A significant portion of the genome is transcribed as long noncoding RNAs (lncRNAs), several of which are known to control gene expression. The repertoire and regulation of lncRNAs in disease-relevant tissues, however, has not been systematically explored. We report a comprehensive strand-specific transcriptome map of human pancreatic islets and β cells, and uncover >1100 intergenic and antisense islet-cell lncRNA genes. We find islet lncRNAs that are dynamically regulated and show that they are an integral component of the β cell differentiation and maturation program. We sequenced the mouse islet transcriptome and identify lncRNA orthologs that are regulated like their human counterparts. Depletion of HI-LNC25, a β cell-specific lncRNA, downregulated GLIS3 mRNA, thus exemplifying a gene regulatory function of islet lncRNAs. Finally, selected islet lncRNAs were dysregulated in type 2 diabetes or mapped to genetic loci underlying diabetes susceptibility. These findings reveal a new class of islet-cell genes relevant to β cell programming and diabetes pathophysiology.

L’allogreffe d’ilots de Langerhans, qui repose sur la transplantation exclusive des ilots endocrines, soit quelques millilitres de tissu, isoles a partir de pancreas de sujets non-diabetiques et infuses dans la veine porte, permet, dans les centres les plus experimentes, d’obtenir des resultats identiques, a court terme, a ceux de la greffe de pancreas (80 % d’insulino-independance a un an). Le developpement clinique de la therapie cellulaire du diabete est donc crucial et se heurte aujourd’hui a trois obstacles majeurs : le rejet immunitaire, la faible implantation initiale des ilots et la duree limitee de leur fonction chez le receveur. Les progres previsibles laissent envisager une extension de ses indications dans les annees a venir chez les patients atteints des formes les plus severes du diabete de type 1. La penurie structurelle des donneurs d’organes limitera, cependant, le developpement de l’allogreffe d’ilots primaires. L’implication dans la production de cellules insulinosecretrices, en vue de leur transplantation chez l’homme, represente un enjeu important pour le traitement du diabete par la therapie cellulaire, en particulier a partir de la proliferation des ilots humains adultes, de l’exploitation des donneurs vivants ou a cœur arrete, de l’obtention des ilots xenogeniques, de la proliferation et la differenciation des cellules souches et des cellules precurseurs, et de la transdifferenciation.
