Within our everyday life, we are confronted with a variety of toxic substances of natural or artificial origin. Toxins are already used, e.g. in medicine, but there is still an increasing number of toxic compounds, representing a tremendous potential to extract new substances. Since predictive toxicology gains in importance, the careful and extensive investigation of known toxins is the basis to assess the properties of unknown substances. In order to achieve this aim, we have collected toxic compounds from literature and web sources in the database SuperToxic. The current version of this database compiles about 60 000 compounds and their structures. These molecules are classified according to their toxicity, based on more than 2 million measurements. The SuperToxic database provides a variety of search options like name, CASRN, molecular weight and measured values of toxicity. With the aid of implemented similarity searches, information about possible biological interactions can be gained. Furthermore, connections to the Protein Data Bank, UniProt and the KEGG database are available, to allow the identification of targets and those pathways, the searched compounds are involved in. Summary In order to contribute to the verification of the hypothesis of Furchgott et al. that the effect of acetylcholine on blood vessels is mediated by factors produced by endothelial cells, the authors tried to provide evidence for the existence of muscarinic receptors on these cells. Using cultivated cells of the endothelium of calve aortas, a specific saturable binding of the potent muscarinic antagonist 3 H-Quinuclidinyl benzilate and therewith the existence of muscarinic receptors on the cells could be demonstrated. Long time cultivation leads to the disappearance of the receptors.

Zusammenfassung Glattmuskulares Myosin wurde unter Anwendung der Methode der indirekten Immunfluo-rescenz in Kryostatschnitten, isolierten Zellen und Zellkulturen der neonatalen Rattenaorta nachgewiesen. Die relativen Myosinmengen von Einzelzellen wurden verglichen, indem die Fluorescenz-intensitat gemessen wurde. Es wurde gezeigt, das gut differenzierte glatte Muskelzellen, die sowohl in Suspensionen aus der gesamten Aorta und aus der Aortenmedia als auch in Aortenzell-kulturen nachgewiesen werden konnten, einen hoheren Myosingehalt besitzen als die Fibroblasten der Adventitia und glatte Muskelzellen mit einem niedrigeren Differenzierungsgrad.

Summary Heparin, sodium butyrate, and the absence of platelet derived factors in the nutrient medium are able to inhibit the decrease of the content of myosin in cultivated arterial smooth muscle cells. Factor VIII and thrombin have no significant influence on this process.

Summary Endothelial cells were able to accomplish at least 120 cell doublings (60 passages) without any decrease of division speed. After that, the doubling time begins to increase. At passage 80, the number of cells was not doubled even after a period of 4 weeks. Temperature stress (41 #x00B0;C for 3 d) leads the cells to normal doubling times again and allows the cultures to reach passage 124. The cells were able to produce factor VIII antigen, but in less quantities, still at passage 124.

Zusammenfassung Der relative mittlere Myosingehalt der Zellen glattmuskularer Organe wurde nach enzymatischer Desintegration der Gewebe immuncytochemisch bestimmt und der prozentuale Anteil von Zellen unterschiedlichen Myosingehaltes am Zellgemisch ermittelt. Als Modellsystem wurden Aorta und Harnblase neonataler Hatten sowie das voll entwickelte Amnion des Huhnereis verwendet. Als Negativkontrolle wurden die von glattmuskularem Myosin freien Zellen einer permanenten Zellinie eingesetzt. Es zeigte sich, das der immuncytochemisch ermittelte relative Myosingehalt mit dem Entwicklungszustand des glattmuskularen Organs gleichsinnig vorlauft.
