increase was made possible by technical progress in specific breeding programmes and force-feeding practice. However, the future of this production is uncertain, at least in Europe. Indeed, although a number of experimental approaches have shown that there is no scientific evidence that validates such adverse comment, this procedure is highly criticised in terms of animal welfare. The Council of Europe therefore adopted two specific recommendations in 1999 and although its practice is not banned at present, it is limited to the areas where it is already practised and only under specific rearing conditions. Therefore, the question is: will it be still possible to produce foie gras in France or elsewhere in Europe in the future and, if not, where and how will it be produced?

Effect of one week of stress on emotional reactivity and learning and memory performances in Japanese quail

Chronic stress is known to induce long term alterations of emotional behaviours as well as cognitive performances leading thereby to welfare or husbandry problems. These stress-induced consequences are observed following long periods of stress lasting from several weeks to several years. The current study examined whether a shorter period of stress (one week) produced similar impairing effects. Two-week old Japanese quail were either submitted to a series of aversive events over consecutive 8 days, at unpredictable times each day (treated animals) or left undisturbed (controls). Following the treatment period, animals were weighed and basal as well as aversive events-induced levels of plasma corticosterone were quantified. Quail were also tested for emotional reactivity in three tests (the tonic immobility test, the hole-in-the-wall and novel object tests) and for spatial reference memory. Although there was no difference in corticosterone levels between the two groups, the treated animals had lower body weight than controls. Behavioural investigations after the treatment period did not reveal any difference between the groups in the three emotional reactivity tests. In the spatial task, treated quail displayed enhanced behavioural flexibility as revealed by their higher performances during the reversal phase of the task. The alteration of growth suggests that a short period of repetitive exposures to unpredictable aversive events can be perceived by quail as stressful. Such a stress period can improve spatial learning performances in quail supporting the critical role played by the duration of the stress period on cognitive performance.

Force-feeding procedure and physiological indicators of stress in male mule ducks

1. The effects of the force-feeding procedure and its different components on various physiological indicators of acute and chronic stress were investigated in male mule ducks before and during a 12-d cramming period. 2. The highest concentration of corticosterone were measured after injection of ACTH agonist, during the pre-experimental period when the ducks were still housed in collective floor pens and at the time of transfer. 3. During the cramming period, corticosterone measured before and after force-feeding did not differ significantly though there was a non-significant trend towards an increase in certain cases. 4. The different components of force-feeding, including manipulation, intubation, force-feeding with a standard or a large amount of food had no significant effect upon corticosterone concentrations. 5. There was no indication from ACTH agonist challenge either of a change in adrenal sensitivity or a change in its responsiveness. 6. The heterocyte-lymphocyte ratio measured before and at the end of the cramming period did not differ significantly. 7. In conclusion, we observed no significant indication that force-feeding is perceived as an acute or chronic stress by male mule ducks, in our experimental conditions. Nevertheless, it remains to be shown that their adrenocorticotropic axis is responsive to acute stressors.
