Soil organic matter mobilization by root exudates

In order to study the different soil organic matter mobilisation by agrarian (Zea mais: cultivars Paolo and Sandek) and forest (Picea abies Karst. and Pinus sylvestris L.) root exudates, three different soils (Dystric Spodic Cambisol--S1, Haplic Luvisol--S2 and Calcaric Cambisol--S3) have been considered. Treating the soils with water (control) or plant root exudates, soil organic matter extracts were obtained. The extracts were characterised by hormone-like activities and gas chromatographic/mass spectrometric (GC/MS) measurements. Water extract and plant root exudates exhibited no hormone-like activity, while the other soil-extracts were endowed with a different hormone-like behaviour. GC/MS data indicated that in the acid soils (S1) Sandek and Picea abies exudates showed a greater ability in extracting organic acid isomers (Cl4COOH, Cl5COOH and Cl7COOH), while in neutral soils (S3) all the exudates were active in separating organic acids. In intermediate conditions (S2), Picea abies and Pinus sylvestris exudates liberated C15COOH isomers, Paolo C11COOH isomers, while Sandek was not effective. The different role of plant root exudates in mobilising bio-molecules from the bulk of the soil is proposed.

Biological Activity of soil organic matter mobilized by root exudates

In order to study the biological activity of soil organic matter mobilized by agrarian (Zea mays: cultivars Mytos and Samantha) and forest (Picea abies Karst. and Pinus sylvestris L.) root exudates, two different soils, an Eutric Cambisol (EC) and a Rendzic Leptosol (RL), were considered. Soil organic matter extracts were obtained by treating the soils with water (control) or plant root exudates. The extracts were characterized by hormone-like activities and gas chromatographic/mass spectrometric (GC/MS) measurements. Their effects on the nitrogen metabolism in maize seedlings were evaluated. The nitrogen organification in the maize seedlings has been greatly stimulated by all the organic acid extracts from the agrarian soil, while the extracts from the forest soil had no influence upon the metabolism; this indicated a probable link between the plant and the environment. The different biological activities of the extracts are discussed.

Soil humic compounds and microbial communities in six spruce forests as function of parent material, slope aspect and stand age

The influences on soil chemical and microbial properties of parent material, north south aspect and time measured as stand age were investigated in six spruce (Picea abies (L.) Karst.) forests located in the alpine range of Northern Italy. Soil samples from A horizons were analysed for humic substances and in parallel Amplified Ribosomal DNA Restriction Analysis (ARDRA) community profiles and microbial biomass carbon and nitrogen content were determined. Chemical data were analyzed by canonical discriminant analysis while the ARDRA fingerprints were ordered in clusters using image analysis software. The geologic parent material was the most determining factor and the aspect-dependent microclimate features also played a distinct role in defining both soil chemistry and microbial community composition; in contrast the composition of the deeper humus layers (OH, A) was stable and similar within a spruce forest cycle time. Most important variables in the construction of the discriminating models resulted soil pH, Dissolved Organic Carbon content and Dissolved Organic Matter phenolic compounds. Bacterial communities appeared to be shaped first and foremost by the substratum, secondly by mountain slope orientation, and thirdly by forest stage, thus confirming the CDA model.

Plant biostimulants: physiological responses induced by protein hydrolyzed-based products and humic substances in plant metabolism

In recent years, the use of biostimulants in sustainable agriculture has been growing. Biostimulants can be obtained from different organic materials and include humic substances (HS), complex organic materials, beneficial chemical elements, peptides and amino acids, inorganic salts, seaweed extracts, chitin and chitosan derivatives, antitranspirants, amino acids and other N-containing substances. The application of biostimulants to plants leads to higher content of nutrients in their tissue and positive metabolic changes. For these reasons, the development of new biostimulants has become a focus of scientific interest. Among their different functions, biostimulants influence plant growth and nitrogen metabolism, especially because of their content in hormones and other signalling molecules. A significant increase in root hair length and density is often observed in plants treated with biostimulants, suggesting that these substances induce a “nutrient acquisition response” that favors nutrient uptake in plants via an increase in the absorptive surface area. Furthermore, biostimulants positively influence the activity and gene expression of enzymes functioning in the primary and secondary plant metabolism. This article reviews the current literature on two main classes of biostimulants: humic substances and protein-based biostimulants. The characteristic of these biostimulants and their effects on plants are thoroughly described.

Effect of a Compost and Its Water-Soluble Fractions on Key Enzymes of Nitrogen Metabolism in Maize Seedlings

The growing concern on long-term productivity of agroecosystems has emphasized the need to develop management strategies to maintain and protect soil resources, particularly soil organic matter (SOM). Among these, the composting process allows both recycling of the increasing amount of organic waste materials and restoration of the content of organic matter in soil. A sequential chemical fractionation into structurally unbound (SU), weakly bound (WB) and strongly bound (SB) compounds was applied to a bulk compost, and its soluble fractions were extracted in water, either after oxidation of compost suspension with an oxygen flux (TEA), or without oxidation but separated into hydrophilic (HiDOM) and hydrophobic (HoDOM) components. The ratio of hydrophilic over hydrophobic compounds decreased in the order HiDOM > TEA > compost > HoDOM, while TEA and compost showed the largest content of SU and WB components, respectively. Such chemically characterized bulk compost and fractions were tested on maize seedlings grown in sand and in hydroponic conditions, and the effects on plant growth and nitrogen metabolism were measured. The structurally complex bulk compost and the hydrophobic HoDOM fraction negatively affected plant growth, whereas the hydrophilic and less-structured fractions (HiDOM and TEA) showed large positive effects on both growth and enzymatic activities of plants. These results suggest that composted organic matter can become useful to stimulate plant growth if the content of potentially bioavailable hydrophilic and poorly structured components is large. These components may be progressively separated from the compost matrix and contribute to the dynamics of natural organic matter in soil.

Capsicum chinensis L. growth and nutraceutical properties are enhanced by biostimulants in a long-term period: chemical and metabolomic approaches

Two biostimulants, one derived from alfalfa plants (AH) and the other obtained from red grape (RG), were chemically characterized using enzyme linked immuno-sorbent assays, Fourier transform infrared (FT-IR) and Raman spectroscopies. Two doses (50 and 100 mL L−1 for RG, and 25 and 50 mL L−1 for AH) of biostimulants were applied to Capsicum chinensis L. plants cultivated in pots inside a tunnel. The experimental design consisted of the factorial combination of treatment (no biostimulant, plus AH, plus RG) at three doses (zero, low, and high) and two time-course applications (at the second and fourth week after transplantation) and the effects were recorded at flowering and maturity. Both biostimulants contained different amounts of indoleacetic acid and isopentenyladenosine; the AH spectra exhibited amino acid functional groups in the peptidic structure, while the RG spectra showed the presence of polyphenols, such as resveratrol. These results revealed that at flowering, RG and AH increased the weights of fresh leaves and fruits and the number of green fruits, whereas at maturity, the biostimulants most affected the fresh weight and number of red fruits. At flowering, the leaves of the treated plants contained high amounts of epicatechin, ascorbic acid, quercetin, and dihydrocapsaicin. At maturity, the leaves of the treated plants exhibited elevated amounts of fructose, glucose, chlorogenic, and ferulic acids. Moreover, green fruits exhibited a high content of chlorogenic acid, p-hydroxybenzoic acid, p-coumaric acid and antioxidant activity, while both AH- and RG-treated red fruits were highly endowed in capsaicin. The 1H high-resolution magic-angle spinning (HRMAS)-nuclear magnetic resonance (NMR) spectra of red fruits revealed that both products induced a high amount of NADP+, whereas RG also increased glucose, fumarate, ascorbate, thymidine and high molecular weight species. Our results suggested that AH and RG promoted plant growth and the production of secondary metabolites, such as phenols.

BIOLOGICAL ACTIVITY OF HUMIC SUBSTANCES EXTRACTED FROM SOILS UNDER DIFFERENT VEGETATION COVER

Abstract Comparisons were made between the chemical compositions of humic substances extracted from three soils covered by different vegetation and their biological activities assayed using 15‐ and 30‐day‐old seedlings of Pinus sylvestris and Picea abies. The growth, “α‐amylase and invertase activity were affected by humic fractions and by gibberellic acid (GA), indicating that humic matter had a gibberellin‐like activity. The isoenzymatic polymorphism in the electrophoretic patterns of esterase was influenced in a similar way by all humic fractions and by indoleacetic acid (IAA), which also suggested that the humic fractions exhibited an auxin‐like activity. The humic fractions extracted from the grassland, exhibiting higher amounts of phenolic and a considerable amount of carboxyl carbon, showed the best metabolic effect. Low-molecular-weight organic acids and hormone-like activity of dissolved organic matter in two forest soils in N Italy

Concentrations of aliphatic acids, phenolic acid, and inorganic nutrients, as well as hormone-like activity, were determined in soil dissolved organic matter (DOM) from two forest sites in N Italy showing differing degrees of silver fir regeneration. In the site where silver fir recruitment was abundant, humification processes prevailed, and the soil DOM had a high content in aliphatic and phenolic acids. This enhanced the hormone-like activity in the soil, which could in turn promote growth of silver fir seedlings. In the site with poor fir recruitment, the soil DOM underwent rapid mineralization and was richer in inorganic nutrients, but had lower concentrations of aliphatic and phenolic acids, and lower hormone-like activity.

Biological Activity of Vegetal Extracts Containing Phenols on Plant Metabolism.

The influence of vegetal extracts derived from red grape, blueberry fruits and hawthorn leaves on Zea mays L. plant growth and the activity of phenylalanine ammonia-lyase (PAL), a key enzyme of the phenylpropanoid pathway, was investigated in laboratory experiments. The extracts were characterized using FT-IR and Raman spectroscopies in order to obtain a pattern of the main functional groups. In addition, phenols content was determined by HPLC, whereas the content of indoleacetic acid and isopentenyladenosine hormones was determined by ELISA test and the auxin and gibberellin-like activities by plant-bioassays. The treated maize revealed increased root and leaf biomass, chlorophyll and sugars content with respect to untreated plants. Hawthorn, red grape skin and blueberry at 1.0 mL/L induced high p-coumaric content values, whilst hawthorn also showed high amounts of gallic and p-hydroxybenzoic acids. PAL activity induced by hawthorn at 1.0 mL/L had the highest values (11.1-fold UNT) and was strongly and linearly related with the sum of leaf phenols. Our results suggest that these vegetal extracts contain more than one group of plant-promoting substances.

Snow vole (Chionomys nivalis Martins) affects the redistribution of soil organic matter and hormone‐like activity in the alpine ecosystem: ecological implications

Abstract In alpine environments, colonies of snow vole (Chionomys nivalis Martins) cause strong pedoturbation, which may affect humification process and soil organic matter (SOM) cycling, with repercussions on the hormone‐like activity of organics. We investigated the effect of snow vole pedoturbation on the chemical and spectroscopic features of soil organic fractions, and the potential hormone‐like activity of humic and fulvic acids (HA, FA). The study site was located on the high‐mountain environment of the Majella massif (central Italy). Pedoturbated and regular soils were morphologically described and characterized for pH and content of total organic carbon, total extractable carbon, HA, and FA. Both HA and FA were extracted and investigated using attenuated total reflectance/Fourier transform infrared (ATR/FTIR), nuclear magnetic resonance with high‐resolution magic angle spinning (HRMAS‐NMR), and 1H‐13C heteronuclear single quantum coherence (HSQC). HA and FA were also tested for their auxin‐like and gibberellin‐like activities. Results provide evidences that bioturbated and regular soils contain a poorly decomposed SOM, but HA and FA with a well‐defined molecular structure. The HA and FA from both bioturbated and regular soils show a hormone‐like activity with a different allocation along the soil profile. In the regular soil, the highest auxin‐like activity was shown by HA and FA from Oe1 horizon, while gibberellin‐like activity was expressed by FA from Oe2 horizon. Burrowing activity determines a redistribution of organics throughout the profile with a relatively high auxin‐like activity in the FA from straw tunnel wall (STW) and gibberellin‐like activity in the HA from vole feces (VF). The relative high presence of carboxylic acids, amides, proteins, and amino acids in the FA from STW and the aromatic moieties in the HA from VF put evidences for their different behavior. The fact that snow vole activity has modified the chemical and biological properties of SOM in these soils otherwise considered governed only by low temperature has important ecological implications such as the preservation of soil fertility and vegetal biodiversity.
