Exposure to waterborne 4-tert-octylphenol induces vitellogenin synthesis and disrupts testis morphology in the South American freshwater fish Cichlasoma dimerus (Teleostei, Perciformes)

Exposure to environmental pollutants may disrupt endocrine functions and cause reproductive effects in human and wildlife populations. Various groups of chemicals have estrogen-like effects, including degradation products of alkylphenol polyethoxylates, such as 4-tert-octylphenol (OP). Laboratory studies have shown that exposure of male fish to xenoestrogens results in induction of circulating vitellogenin (Vtg), inhibition of testicular growth, testis abnormalities and formation of intersex gonads. In this study, the impact of the exposure to waterborne OP on reproductive aspects in the South American cichlid fish Cichlasoma dimerus was evaluated using qualitative changes in the levels of Vtg in plasma and surface mucus and histological alterations in the liver and gonads as endpoints. Adult males and females were exposed to OP via immersion during 60 days in aquaria under semi-static conditions, water changes being made every 84 h. Treatment groups were: control (ethanol 0.005%), OP 30, 150 and 300 microg/L. Using Western and Dot blot analysis, Vtg was detected in plasma and mucus of control and treated females and treated males, while no Vtg was observed in samples from control males. Morphological changes in the hepatocytes due to the accumulation of Vtg were observed in OP-exposed males. Impairment of testicular structure became apparent in males treated with the highest OP concentrations. The most salient pathological change was the alteration of lobular organization with increased testicular fibrosis and progressive disruption of spermatogenesis. No major changes were observed in ovarian architecture. Our results indicate that detection of Vtg in surface mucus may be a sensitive and non-invasive biomarker of the endocrine disrupting effects of environmental estrogens, resulting in a useful method for field monitoring.

Ultrastructure of the testis in Synbranchus marmoratus (Teleostei, Synbranchidae): the germinal compartment

Synbranchus marmoratus, is a protogynic diandric species in which two types of males, primary and secondary, are found. In both types, the germinal compartment in the testes is of the unrestricted lobular type, but in secondary (sex reversed females) males the lobules develop within the former ovarian lamellae. In the present study, the germinal compartment was examined in both types of males using light microscopy as well as scanning and transmission electron microscopy. Germinal compartment is limited by a basement membrane and contains Sertoli and germ cells. During maturation, processes of Sertoli cells form the borders of spermatocysts containing isogenic germ cells. Characteristically, type A and type B spermatogonia have a single nucleolus and grouped mitochondria associated with dense bodies or nuage. Type B spermatogonia, spermatocytes and spermatids are joined by cytoplasmatic bridges and are confined within spermatocysts. Secondary spermatocytes are difficult to find, indicating that this stage is of short duration. Biflagellated spermatozoa have a rounded head, no acrosome, and possess a midpiece consisting of two basal bodies, each of which produces a flagellum with a typical 9+2 microtubular composition. No associations occur between sperm and Sertoli cells. There were no differences between spermatogenesis in primary and secondary males in this protogynic, diandric fish.

Characterization of molecular variants of GnRH, induction of spermiation and sex reversal using salmon GnRH-A and domperidone in the protogynous diandric fish, Synbranchus marmoratus Bloch, (Teleostei, Synbranchidae)

This paper studies the molecular variants of gonadotropin-releasing hormone (GnRH) present in the brain of the protogynous swamp eel, Synbranchus marmoratus, and the effects of the administration of salmon GnRH analogue (sGnRH-A) and the dopamine receptor antagonist, domperidone (DOM) on final maturation and gamete release in this species. Evidence for the presence of two GnRH variants, sGnRH and cIIGnRH were obtained by reverse phase high-pressure liquid chromatography (RP-HPLC) and radioimmunoassay with different antisera. The effects of treatment with sGnRH-A+DOM were checked by three ways: oocyte and milt release by stripping, histological analysis of the gonadal tissue, and androgen serum levels at different times throughout the experiment. In males, spermiation was induced after three weeks of treatment. In the female group, sGnRH-A+DOM did not induce ovulation at the end of the experiment. Histological analysis of the gonads from the female group showed evidence of sex reversal. All the treated fish had elevated androgen serum levels from the third week, with respect to control fish. In all cases, serum estradiol levels were undetectable. These results suggest that treatment with sGnRH analog and DOM induce sex reversal in female and spermiation in males.

Nucleotides in embryos in the stages of morula, gastrula, and neurula☆

Levels of nucleotides and sugar nucleotides in embryos of Bufo arenarum at the stages of morula, gastrula, and neurula have been measured. The total amounts of purine nucleoside diphosphates decreased from morula to gastrula, but increased sharply from gastrula to neurula. The levels of ADP followed this pattern, but those of GDP did not change significantly through the three stages. Purine nucleoside triphosphate levels, which had increased immediately after fertilization, remained almost constant through morula, gastrula, and neurula. As with the purine nucleoside diphosphates, the adenine nucleotide decreased from morula to gastrula, and increased from gastrula to neurula. In contrast, the level of GTP showed a sharp maximum at gastrula. The total pyrimidine nucleoside triphosphate did not change significantly from morula through neurula. As in previous stages of development, only uridine sugar nucleotides were detected. A sharp increase of the galactosyl ester of nucleotides was found at gastrula.

Nucleotides and nucleotide sugars in early development of bufo arenarum: Part I. mature oocytes

Abstract A study of nucleotides and nucleotide sugars in mature oocytes of Bufo arenarum has been performed. Two samples of oocytes from ovulations of ten females each were analyzed by ion-exchange chromatography and the following nucleotides and nucleotide sugars found 2 : NAD, UDPGNAc, UDPGalNAc, UDPG, UDP, NADP, CTP, UTP, UDPGA, ADP, GDP, GTP, and ATP. The nucleotides and sugar-nucleotides were identified on the basis of chemical, enzymic, and chromatographic analysis. To minimize the decomposition of unstable substances, the extraction was performed with trichloroacetic acid in the cold, the contact of the sample with the acid was minimal and ammonium chloride was used for the elution from the Dowex 1-resin. In order to detect compounds present in minute amounts, very large samples were used (60,000 to 120,000 oocytes). A nucleotide of complex structure, which binds weakly to the column, was detected in the two samples analyzed.
