An irreversible one-pot enzymatic synthesis of sialyl oligosaccharides has been achieved with a β-galactosidase-catalyzed galactosylation of an acceptor followed by a sialyltransferase-catalyzed sialylation with regeneration in situ of CMP-sialic acid.

Abstract The Ca 2+ binding properties of the cell-surface carbohydrate sialyl Lewis x, SLe x , and its 2-, 3-, and 4- deoxygenated-fucose analogs, were examined by pneumatically-assisted electrospray (ion spray) mass spectrometry. Computer-assisted modeling and mass spectral evidence suggest that Ca 2+ is coordinated primarily to the GalGlcNAc moeity of SLe x .
