Detection of polymorphisms on strawberry (Fragaria spp.) based on cleavage amplified polymorphic sequence

Strawberries (Fragaria spp.) represent a fruit of high economic value. However, its cultivation in Indonesia has not been optimal. Along with the development of technology, plant researchers and breeders have been developing new cultivars to improve the quality of strawberry plants. Morphological selection of superior quality cultivars was found less effective, leading to the development of molecular research. One method to evaluate the result of breeding strawberry plants is to use the Cleavage Amplified Polymorphic Sequence (CAPS) markers, which can be used for the identification of octoploid strawberry cultivars. The purpose of the present study was to investigate the polymorphic banding pattern of strawberry cultivars grown in Indonesia, to determine CAPS marker capabilities for detection of polymorphisms, and to determine the degree of similarity of strawberry cultivars in Indonesia based on the CAPS markers. Molecular characters of melon (Cucumismelo L. “Tacapa”) in response to karst critical land

Yogyakarta district has 158.600 ha critical land and spread off in three Agro Ecosystem zones. Two of them are karsts critical land. Critical lands which contain calcium carbonate in high concentration and water dehydration in upper surface give abiotic stress in wide range of plant. Melon cultivar TACAPA has superior characteristic derived from parental crossing, ♀ Action 434 and ♂ PI 371795 and potential to be developed in karsts critical land. Abscicic acid (ABA) is a phytohormone expressed by plant in abiotic stress condition. CmBG1 is a gene which regulate ABA hormone in melon. The purposes of this research were examining the molecular character of melon cultivar TACAPA in response to karsts critical land in order to study molecular characterization of CmBG1 gene. Karakterisasi Kromosom Stroberi (Fragaria vesca L. subsp. californica Cham. & Schltdl. cv. Californica) Hasil Poliploidisasi

Fruit consumption such as strawberries always increase in Indonesia and reaching about 210 tons in 2011. Strawberry (Fragaria vesca L. subsp. californica Cham. and Schltdl. cv. Californica) has been developed in Indonesia, especially in Banyuroto, Sawangan, the district of Magelang, Central Java. In order to improve the fruit productivity (quality and quantity), genetic characterization including chromosomes character of this fruit are needed to be investigated. Therefore, the research was conducted to determine the chromosomes number, chromosomes size, chromosomes shape and karyotype of control and strawberries treated by colchicine as mutagenic agent. The method was squash and flow cytometry method. Whereas the cell suspension sample were observed used flow cytometer instrument. The result of research shown that the chromosome number of strawberry Californica in normal and treatment strawberries was 2n=14. The chromosomes shape of normal strawberry was 10 metacentric and 4 submetacentric. Whereas the chromosomes shape of treatment strawberries used colchicine was 10 metacentric and 4 submetacentric. Flow cytometric analysis shown that control strawberries and treated strawberries has diploid ploidy level.
