SPIRULINA PLATENSIS PROTECTS AGAINST GENTAMICIN-INDUCED NEPHROTOXICITY IN RATS

The present study aimed to investigate the protective effect of Spirulina platensis (SP) on gentamicin sulphate (GS)‐induced changes in the levels of lipid peroxidation and endogenous antioxidants in the kidney of rats. Sprague‐Dawley rats were treated in separate groups as follows for 7 consecutive days: control (C), gentamicin sulphate (100 mg/kg i.p.) (GS), Spirulina platensis (1000 mg/kg orally) (SP) and Spirulina platensis (1000 mg/kg orally) plus gentamicin sulphate (100 mg/kg i.p.) (SP + GS). The degree of protection was evaluated by determining the effects of Spirulina platensis on malondialdehyde (MDA), glutathione (GSH), superoxide dismutase (SOD), glutathione peroxidase (GPX) and nitric oxide (NO), and plasma creatinine and urea levels were estimated in kidney homogenates to evaluate antioxidant activity, and the kidney was histologically examined as well. Spirulina platensis elicited significant nephroprotective activity by decreasing lipid peroxidation (MDA) and elevated the levels of GSH, SOD, GPX, NO, creatinine and urea. Furthermore, these biochemical observations were supplemented by histological examination of the rat kidneys. In conclusion, the present study indicates a very important role of reactive oxygen species (ROS) and the relation to renal dysfunction and point to the therapeutic potential of Spirulina platensis in gentamicin sulphate induced nephrotoxicity. Cyclophosphamide İle İndüklenmiş Ratlarda Karaciğer Enzimleri (AST, ALT, ALP) ve Histopatolojisi Üzerine Naringin’in Protektif Etkileri

Bu calismada Cyclophosphamide (CYP) uygulamasi yapilan ratlarda karaciger enzimleri (Aspartat aminotransferaz (AST), Alanin aminotransferaz (ALT), Alkalen fosfataz (ALP) ve histopatolojisi uzerine Naringin’in protektif etkilerinin arastirilmasi amaclanmaktadir. Arastirmamizda yaklasik 200-250 g agirliginda, 40 adet erkek eriskin Sprague Dawley irki rat kullanildi ve 5 grup olusturuldu. Karaciger enzimlerinin analizleri otoanalizorde ve spektrofotometrede yapildi. Hematoksilen eosin ile boyanan karaciger doku ornekleri isik mikroskobunda incelendi. CYP’nin ratlarda hepatotoksiteye yol actigi ve karaciger enzimlerini artirdigi goruldu. AST, ALT ve ALP degerleri Kontrol grubunda 58±11, 32±8, 38±6; CYP grubunda 237±42, 168±44, 74±11; Naringin 50+CYP grubunda 223±33, 158±42, 62±9; Naringin 100+CYP grubunda 117±25, 107±24, 48±8; Naringin 100 grubunda ise 54±9, 31±7, 36±6 sirasiyla olarak bulundu. Kontrol grubundan elde edilen degerlere gore CYP verilen gruplarda ki AST, ALT ve ALP degerlerinin tamaminda P<0.001 duzeyinde artis soz konusudur. CYP’nin karaciger dokusunda olusturdugu dejeneratif ve nekrotik doku hasarinin Naringin’in etkisiyle dozlara bagli olarak azaldigi saptandi. Sonuc olarak, Naringin’in farkli dozlarinin, CYP ile induklenen hepatotoksisitede protektif etkili oldugu belirlendi.

Determination and pharmacokinetics of recombinant human growth hormone in rabbit plasma by spectrofluorometry method.

In this study, a new spectrofluorometry method was developed for the determination of human growth hormone (somatotropin) in rabbit plasma. The validation was assessed under a variety of conditions (FDA, Bioanalytical Method Validation Guidline). Calibration curves were linear between the concentration range of 0.125-8.0 μg/mL. Intraand inter-day precision values for somatotropin in plasma were less than 13.5, and accuracy (relative error) was better than 12.0%. The recoveries for all samples were >84.0%. The limits of detection (LOD) and quantification (LOQ) of somatotropin were 0.075 and 0.10 μg/mL, respectively. The described spectrofluorometry method has adequate sensitivity and specificity to study pharmacokinetics of somatotropin in rabbits, and could be adapted also to clinical pharmacokinetic study.
