Dioxin contamination and growth and development in great blue heron embryos

A great blue heron colony located near a pulp mill in British Columbia failed to fledge young in 1987, with a concurrent sharp increase in polychlorinated dibenzo-p-dioxin (PCDD) and polychlorinated dibenzofuran (PCDF) levels in their eggs. In 1988 we tested the hypothesis that the PCDD and PCDF contamination caused reproductive failure by increasing mortality of the heron embryos in ovo. Pairs of great blue heron eggs were collected from three British Columbia colonies with low, intermediate, and high levels of dioxin contamination: Nicomekl, Vancouver, and Crofton, respectively. One egg of each pair was incubated under laboratory conditions at the University of British Columbia (UBC) while the other egg was analyzed for PCDDs and PCDFs. All incubated eggs were fertile. All eggs from the Nicomekl colony hatched, while 13 of 14 eggs from Vancouver and 12 of 13 eggs from Crofton hatched. Subcutaneous edema was observed in 4 of 12 chicks from Crofton and 2 of 13 chicks from Vancouver. No edema was seen in the chicks from Nicomekl. There was a small, but significant, negative regression of plasma calcium concentration, yolk-free body weight, tibia length, wet, dry, and ash weight, beak length, and kidney and stomach weight of the hatched chicks on the tetrachlorodibenzo-p-dioxin (TCDD) level of the paired eggs. Fewer down follicles were present on the heads of TCDD-contaminated chicks. Hence while dioxins did not cause mortality of the heron embryos in ovo, the depression of growth and the presence of edema are suggestive that dioxins at the levels found in the environment have an adverse effect on the development of great blue heron embryos.

Comparison of the antioxidant properties of emu oil with other avian oils

The antioxidant properties of emu oil were compared with oils derived from the fat of other avian species. We first examined their free radical scavenging activity against the 2,2-diphenyl-1-picryl hydracyl radical. The concentration of emu oil in the test solution that caused 50% neutralisation (IC50) was variable (24.5 ± 5.9 mg/mL, range 5.3–55.4 mg/mL), but similar to values obtained for other ratites (10.7 ± 5.9 mg/mL). In contrast, the IC50 values for duck and chicken oil were much higher (118.0 ± 8.1 mg/mL). The variability in the radical scavenging activity of emu oil preparations may reflect variations in the diets of the birds, the processing protocol and/or the storage age of the oil. We also evaluated some of the ratite oils for their inhibitory capacity on human erythrocyte membrane oxidation, by measuring the reduction of the thiobarbituric acid-reactive substance (TBAR) production. Emu oil had a greater effect in decreasing TBAR production than either the ostrich or rhea oil, suggesting that it offers more protection than the other ratite oils against oxidative damage. In conclusion, we demonstrated that emu oil has both antioxidant properties in vitro and a protective role against oxidative damage in a model biological membrane system. The antioxidant or radical scavenging properties of emu oil appear to be due to minor constituents in the non-triglyceride fraction of the oil, while its high ratio of unsaturated to saturated fatty acids (UFA : SFA) offers protection against oxidative damage.

Comparison of renal and salt gland function in three species of wild ducks.

SUMMARY Three processes central to osmoregulation of marine birds were compared in three species of ducks that differ in habitat affinity, diet and saline tolerance. These processes are filtration of Na+ and water from the plasma by the kidneys, their reabsorption along the renal tubules, and secretion by the salt glands. Barrows goldeneyes Bucephala islandica, the most marine species, have the highest rates for all three processes and only this species can secrete all the infused salt via the salt glands. Rates of all three processes are lower in mallards Anas platyrhynchos, the most freshwater species. Following saline acclimation, mallards could excrete all the infused Na+ by a combined Na+ excretion of the kidneys and salt glands. Canvasbacks Aythya valisineria, despite being more saline tolerant than mallards, are unable to excrete all the infused Na+. They produce a large volume of urine (like mallards) that has a low [Na+] (like goldeneyes). Salt gland secretion Na+ concentration did not differ among the three species, but only goldeneyes secrete at a rate sufficient to eliminate all infused Na+ via the salt glands. Differences in saline tolerance of these ducks species cannot be fully explained by differences in their filtration, reabsorption and secretion of Na+ and water, suggesting that the intestinal tract plays an important role.

EFFECT OF CADMIUM ON PEKIN DUCK TOTAL BODY WATER, WATER FLUX, RENAL FILTRATION, AND SALT GLAND FUNCTION

The following hypotheses were examined using Pekin ducks (Anas platyrhynchos) as a model for marine ducks: cadmium (Cd) intake affects (1) salt gland and/or kidney function of ducks and (2) osmoregulation differently in male and female ducks. Birds were fed 0, 50, or 300 microg Cd/g food. They were gradually acclimated to 450 mM NaCl and then drank 300 mM NaCl for 3 mo while salt gland secretion (SGS), glomerular filtration rate (GFR), total body water (TBW), and water flux (WF) were measured in ducks eating control and high-Cd diets. Cadmium ingestion did not markedly affect body mass, but significantly enlarged the salt glands and kidneys. Enhancement of kidney mass was greater in males. Cadmium ingestion did not affect TBW or WF, but tended to increase interstitial fluid space at the expense of intracellular fluid. Sex did not affect TBW, but males had greater WF. Birds that ate Cd diets, especially the higher Cd diet, exhibited renal tubular damage and lower GFR. Both hypotheses were accepted since birds that ate Cd had lower GFR and delayed onset of SGS in response to salt challenge and the effects of Cd on the size and response of excretory organs were sexually disparate. Cadmium appeared to compromise the salt excretion of Pekin ducks. However, they are less salt tolerant than seaducks and, in this study, had higher organ Cd concentrations than those seen in wild ducks. Further studies are needed to determine if these observations also apply to the more salt-tolerant seaducks.

Effect of diatomaceous earth on parasite load, egg production, and egg quality of free-range organic laying hens.

The effectiveness of diatomaceous earth (DE) as a treatment against parasites and to increase feed efficiency and egg production of organically raised free-range layer hens was evaluated in 2 breeds of commercial egg layers [Bovan Brown (BB) and Lowmann Brown (LB)] that differ in their resistance to internal parasitic infections. Half the hens of each breed were fed diets supplemented with DE (2%). Their internal parasite loads were assessed by biweekly fecal egg counts (FEC) and by postmortem examination of the gastrointestinal tract. Supplementing DE in diets of LB hens, the more parasite-resistant breed, did not significantly affect their FEC and adult parasite load. However, BB hens treated with dietary DE had significantly lower Capillaria FEC, slightly lower Eimeria FEC, fewer birds infected with Heterakis, and significantly lower Heterakis worm burden than control BB hens. Both BB and LB hens fed the diet containing DE were significantly heavier, laid more eggs, and consumed more feed than hens fed the control diet, but feed efficiency did not differ between the 2 dietary treatments. Additionally, BB hens consuming the DE diet laid larger eggs containing more albumen and yolk than hens consuming the control diet. In a subsequent experiment, the effectiveness of DE to treat a Northern fowl mite (Ornithonyssus sylviarum) infestation was tested. Relative to controls, both breeds of hens that were dusted with DE had reduced number of mites. The results of this study indicate the DE has the potential to be an effective treatment to help control parasites and improve production of organically raised, free-range layer hens.

Hematological and serum biochemical profile of farm emus (Dromaius novaehollandiae) at the onset of their breeding season

Blood profiling is a helpful tool in detecting the health status, metabolic diseases, nutritional deficiencies, and welfare of animals. Body weights, body temperatures, hematological and serum biochemical parameters, enzymes, and electrolytes in both sexes of farm emus at the beginning of their breeding season (November in Canada), were determined. The reference interval for each analyte was also calculated. Emus have lower body temperatures (37.2 ± 0.2) than other poultry species. There was no significant between-sex difference in BW, body temperature, and all the hematological and enzyme parameters measured. However, females had significantly (P < 0.001) higher serum calcium, phosphorus, albumin, total protein, globulin, and triglyceride levels than males, probably in preparation for egg laying. We also examined our findings in light of their sex-role reversal in incubation and brooding. Contrary to other avian species in which only females incubate and brood, there was no sex difference in the hematological and enzyme parameters measured in emus. We found that emus are similar to other ratite species with respect to the changes in protein, globulin, triglyceride, and calcium levels. The findings from our study contribute to the database for reference emu hematological and serum enzyme, metabolite, and electrolyte values.

INTERACTION OF OSMOTIC AND VOLEMIC COMPONENTS IN INITIATING SALT-GLAND SECRETION IN PEKIN DUCKS

Pekin Ducks (Anas platyrhynchos) were infused with 1,000 mM NaCl. Changes in plasma osmolality (Osmpl); relative plasma volume (RPV); and plasma concentrations of electrolytes, arginine vasotocin, and angiotensin II were determined at initiation of salt-gland secretion and at 5 and 11 h post-infusion. Changes in these parameters and in volume and concentration of secretion produced were not different between the sexes. Although Osmpl and RPV were not related prior to infusion, they were linearly related at the onset of secretion (r2 = 0.51, P = 0.013). Osmpl and RPV were equally effective stimuli for secretion. Initial RPV was a strong determinant of secretion volume (P < 0.01) and total sodium (P < 0.001) and potassium (P < 0.001) secreted, but initial Osmpl was not. Plasma arginine vasotocin concen- tration was increased by NaCl infusion but not by subsequent dehydration. Plasma angioten- sin II concentration was not significantly affected by NaCl infusion or by dehydration. These experiments are the first to directly relate the secretory response of the salt glands to the physi- ological state of ducks prior to salt loading and to describe the interactive simultaneous changes

Effects of cadmium ingestion on plasma and osmoregulatory hormone concentrations in male and female Pekin ducks.

Effects of ingested cadmium (Cd) on body mass and plasma, urine, salt-gland secretion, and osmoregulatory hormone concentrations were assessed in male and female Pekin ducks, Anas platyrhynchos, acclimated to 450 mM NaCl over 6 wk and then held an additional 13 wk on 300 mM NaCl (prolonged saline exposure). Groups of six birds ate diets containing 0 (control), 50 (low-Cd diet), or 300 (high Cd diet) w g Cd/g food. Ducks that ingested Cd, especially females, lost body mass. Cadmium ingestion did not affect salt-gland secretion concentration. Control males had higher plasma osmolality and lower relative plasma volume. These increased in both sexes during saline acclimation. The high-Cd diet suppressed the increase in plasma osmolality in both sexes, but a rise in relative plasma volume occurred only in females. Following prolonged exposure to saline, plasma osmolality and relative plasma volume were reduced in control ducks but further elevated in ducks on the high-Cd diet. Cadmium ingestion suppressed the increase in urine osmolality that occurred in control ducks during saline acclimation. Neither saline acclimation nor Cd ingestion affected plasma concentrations of arginine vasotocin or prolactin. Arginine vasotocin was not correlated with plasma osmolality; prolactin was negatively correlated with plasma osmolality, but only in males. Cadmium suppressed the increase in angiotensin II that occurred at higher salinities in control and low-Cd males. This study examined the effects of gradually increasing body cadmium content on osmotic homeos-tasis. Cadmium affected plasma and urine, but not salt-gland secretion, concentrations and some of these responses were sexually disparate. Cadmium did not affect osmoregulatory hormones (arginine vasotocin and prolactin) by which observed changes in plasma concentration might have been influenced.

Suspected Sodium Toxicity in Hand-Reared Great Blue Heron (Ardea herodias) Chicks

Sodium toxicity was suspected in hand-reared great blue heron (Ardea herodias) chicks fed herring frozen in brine (seawater). Affected chicks were lethargic with stiff legs that extended to the posterior, and breathing was labored. Chicks regurgitated food or refused to eat. All chicks that were fed herring exclusively and eight of the 10 chicks fed a mixed diet (herring and salmonids) died, whereas all chicks fed only salmonids survived. Renal lesions ranged from mild to marked generalized nephrosis, which was characterized by degeneration and necrosis of the proximal convoluted tubular epithelium and dilation of the distal convoluted tubules and collecting ducts. These observations suggest that fish frozen in brine is unsuitable food for hand-rearing of young herons.

The effect of diet and host genotype on ceca microbiota of Japanese quail fed a cholesterol enriched diet

Two Japanese quail strains, respectively atherosclerosis-susceptible (SUS) and –resistant (RES), have been shown to be good models to study cholesterol metabolism and transportation associated with atherosclerosis. Our objective was to examine possible difference in cecal microbiota between these strains when fed a control diet and a cholesterol enriched diet, to determine how host genotype and diet could affect the cecal microbiome that may play a part in cholesterol metabolism. A factorial study with both strains and two diets (control, cholesterol) was carried out. Cecal content was collected from 12 week old quail that have been on their respective diets for 6 weeks. DNA was extracted from the samples and the variable region 3–5 of the bacterial 16S rRNA gene was amplified. The amplicon libraries were subjected to pyrosequencing. Principal Component Analysis (PCA) of β-diversity showed four distinct microbiota communities that can be assigned to the 4 treatment groups (RES/control, RES/cholesterol, SUS/control, SUS/cholesterol). At the Phylum level, the 4 treatment groups has distinct Firmicutes community characteristics but no significant difference in Bacteroidetes. Eubacterium dolichum was rare in RES/control but became overabundant in RES/cholesterol. An unclassified species of Lactobacillaceae was found in abundance in SUS/control but the same species was rare in RES/cholesterol. On the other hand, two Lactobacillus species were only found in RES/control and an unclassified Lachnospiraceae species was abundant in RES/cholesterol but rare in SUS/control. The abundance of four species of Lachnospiraceae, three species of Ruminococcaceae and one species of Coprobacillaceae was positively correlated with plasma Total Cholesterol, plasma LDL, and LDL/HDL ratio. Our study of cecal microbiota in these quail has demonstrated that selection for susceptibility/resistance to diet induced atherosclerosis has also affected the quails cecal environment to host distinctly different cecal microbiome.
