Effects of Different Temperatures on Testis Structure and Function, with Emphasis on Somatic Cells, in Sexually Mature Nile Tilapias (Oreochromis niloticus)

Abstract The Nile tilapia (Oreochromis niloticus) is economically one of the most important freshwater fish and is an excellent model for studies under laboratory conditions. Temperature is considered a very important modulator of reproductive activity in fish, although few studies have specifically addressed the effects of this key factor on morphological and functional aspects of teleost testes. Therefore, our main objectives in the present study were to analyze the effects of different temperatures (20, 25, 30, and 35°C) on testicular somatic and germ cells in sexually mature Nile tilapias. Compared with fish kept at other temperatures, tilapias maintained at 20°C demonstrated increased (P < 0.05) Sertoli cell and Leydig cell proliferation, volume density and frequency of most type B spermatogonia, and germ cell apoptosis. Conversely, tubular fluid secretion was decreased (P < 0.05) in the same animals. Although not significant, type A spermatogonia proliferation followed the pattern established for Sertoli cell and Leydig cell mitotic activity, suggesting that they preferentially would proliferate at lower temperatures. Based on most results found in our study and considering that tilapias are nonseasonal breeders, we suggest a model for temperature action on tilapia testes in which lower temperature (20°C) would favor type A spermatogonial renewal, Sertoli cell and Leydig cell proliferation, and germ cell apoptosis, whereas higher temperatures (30–35°C) would trigger rapid germ cell differentiation. Thus, tilapias could potentially be utilized in studies involving hormones and factors related to Sertoli cell and Leydig cell proliferation and spermatogonial self-renewal or differentiation.

Reproductive biology and feeding of Curimatella lepidura (Eigenmann & Eigenmann) (Pisces, Curimatidae) in Juramento reservoir, Minas Gerais, Brazil

Reproductive biology and feeding of Curimatella lepidura (Eigenmann & Eigenmann, 1889) were studied in Juramento reservoir, Sao Francisco River basin, Southeastern Brazil. Histological analyses and gonadosomatic indexes revealed females and males in reproductive activity from October to March and total spawning occurring from January to March coupled with the peak of spermiating males. In the dry season, the fishes accumulated energetic reserves for reproduction during a short rainy season. The species presented sexual dimorphism, being females larger than males and sexual maturation occurring close to 7.7 cm standard length for females and 7.1 cm for males. C. lepidura presented iliophagous feeding habit, ingesting mainly sediment/detritus and a small amount of acari, algae, Tricoptera insects and Ostracoda crustaceans, suggesting a probable role in nutrient recycling of the Juramento reservoir.

Successful xenogeneic germ cell transplantation from Jundia catfish (Rhamdia quelen) into adult Nile tilapia (Oreochromis niloticus) testes

Fish germ cell transplantation presents several important potential applications for aquaculture, including the preservation of germplasm from endangered fish species with high genetic and commercial values. Using this technique in studies developed in our laboratory with adult male Nile tilapias (Oreochromis niloticus), all the necessary procedures were successfully established, allowing the production of functional sperm and healthy progeny approximately 2months after allogeneic transplantation. In the present study, we evaluated the viability of the adult Nile tilapia testis to generate sperm after xenogeneic transplant of germ cells from sexually mature Jundia catfish (Rhamdia quelen) that belong to a different taxonomic order. Therefore, in order to investigate at different time-periods post-transplantation, the presence and development of donor PKH26 labeled catfish germ cells were followed in the tilapia seminiferous tubules. From 7 to 20days post-transplantation, only PKH26 labeled spermatogonia were observed, whereas spermatocytes at different stages of development were found at 70days. Germ cell transplantation success and progression of spermatogenesis were indicated by the presence of labeled PKH26 spermatids and sperm on days 90 and 120 post-transplantation, respectively. Confirming the presence of the catfish genetic material in the tilapia testis, all recipient tilapias evaluated (n=8) showed the genetic markers evaluated. Therefore, we demonstrated for the first time that the adult Nile tilapia testis offers the functional conditions for development of spermatogenesis with sperm production from a fish species belonging to a different order, which provides an important new venue for aquaculture advancement.

Stocking density in the larviculture of Nile tilapia in saline water

The objective of this study was to assess the possibility of increasing the stocking density in the larviculture of Nile tilapia, Oreochromis niloticus, in saline water. Six-day post-hatched larvae (average weight of 0.09±0.002 g), at the start of the exogenous feeding, were stocked in twelve 8 L tanks at densities of 1, 10, 20 and 30 larvae/L. The larvae were fed a diet consisting of 40% crude protein five times per day. During larviculture, water was kept at a salinity of 2 g of common salt/L. At the end of the 28-day experiment, the survival rate, length, weight, temperature, water salinity and conductivity were not affected by the different treatments. The dissolved oxygen and pH, however, decreased as density increased, whereas total ammonia concentration, turbidity and biomass showed a direct relationship with the increase of stocking density. Consequently, the present study shows for the first time that the larviculture of Nile tilapia is possible with up to 30 larvae/L in water with 2 g of salt/L.

Digestibilidade de ingredientes em dietas para juvenis de pacamã (Lophiosilurus alexandri)

The objective of this work was to determine the apparent digestibility coefficient (ADC) of six ingredients, in practical and purified reference diets, and to assess the influence of these reference diets on the digestibility of the ingredients in pacman catfish juveniles. The following ingredients were evaluated: fish meal, meat and bone meal, soybean meal, roasted whole soybean, wheat meal, and corn protenose. There was interaction between the reference diet and the studied ingredients. The purified diet provided higher ADC for crude protein and gross energy. Pacman catfish showed high ADC for protein ingredients of vegetable origin for the two reference diets.

Modelagem das trajetórias médias do peso vivo e do rendimento de carcaça de tilápias do Nilo (Oreochromis niloticus) por meio de funções B-spline

Employing the method of least squares and quadratic B-spline polynomials, different statistical models were tested to identify the most appropriate to model the mean trajectories of live weight and carcass yield of Nile tilapia (Oreochromis niloticus). Data of live weight (8,758) and carcass yield (2,042) of tilapias with ages between 106 and 245 days were obtained from 72 families derived from 36 males and 72 females. The sex and tank variables were considered as classificatory and the coefficients of quadratic polynomials B-spline of two to five intervals of the same size were used as covariables. According to most fit criteria used, the models with quadratic B-spline polynomial with five intervals of the same size presented the best adjustments. The increase in number of intervals of B-spline polynomial improved the fit of the polynomials to the data. The inclusion of classificatory effects from sex, tank, the interaction of these effects and the quadratic polynomial B-spline nested in this interaction indicated that, over time, each sex, grown in different tank, presented different mean trajectory, necessitating the inclusion of nesting time in the interaction sex x tank in order to avoid the under or overestimation of breeding values of the selection candidates in breeding programs of this species.
