Application of a fish health assessment index and associated parasite index to Clarias gariepinus (Teleostei: Clariidae) in the Vaal River system, South Africa

The aim of this study was to determine if the fish health assessment index (HAI) developed in the USA and associated parasite index (PI), when applied to Clarias gariepinus at two localities in the upper and middle Vaal River system, could distinguish between localities on the grounds of water quality. Elevated HAI values, correlating with poorer chemical and physical water quality, were recorded from the Vaal River Barrage site. Parasite data were congruent with the main PI premise that prevalence and intensity of endoparasite infection would be higher at the more polluted locality, with the converse being recorded for ectoparasites. It was therefore possible to distinguish between localities with different water quality, based on parasites present on the hosts.

Prophylactic treatment of flea-infested cats with an imidacloprid/flumethrin collar to forestall infection with Dipylidium caninum

BackgroundThe objective of the study was to determine the sustained effectiveness of 10% imidacloprid (w/w) and 4.5% flumethrin (w/w) incorporated in a slow-release matrix collar in preventing Dipylidium caninum infection in cats following repeated laboratory-infestations with fleas infected with metacestodes.MethodsEfficacy against infection with D. caninum was evaluated by infesting 16 cats with the flea Ctenocephalides felis felis infected with metacestodes of the tapeworm. Medicated collars were fitted to 8 of the cats and infestation of each cat with 200 fleas from a suitably infected batch commenced 7 days later and continued at weekly intervals until Day 28. Efficacy against fleas was evaluated 24 h after each infestation. Infection of the cats with D. caninum was verified by daily examination of the cats’ faeces and immediate surroundings for proglottids from Day 21 to Day 60. Calculation of the prophylactic effectiveness of the collars in preventing infection of the cats with D. caninum was based on the difference in the geometric mean number of scoleces recovered from the gastrointestinal tracts of collared compared to untreated cats at necropsy on Day 61.ResultsEfficacy of the collars against infestation of the cats with fleas was 99.9% on Day 7 and 100% at each subsequent weekly assessment. Infection of the fleas with metacestodes was ≥40% in 7 to 13 day old fleas, but progressively decreased thereafter. At necropsy all the control cats were infected with D. caninum and harboured between 19 and 346 scoleces with a geometric mean of 58.3. A single treated cat was infected and harboured 2 scoleces. Effective prevention of infection with D. caninum, based on a comparison of the geometric mean numbers of scoleces recovered from control and treated cats, was 99.7%.ConclusionThe insecticidal components of the medicated collars are capable of rapidly eliminating newly-acquired infestations of fleas that are infected with the metacestodes of D. caninum, thus preventing infection with the cestode in collared cats.

The Efficacy of a Topically Applied Imidacloprid 10 % / Moxidectin 2.5 % Formulation (Advocate®, Advantage® Multi, Bayer) against Immature and Adult Spirocerca lupi Worms in Experimentally Infected Dogs

This study investigated the efficacy and safety of an imidacloprid 10 %/moxidectin 2.5 % spot-on combination (Advocate®, Advantage® Multi, Bayer) against immature and mature stages of Spirocerca lupi in experimentally infected dogs. 24 dogs were allocated to 3 groups and infected with approximately 10 L3 larvae of S. lupi orally on study day (SD) +2, +14, +28 and +42. Group 1 remained as untreated control group. Group 2 dogs were treated on SD –28, 0, and thereafter monthly until Day 280 (12 treatments). Group 3 dogs were treated weekly on 19 occasions starting on SD +170. The dosage for all treatments was the licensed dose of 10–25 mg imidacloprid/2.5–6.25 mg moxidectin per kg body weight. All dogs were examined on SD +169 or +176 by endoscopy. Group 3 dogs were additionally examined approximately every two weeks up to Day 296. On Day +308 or +310, all dogs were necropsied to recover S. lupi worms and to quantify lesions in the thoracic aorta and oesophagus. Dogs in the control group were adequately infected with S. lupi, demonstrated by the extensive damage to the thoracic aorta, the nodules in the oesophagus and the large numbers of worms recovered. In total 144 worms were collected (geometric mean of 16.8 worms per dog). Dogs in group 2 had no or very slight damage to the thoracic aorta and no nodules or worms in the oesophagus, indicating 100 % efficacy of the monthly treatments. Dogs in group 3 were also adequately infected, showing nodules in the oesophagus before initiation of weekly treatment, and at necropsy extensive damage was seen in the thoracic aorta. After treatment, three dogs of 8 still had a few nodules and in total three worms (GM of 0.25 per dog) were recovered, demonstrating an efficacy of 98.5 % against adult S. lupi. All dogs tolerated the treatment well and no treatment- related adverse events occurred.

Efficacy of afoxolaner plus milbemycin oxime chewable tablets against naturally acquired intestinal nematodes in dogs

The efficacy of oral afoxolaner plus milbemycin oxime combination chewable tablets (NexGard Spectra, Merial) against naturally acquired intestinal nematode infections in dogs was evaluated in six negative control, blinded studies including a total of 114 dogs. Dogs were selected based on a pre-treatment fecal examination indicating patent infections with hookworms (two studies), Toxocara or Toxascaris ascarids (one study each) or Trichuris whipworms (two studies). In each study, dogs were assigned to blocks of two animals each, based on decreasing pre-treatment body weight and were randomly allocated to one of two groups consisting of eight, nine or 10 dogs: untreated (control) or treated with the combination chewable tablet formulation. Chewable tablets were combined to provide doses of actives as close as possible to the minimum effective dose of afoxolaner and milbemycin oxime, i.e., 2.5 mg/kg body weight and 0.5 mg/kg body weight, respectively, once on Day 0. For parasite recovery and count, dogs were euthanized humanely and necropsied seven or eight days after treatment. A single treatment with afoxolaner plus milbemycin oxime chewable tablets provided 94.8% and 90.9% efficacy against adult Ancylostoma braziliense and A. caninum, respectively, 97.8% and 99.4% efficacy against adult Toxocara canis and Toxascaris leonina, respectively, and ≥98.3% efficacy against adult Trichuris vulpis. Compared to untreated controls, nematode counts of the treated dogs were significantly reduced (F-test; p<0.002). In addition, analysis of the pooled data across studies revealed that treatment with afoxolaner plus milbemycin oxime chewable tablets reduced adult Uncinaria stenocephala burdens by 74.9% (p=0.002). All dogs tolerated the treatment well based on clinical observations post-treatment and daily clinical observations. No adverse experiences or other clinical problems related to the treatment were observed throughout the studies. The results of this series of controlled studies demonstrated high efficacy and excellent acceptability and safety of the afoxolaner plus milbemycin oxime chewable tablets when administered for treatment of a broad range of canine intestinal nematode infections.

Monogenean parasites from fishes of the Vaal Dam, Gauteng Province, South Africa. I. Winter survey versus summer survey comparison from Labeo capensis (Smith, 1841) and Labeo umbratus (Smith, 1841) hosts.

Indigenous South African Labeo spp. show promise with regard to development of semi-intensive aquaculture, yet little research on their monogenean fauna has been conducted. Ecological aspects of monogenean fauna of the moggel Labeoumbratus (Smith 1841) and the Orange River mudfish Labeo capensis (Smith 1841), as recorded during both winter and summer sampling surveys, are reported here. Fish were collected using gill nets, euthanized and gills removed and examined to both quantify parasite numbers and distribution on the gills. Results obtained support the hypothesis that gill site preference is not due to active choice for a particular attachment site, but rather a result of water flow over gills during respiration in conjunction with fish behaviour and habitat use. Interaction between individual elements investigated (temperature effects, parasite population dynamics and host population dynamics) may be largely responsible for seasonal differences in infection statistics of monogenean parasites. Such interactions should be investigated in future large scale ecological studies, in combination with experimental studies, to further elucidate these effects.

Efficacy of oral afoxolaner plus milbemycin oxime chewables against induced gastrointestinal nematode infections in dogs.

The efficacy of oral afoxolaner plus milbemycin oxime combination chewables against induced gastrointestinal nematode infections in dogs was evaluated in six separate studies. Two studies were performed to evaluate the efficacy of the product against Toxocara canis, two studies evaluated the efficacy against Toxascaris leonina, one study evaluated the efficacy against Ancylostoma braziliense, and one study evaluated the efficacy against Ancylostoma caninum. In the A. caninum study, the efficacy of milbemycin oxime alone and afoxolaner alone was also evaluated. Dogs in all studies were inoculated with infective eggs or larvae and confirmed to have patent infections based on a fecal examination prior to allocation to study group and treatment. Each study utilized a randomized block design with blocks based on pre-treatment body weight. All dogs were assigned to blocks based on body weight, and then each dog within a block was randomly assigned to treatment group. There were two groups of 10 dogs each in the T. canis, T. leonina, and A. braziliense studies: 1) an untreated (control) group and 2) a group treated with afoxolaner plus milbemycin oxime chewables (NexGard Spectra(®), Merial). This group was treated at a dose as close as possible to the minimum effective dose of afoxolaner and milbemycin oxime (2.5mg+0.5mg per kg body weight, respectively) once on Day 0 using whole chews. There were four groups of 10 dogs each in the A. caninum study: 1) untreated (control), 2) NexGard Spectra(®) as described above, 3) milbemycin oxime alone (dose of at least 0.5mg per kg of body weight) and 4) afoxalaner alone (dose of at least 2.5mg per kg body weight). For parasite recovery and counts, dogs were euthanized humanely and necropsied seven days after treatment. The efficacy of the afoxolaner plus milbemycin oxime combination was ≥98% against T. canis, ≥95.8% against T. leonina, and 90.2% against A. braziliense. Efficacy of the combination against A. caninum was 99.7%, while the efficacy of milbemycin oxime alone was 99.6% and the efficacy of afoxolaner alone was 2.1%. Dogs treated with afoxolaner plus milbemycin oxime chewables had significantly (p≤0.0002) fewer nematodes than the untreated controls in all studies. There were no adverse events or other health problems that were related to treatment with Nexgard Spectra(®) in these studies. The results of these controlled studies demonstrate the high efficacy of the afoxolaner plus milbemycin oxime chewables against a broad range of canine intestinal nematode infections.

Prophylactic treatment of flea-infested dogs with an Imidacloprid/Flumethrin collar (Seresto®, Bayer) to preempt infection with Dipylidium caninum

The objective of the study was to determine the sustained effectiveness of 10 % imidacloprid and 4.5 % flumethrin, incorporated in a slow-release matrix collar, in preventing Dipylidium caninum infection in dogs after repeated laboratory infestations with fleas infected with metacestodes of this tapeworm. Efficacy against infection with D. caninum was evaluated by infesting 16 dogs with cat fleas (Ctenocephalides felis) on study days 7, 14, 21, 28, 35 and 42, from batches suitably infected with D. caninum metacestodes. Prior to each post-treatment infestation the D. caninum infection rate for the fleas was determined by microscopically examining 100 fleas for D. caninum metacestodes. The D. caninum prevalence in the fleas used for infestations ranged from 23 % to 52 %. Medicated collars were fitted to 8 of the dogs on study day 0. The weight of the IVP collars varied between 35.48 g and 38.48 g (average 37.16 g), whilst animal weight varied between 12.20 kg and 17.98 kg (treated group, n = 8, average 14.79 kg). Seven days later infestation of each of the 16 dogs with 250 fleas commenced. Infestations continued at weekly intervals until Day 42 with efficacy against fleas evaluated 24 hours after each infestation. From Days 21 to 74, infection of the dogs with D. caninum was verified (daily examination of faeces and cages for the presence of expelled proglottids). Calculation of prophylactic effectiveness of the collars in preventing infection with D. caninum was based on the difference in geometric mean numbers of scoleces between groups at necropsy on Day 75. Effective prevention of infection with D. caninum was found to be 96.6 %. Efficacy of the collars against fleas was ≥ 99.9 % for the duration of the assessment period. Newly acquired infestations of fleas are rapidly eliminated by the insecticidal components of the medicated collars over a period of several months. In the event of fleas being infected with metacestodes, with D. caninum can be prevented in collared dogs, concurrently reducing the likelihood of transmission to humans.

Evaluation of the fish health assessment index in the Olifants River system, South Africa

The fish health assessment index (HAI) biomonitoring technique, developed in the USA and subsequently tested in South Africa, was re-evaluated in the Olifants River system using Clarias gariepinus, Oreochromis mossambicus and Labeobarbus marequensis during drought and flood conditions in 1994–1997 at the same sample sites in the Kruger National Park as used previously, as well as at Loskop Dam and Bronkhorstspruit Dam in the upper catchment. Parasite numbers recorded from Clarias gariepinus supported the HAI hypothesis that higher endoparasite and lower ectoparasite numbers occur at highly impacted areas. The apparent effects of pollution type on the ratio between ectoparasites and endoparasites are discussed. Environmental stressors such as flood conditions resulted in immunological responses, reflected statistically as changes in white blood cell percentage (WBC%). It is suggested that endoparasite and WBC% data provide the best overall assessment of fish condition in the Olifants River system. Modifications made to the HAI included further development of the original parasite presence/absence component into a more extensive parasite index component. The modified HAI index shows promise as a long-term biological monitoring tool in South Africa.

Monogenean Parasite Species Descriptions from Labeo spp. Hosts in the Vaal Dam, South Africa

Specimens of Labeo capensis (n = 13) and Labeo umbratus (n = 26) from the Vaal Dam (South Africa) were collected and examined for gill and skin monogenean parasites. Three new Dactylogyrus and one new Dogielius species are described. Dactylogyrus iwani n.sp. (longer inner root on anchor and predominates onL. capensis) and D. larindae n.sp. (shorter inner root on anchor and predominates on L. umbratus) show similarities in male copulatory organ (MCO) and anchor structure with representatives of the D. pseudanchoratus group. Dactylogyrus nicolettae n.sp. differs completely with regard to anchor morphology (stout with a curved inner root and reduced outer root), yet MCO morphology (curved penis and simple accessory piece structure of similar shape and size) are similar in all three species described. Dogielius intorquens n.sp. was collected from both hosts. Two forms, corresponding to host species and differing in anchor and MCO size but not shape, are described. This brings the number of Dactylogyrus species described from African Labeo spp. to 29, with D. intorquens n.sp. constituting the 16th Dogielius species from African Labeo spp. hosts.

The efficacy of a generic doxycycline tablet in the treatment of canine monocytic ehrlichiosis

The objective of the present study was to evaluate the therapeutic efficacy of a generic doxycycline tablet (DoxyVet®) against Ehrlichia canis infection in dogs. Canine monocytic ehrlichiosis is caused by the bacterium E. canis and transmitted by the brown kennel tick (Rhipicephalus sanguineus). Six disease-free and tick-free dogs were infested with E. canis-infected ticks. Once diagnosed (with polymerase chain reaction [PCR] analysis and platelet counts) as positive for infection, doxycycline tablets were administered orally once a day for 20 consecutive days, at a target dose level of 10 mg/kg. The actual dose administered was calculated as ranging between 10 mg/kg and 11.7 mg/kg. The PCR analysis, 28 days after the first administration of the tablets, failed to detect E. canis in any of the dogs. On Day 56 of the study, four of the dogs were diagnosed with E. canis for the second time and a fifth dog was diagnosed on Day 70. The platelet counts of the sixth dog remained within normal levels and it was discharged from the study on Day 84. Doxycycline tablets were then administered to the remaining five infected dogs for 28 consecutive days. Four of these dogs had no positive PCR results during the following 3 months. The fifth dog was diagnosed with E. canis for the third time 58 days after the last tablets of the second treatment had been administered, after which it was rescue treated (doxycycline for a further 28 days). The results indicate that doxycycline administered in tablet form (DoxyVet®) at 10 mg/kg – 11.7 mg/kg body mass once daily for 28 consecutive days clears most dogs of infection. The importance of a concomitant tick-control programme is therefore stressed.
