Lack of effect of two 17-substituted testosterones on hepatic sulfobromophthalein clearance in dogs

Abstract No effect on hepatic function in dogs given 10–100 mg per day of methyltestosterone or norethandrolone for periods up to 50 days could be demonstrated by the sulfobromophthalein retention test or the sulfobromophthalein clearance rate. Apparently the impairment of hepatic sulfobromophthalein clearance caused by these drugs in humans does not occur in the dog.

Survival Time of Endotoxin Treated Rats as Response Metameter for Corticotropin and Hydrocortisone Bioassays.

Summary 1. Small doses of hydrocortisone decrease the amount of hypothermia and increase survival time of adrenalectomized rats given a lethal amount of E. coli lipopoly-saccharide (endotoxin). 2. Using survival time as the criterion of response, the dose response relationships for hydrocortisone in the adrenalectomized rat and for corticotropin in the hypophysectomized rat were determined. When the log of dose was plotted vs. response, the lines were linear in ranges of 10 to 200 μg of hydrocortisone and 0.5 to 4.5 milliunits of corticotropin. The indices of precision were approximately 0.3 and 1.0 for hydrocortisone and corticotropin, respectively. 3. The effect of sex and age of animals and time of administration on response to hydrocortisone were studied.

The effects of some centrally acting drugs on the glucocorticold induction of hepatic tyrosine transaminase

Abstract The effects of some centrally acting drugs on hepatic tyrosine transaminase activity with and without cortisol in the adrenalectomized male and female rat have been determined. Tyrosine transaminase activity in the female adrenalectomized rat is higher than in the male and is less sensitive to drug augmentation of cortisol induction. Cortisol induction of tyrosine transaminase can be augmented by SKF 525-A 3 hours after cortisol injection. Induction of the enzyme by 16β-methylprednisone, oxylone, and fluocinolone acetonide can be augmented by SKF 525-A and by chlorpromazine.
