Oxidative Conversion of α,α-Disubstituted Acetamides to Corresponding One-Carbon-Shorter Ketones Using Hypervalent Iodine (λ5) Reagents in Combination with Tetraethylammonium Bromide

Eknath V. Bellale; Dinesh S. Bhalerao; Krishnacharya G. Akamanchi

Alpha,alpha-disubstituted acetamides undergo oxidative dehomologation to give one-carbon-shorter ketones when reacted with a hypervalent iodine (lambda(5)) reagent in combination with tetraethylammonium bromide (TEAB) in various solvents. In further studies, one such combination of a hypervalent iodine (lambda(5)) reagent, o-iodoxybenzoic acid, and TEAB has been established as a new, mild, efficient, and general method for the transformation.

MILD AND EFFICIENT METHOD FOR α-THIOCYANATION OF KETONES AND β-DICARBONYL COMPOUNDS USING BROMODIMETHYLSULFONIUM BROMIDE-AMMONIUM THIOCYANATE

Dinesh S. Bhalerao; Krishnacharya G. Akamanchi

An efficient and convenient method for α-thiocyanation of ketones and β-dicarbonyl compounds has been developed using a reagent combination of bromodimethylsulfonium bromide (BDMS) and ammonium thiocyanate in acetonitrile. The developed method is mild and gave good yield of the products at room temperature.

A novel approach for the synthesis of omeprazole, a potent antiulcer drug, is described. The synthetic procedure involved the formation of an ester of the 5-methoxy thiobenzimidazole followed by coupling of the ester with the Grignard reagent of 2-chloromethyl-4-methoxy-3,5-dimethyl-pyridine.
