Changes in incidence of sex chromatin in subcultured cells.

Sex chromatin counts of subcultured cells of both female human mammary tumor and female rabbit kidney show a considerable drop from an initial high level. Cultures in which sex chromatin persists also retain the viral insensitivity of their source material.

Observations during in vitro growth of cells derived from rabbit kidney

Abstract Two cell lines derived from rabbit kidney have been established in vitro and propagated through 35 passages for approximately 14 months. Both cell lines have retained throughout this period, the morphological, immunological and adhesive properties, as well as original host susceptibility to a number of viruses, which characterize these cells in primary culture. One of the cell lines, RbKi-C, revealed a shift in modal chromosome number from the near diploid to triploid after approximately 10 months cultivation. No accompanying change in morphology or any other cellular property measured was observed. The relative influence of the genotype of cultured cells on permanent alterations in morphology and malignancy is discussed.

A simple stain for mitochondria.

Summary 1. An end-point method of staining mitochondria is described. The staining reaction involves: 1) the combination of mercury a) with RNA and a constituent of dilute solutions of toluidine blue, and b) with lipid of the mitochondrion; 2) subsequent treatment of the cell with a solution of ammonium molybdate and potassium ferrocyanide to precipitate the Hg-RNA-dye complex onto the Hg-lipid of the mitochondrion. 2. The highest degree of permeability to the reagents used and, therefore, the best staining results were obtained in smear preparations which were fixed in the wet state. The method was also applicable to frozen sections.

Infectious Bovine Rhinotracheitis (IBR) IV. Cytological Changes in Infected Bovine Kidney and HeLa Cultures

Summary In both bovine kidney and HeLa cultures, extensive focal nuclear degeneration followed inoculation with a HeLa-adapted IBR virus strain. The pattern of nuclear and cytoplasmic degeneration was similar for the native host (PBK) and the HeLa cell. In cells of both types, nuclear inclusions were Feulgen-negative, and morphologic nuclear changes were accompanied by a reduction of DNA.
