BACKGROUNDnAnimal behavioral models of voluntary ethanol consumption represent a valuable tool to investigate the relationship between age and propensity to consume alcohol using an experimental methodology. Although adolescence has been considered as a critical age, few are the studies that consider the preadolescence age. This study examines the ethanol consumption/preference and the propensity to show an alcohol deprivation effect (ADE) after a short voluntary ethanol exposure from a developmental perspective.nnnMETHODSnThree groups of heterogeneous Wistar rats of both sexes with ad libitum food and water were exposed for 10 days to 3 ethanol solutions at 3 different ontogenetic periods: preadolescence (PN19), adolescence (PN28), and adulthood (PN90). Ethanol intake (including circadian rhythm), ethanol preference, water and food consumption, and ADE were measured.nnnRESULTSnDuring the exposure, the 3 groups differed in their ethanol intake; the greatest amount of alcohol (g/kg) was consumed by the preadolescent rats while the adolescents showed a progressive decrease in alcohol consumption as they approached the lowest adult levels by the end of the assessed period. The pattern of ethanol consumption was not fully explained in terms of hyperphagia and/or hyperdipsia at early ages, and showed a wholly circadian rhythm in adolescent rats. After an abstinence period of 7 days, adult rats showed an ADE measured both as an increment in ethanol consumption and preference, whereas adolescent rats only showed an increment in ethanol preference. Preadolescent rats decreased their consumption and their preference remained unchanged.nnnCONCLUSIONSnIn summary, using a short period of ethanol exposure and a brief deprivation period the results revealed a direct relationship between chronological age and propensity to consume alcohol, being the adolescence a transition period from the infant to the adult pattern of alcohol consumption. Preadolescent animals showed the highest ethanol consumption level. The ADE was only found in adult animals for both alcohol consumption and preference, whereas adolescents showed an ADE only for preference. No effect of sex was detected in any phase of the experiment.

Rats were conditioned across 2 consecutive days where a single unsignaled footshock was presented in the presence of specific contextual cues. Rats were tested with contexts that had additional stimulus components either added or subtracted. Using freezing as a measure of conditioning, removal but not addition of a cue from the training context produced significant generalization decrement. The results are discussed in relation to the R. A. Rescorla and A. R. Wagner (1972), J. M. Pearce (1994), and A. R. Wagner and S. E. Brandon (2001) accounts of generalization. Although the present data are most consistent with elemental models such as Rescorla and Wagner, a slight modification of the Wagner-Brandon replaced-elements model that can account for differences in the pattern of generalization obtained with contexts and discrete conditional stimuli is proposed.

Forward and backward blocking of taste preference learning was compared in rats. In the forward condition, thirsty rats were exposed to a flavor (A) in sucrose solution (+) or in water (-), after which they were exposed to A in compound with another flavor (B) in sucrose solution (i.e., AB+). In the backward condition, these phases were reversed. Consumption of B alone was assessed when rats were food deprived. In the forward condition, rats given A+ consumed less B than rats given A-, providing evidence of forward blocking, whereas in the backward condition, rats given A+ drank more of B than those given A-. Subsequent experiments found that alternating but not blocked preexposure to A and B, when given prior to training, produced blocking of B whether A+ was given before or after AB+, suggesting that prior failures to observe backward blocking reflect failures of discrimination.

In one experiment half of the animals were trained to avoid a signaled footshock by jumping (30 or 160 trials), whereas the rest of the animals received the same events as yoked. For all of them the termination of the warning signal and of the shock was followed by a safety signal. Several tests were conducted to assess the ability of the stimuli to suppress licking by measuring the latency in completing 25 consecutive licks in the presence of the stimuli. Fear of the warning signal and inhibitory properties of the safety signal (summation and retardation tests) were measured. The results showed that there were no differences in fear to the warning signal, and that the safety signal behaves as a conditioned inhibitor only for animals trained with a long avoidance procedure, but not in the yoked (classical conditioning) procedure. These results highlight the role played by the avoidance response and its consequences in avoidance learning.

In two experiments, rats received preexposure consisting of six intraperitoneal injections of lithium chloride (LiCl). This treatment reduced the magnitude of the unconditioned response (UR; suppressed consumption of a novel flavor) evoked by an additional injection (Experiment 1) or by oral consumption (Experiment 2) of LiCl. In both experiments, preexposure also attenuated the acquisition of a conditioned aversion with an LiCl injection as the unconditioned stimulus (US) but had no effect on the aversion produced when the US was oral consumption of LiCl (Experiment 2). These results are consistent with the view that the reduced ability of the preexposed US to serve as a reinforcer depends on blocking by injection-related cues and is independent of habituation of the UR recorded in the present study. Possible interpretations of this dissociation are discussed.

Two experiments, using rats as subjects, examined the role of contextual cues in producing the unconditioned stimulus (US) pre-exposure effect in conditioned taste aversion. Experiment 1 showed a significant US pre-exposure effect, when the pre-exposure was conducted in a familiar context, and that a change of context between the pre-exposure and conditioning phases did not attenuate this effect. Experiment 2 demonstrated that extinction of injection-related cues after the pre-exposure stage attenuated the US pre-exposure effect, when the pre-exposure was conducted in either a familiar or a novel context. Taken together, these results support the associative explanation of the US pre-exposure effect in terms of blocking, incorporating a role for injection-related cues in the context blocking analysis of the US pre-exposure effect.

In two experiments rats were preexposed to neutral stimuli. Both experiments used a between-subjects design in which a paired group was preexposed to intermixed presentations of A→ Band AX, and an unpaired control group was preexposed to intermixed presentations of A, B, and AX. After the conditioning of B, in Experiment 1, conditioned responding to X was acquired more slowly in the paired than in the unpaired group. Furthermore, in Experiment 2, Xreduced conditioned responding to a separately trained excitor in a summation test but only in the paired group. Together, these results provide evidence of an inhibitory form of sensory preconditioning.

In one experiment using conditioned taste aversion and the unconditioned stimulus (US) preexposure procedure, one group of rats was given LiCl exposure for 3 days, whereas two other groups received saline. Following this phase, all groups were given a novel flavour (saccharine) to drink following either LiCl (group preexposed and one of the control groups) or saline injections (the remaining control group) and the consumption of the flavour was assessed. After this neophobia test, the acquired saccharine aversion was evaluated. The results show that three LiCl injections are enough to produce a US preexposure effect on backward excitatory taste aversion conditioning, whereas this number of injections procedure does not produce habituation of the increment in neophobia, an unconditioned response to the LiCl. The results are discussed taking into account different mechanisms involved in US preexposure effect.

This article studied the role of contextual cues, present at the time flavor conditioning occurs, on intake behavior in rats. In three experiments animals were given flavor-sucrose pairings in one distinctive context (Context A) whereas the flavor was presented unreinforced in an alternative context (Context B). Experiments 1 and 2 used a simple Pavlovian discrimination procedure (A: X+, B: X-) and tested consumption of flavor X in each context. Consumption of the flavor was higher in Context A than in Context B. In Experiment 2 rats were given a treatment (exposure to water in the context) designed to extinguish associations between the context and the reinforcer. This procedure did not affect the ability of the context to control intake of flavor X. Experiment 3 used a biconditional discrimination procedure (A: X+, Y-; B: X-, Y+; where X and Y were different flavors) in which no single context or flavor predicted reinforcement. The rats learned this discrimination, consuming more of each flavor in the context in which it had previously been reinforced. The results are interpreted in terms both of the effects of direct associations between context and events presented in them, and in terms of the modulatory or occasion-setting properties of the context.

In three experiments, rats given pairings of a neutral flavor with sucrose showed a preference for that flavor when subsequently allowed to choose between it and water. Preexposure to the flavor produced a latent inhibition effect (reduced the size of the preference) when the rats were hungry during the test (Experiments 1 and 2). Rats that were not hungry during the test failed to show latent inhibition (Experiments 1-3). Experiment 3 confirmed that sucrose-flavor pairings were capable of producing a preference even in nonhungry rats. It is argued that the preference shown by rats that are hungry on test depends on a flavor-nutrient association, a form of learning that is susceptible to latent inhibition in the same way as standard conditioning procedures are. The failure to obtain latent inhibition in nonhungry rats suggests that the preference obtained in these conditions depends on a different form of learning that is less susceptible to the effects of stimulus exposure.
