Evaluation of the Brix refractometer as an on‐farm tool for the detection of passive transfer of immunity in dairy calves

OBJECTIVES To investigate the accuracy of both the optical and digital Brix refractometers compared with radial immunodiffusion (RID) for determining the immunoglobulin G (IgG) concentrations in dairy calf serum. DESIGN The experiment design was a cross-sectional survey of four dairy farms. Serum was sampled from 12 calves from each farm at approximately 48 hours of age. METHODS Serum IgG concentrations of 48 calves were measured using RID and both types of Brix refractometer. RESULTS IgG concentrations measured by Brix refractometer scores correlated with RID results: 0.74 and 0.71 for the digital and optical devices, respectively. The minimum Brix score that identified calf serum with success of passive immunity (>1000 mg/dL RID IgG) with 100% accuracy was 10% for both devices. The optical and digital devices performed similarly at identifying IgG concentrations in calf serum, with a concordance of 87%. CONCLUSION Brix refractometer score ≥10% can be used to classify calves with successful transfer of passive immunity and the devices are sufficiently accurate for use as a simple, inexpensive on-farm tool for the monitoring of neonatal dairy calf immunity levels.

Differential rumination, intake, and enteric methane production of dairy cows in a pasture-based automatic milking system.

Proper performance monitoring of cows on pasture-based diets is crucial to inform nutritional recommendations that minimize undesirable effects of high ruminant CH4 emissions into the environment. The prediction of linkages between rumination patterns, methane emissions, and correlated production traits of cows in a pasture-based automatic milking system was tested. A previous 10-d baseline measurement of rumination activity by acoustic methodology of 156 Holstein-Friesian cows was used for frequency analysis of rumination time and identification of 2 treatment groups (n = 37 cows/group) represented by cows with consistently high (HR; 75th rumination percentile = 617.55 ± 81.37 min/d) or low (LR; 25th rumination percentile = 356.65 ± 72.67 min/d) rumination. The HR and LR cows were paired by nearest parity, days in milk, body weight (BW), and previous 10-d milk production, and within pairs randomly assigned to 1 of 2 experimental groups managed on a voluntary milking system with diets consisting of at least 75% pasture, plus concentrates. Animal traits, including rumination time, mass flux of CH4 (QCH4) and carbon dioxide (QCO2), milk production, and estimated dry matter intake according to individual QCO2 fluxes over a 22-d period were analyzed with repeated measure mixed models for a completely randomized design, structural equation modeling, and nonlinear regression. High rumination and methane was seen in older and heavier cows that had greater estimated dry matter intake and milk production. A consistent difference in rumination time and QCH4 across days was detected between HR and LR, even after adjustment for metabolic BW. Estimated dry matter intake had direct positive effects on rumination and QCH4, but no independent direct effect of rumination on QCH4 was detected. The LR cows produced more QCH4/milk, associated with lower milk, BW, concentrate intake, and greater activity at pasture. A typical dilution of maintenance effect on QCH4/milk was detected as a consequence of increasing milk yield and similar significant reduction of QCO2/milk. The results raise challenging questions regarding the rumination patterning of grazing dairy cows and alternatives to reduce ruminant methane emissions in grazing dairy cows.

Seasonal diet selection by ewes grazing within contrasting grazing systems

Grazing management systems seek to control the relationship among animals, plants and soil by regulating the number of animals and the duration and location of animals. A greater understanding of the diet selection and activity of livestock within grazing systems will assist producers to make informed management decisions about their management within complex pastures. In the present paper, differences in the diet quality, selection and activity of ewes managed within contrasting systems (continuous-grazing (CG) and an intensive (20-paddock) rotational-grazing (RG) system) within a native pasture in the Central Tablelands of New South Wales, using non-invasive methodologies, are described. During two time periods (late spring and early autumn), the animals grazing within the CG system consumed a diet of higher quality and spent less time active than did those within the RG system. These differences resulted in higher individual animal production of CG animals that were able to maintain the herbage of preferred areas in a vegetative and highly nutritious state. The grazing animals selected the green herbage of higher quality than the average pasture and adjusted their selection seasonally. An underlying mechanism driving selection is the green : dead ratio of the herbage. Practically the results indicated that the green : dead ratio (or greenness) of herbage may provide a management trigger to enhance the production of animals grazing within a RG system, in particular during periods of higher requirement.

Predicting the quality of browse-containing diets fed to sheep using faecal near-infrared reflectance spectroscopy

The potential of data collected from past feeding trials to derive faecal near-infrared reflectance spectroscopy (fNIRS) calibrations for predicting the attributes of browse-containing sheep diets was examined. Reference data and faecal near-infrared spectrum pairs (n = 240) originated from five feeding trials involving 40 diets consisting of varying levels of fresh browse and oaten chaff. The fNIRS calibrations were developed to predict crude protein (CP), total phenolics (TP), total tannin (TT) and phosphorus (P) contents, protein precipitation capacity of tannin (PPC), in vivo digestibility of dry matter (DMD), organic matter (OMD) and crude protein (CPD) and in vitro OMD (IVOMD), metabolisable energy (ME) and short chain fatty acid production (eSCFA) in the diet. The precision of calibrations was evaluated by the coefficient of determination (R2c) and standard error (SEC) of calibration. The predictive ability of calibrations was evaluated by standard error of cross-validation (SECV), standard error of prediction (SEP), slope of the validation regression and the ratio of the standard deviation of the reference data to the SECV (RPD). For all fNIRS calibrations, R2c was >0.80 and SEC was close to the respective SECV. Slope of the validation regressions did not deviate from 1 for chemical attributes but deviated from 1 for functional attributes (except eSCFA). The RPD of DMD and OMD was 3 for CP, TP, TT, PPC, P, CPD, IVOMD, ME and eSCFA calibrations. Data derived from the past feeding trials could be used to derive robust fNIRS calibrations to predict chemical attributes (CP, TP, TT, PPC, P) of browse-containing sheep diets. Although, fNIRS calibrations predicting dietary in vitro functional properties (digestibility and ME) were superior to those predicting in vivo functional properties, both were not so robust. Statistics of fNIRS calibrations derived using reference data originating from in vitro methods needs to be carefully interpreted.

Using faecal DNA to determine consumption by kangaroos of plants considered palatable to sheep.

Disagreement exists within the scientific community with regards to the level of competition for feed between sheep and kangaroos in the Australian rangelands. The greatest challenge to solving this debate is finding effective means of determining the composition of the diets of these potential grazing competitors. An option is to adopt a non-invasive approach that combines faecal collection and molecular techniques that focus on faecal DNA as the primary source of dietary information. As proof-of-concept, we show that a DNA reference data bank on plant species can be established. This DNA reference data bank was then used as a library to identify plant species in kangaroo faeces collected in the southern rangelands of Western Australia. To enhance the method development and to begin the investigation of competitive grazing between sheep and kangaroos, 16 plant species known to be palatable to sheep were initially targeted for collection. To ensure that only plant sequences were studied, PCR amplification was performed using a universal primer pair previously shown to be specific to the chloroplast transfer RNA leucine (trnL) UAA gene intron. Overall, genus-specific, single and differently sized amplicons were reliably and reproducibly generated; enabling the differentiation of reference plants by PCR product length heterogeneity. However, there were a few plants that could not be clearly differentiated on the basis of size alone. This prompted the adoption of a post-PCR step that enabled further differentiation according to base sequence variation. Restriction endonucleases make sequence-specific cleavages on DNA to produce discrete and reproducible fragments having unique sizes and base compositions. Their availability, affordability and simplicity-of-use put restriction enzyme sequence (RES) profiling as a logical post-PCR step for confirming plant species identity. We demonstrate that PCR-RES profiling of plant and faecal matter is useful for the identification of plants included in the diet of kangaroos. The limitations, potential and the opportunities created for researchers interested in investigating the diet of competing herbivores in the rangelands are discussed.

Pumpkin kernel and garlic as alternative treatments for the control of Haemonchus contortus in sheep.

Two alternative remedies for controlling Haemonchus contortus in sheep (pumpkin kernel and garlic) were investigated. The experiment involved 18 4-month-old Merino ram lambs with six lambs per treatment group. The lambs were initially drenched to render them worm free, and then infected with 4000 L3 H. contortus larvae, 2 weeks later. Four weeks after infection, faecal egg counts were performed and lambs were allocated into treatment groups and fed their respective diets (control, pumpkin kernel or garlic) for 2 weeks. The lambs were combined into a single group grazing pasture for the last week of the experiment. Faecal worm egg counts (WEC) were carried out weekly for 3 weeks following allocation to treatment diets. Clinical signs of infection observed included liveweight, body condition score and voluntary feed intake. The pumpkin kernel treatment resulted in a 65.5% decrease in the initial level of WEC, but this increased back to the initial level as soon as animals came off treatment. The garlic resulted in a 64.4% decrease in WEC from the initial level and this increased slightly (to 25.5% of the initial level and 43.5% lower than the control) when the animals came off treatment, suggesting that there was a residual effect of the garlic and/or that the garlic had an effect beyond decreasing the fecundity of the parasites. There was no significant difference (P > 0.05) between treatments in liveweight, body condition score or voluntary feed intake. Throughout the experiment voluntary feed intake and liveweight increased while body condition scores remained stable. We concluded that pumpkin kernel and garlic show potential for parasite control by affecting the fecundity of the parasites. Our results also indicate that with good nutrition lambs can cope with high parasite burdens and still be productive during the early stages of infection.

Prevalence and effect of Theileria orientalis infection in homebred calves in the Gloucester region of New South Wales, Australia

Objectives To examine the prevalence of Theileria orientalis in the Gloucester area of New South Wales and its effect on individual animals. Methods Blood samples (EDTA and clotted blood) were collected from a total of 55 calves and their dams from 6 properties over a 16‐week period. A total of 202 and 190 blood samples were collected from the calves and dams, respectively, and were examined via blood film for the presence of intraerythrocytic T. orientalis piroplasms. Packed cell volume (PCV) was measured to determine infection resulting in anaemia. The presence of antibodies against the T. orientalis major piroplasm surface protein (MPSP) was tested using ELISA. Results The overall prevalence of T. orientalis infection in both dams and calves from all herds examined was 95%. Mean peak parasitaemia was observed in calves between 6 and 9 weeks of age (P = 0.051), coinciding with a decline in mean PCV. Only 3 (6%) of the blood samples collected from the dams were positive for Theileria‐associated antibodies and no significant relationship (P > 0.05) was found between the presence of antibodies in the dams and PCV levels in the calves. There was no evidence that passive transfer of antibodies from dams to calves protected the calves against a decline in PCV. Conclusion This study confirmed a high prevalence of low‐level Theileria infection, but low MPSP seroconversion rates, in dams and calves in an area where the disease has been endemic for a number of years.

Acute renal failure associated with Amaranthus species ingestion by lambs

CASE REPORT This report documents an outbreak of acute renal failure in a flock of 9-month-old White Suffolk-cross ewe lambs. The mortality rate was moderate but the morbidity was high. The lambs had grazed a wheat stubble paddock in which there were several weed species, including Amaranthus spp. CLINICAL SIGNIFICANCE Renal failure in livestock has been reported internationally following the ingestion of Amaranthus spp. and although it has previously been suspected in Australia no cases have been documented. It is important for Australian veterinarians and farmers to realise the toxic potential of this group of plants.

Effects of increasing the inclusion level of Atriplex amnicola in the diet of sheep.

The effects of increasing the level of inclusion of fresh Atriplex amnicola Paul G.Wilson foliage in an oaten chaff (Avena sativa L.) diet fed to sheep were investigated. Six experimental diets were formulated to contain varying amounts [0, 161, 362, 496, 650 and 836 g/kg diet dry matter (DM)] of A. amnicola, and these diets were fed to individually penned sheep according to a Latin square design. Feed and faecal samples were analysed for crude protein, neutral detergent fibre, acid detergent fibre, acid detergent lignin, and ash contents. Mineral contents and 24 h in vitro gas production were measured in freeze-dried feed samples. As the level of inclusion of A. amnicola increased, digestibility of DM, organic matter and neutral detergent fibre increased (P < 0.05), reaching the peak at the 496 g/kg DM inclusion level and then decreasing (P < 0.05) thereafter. Acid detergent fibre digestibility followed a similar trend, but peak digestibility was at 362 g/kg DM inclusion level. Digestibility of crude protein continued to increase (P < 0.05) with increasing inclusion of A. amnicola. Gas production (24 h) from A. amnicola was lower (P < 0.05) than that from oaten chaff (24.4 v. 45.3 mL/200 mg DM) and also had lower metabolisable energy content (in vitro ME, 6.3 v. 8.6 MJ/kg DM). As the level of inclusion of A. amnicola in the diet increased, gas production and in vitro metabolisable energy content decreased (P < 0.05). The 496 g/kg DM inclusion level provided the optimum N : energy ratio for fermentation of dietary fibre. At this level of inclusion the diet was rich in Na, Ca, P, Mg, K, S, Mn and Zn. High inclusion levels improved the crude protein value of the diet but compromised fibre digestibility.

Expeller Barrel Dry Heat and Moist Heat Pressure Duration Induce Changes in Canola Meal Protein for Ruminant Utilisation

Simple Summary Canola meal, a by-product of oil production from canola seed, is a source of protein commonly incorporated into dairy and feedlot rations. Processing conditions and pressure treatments can alter the quality of protein in canola meal. In this study, the impact of expeller dry heat and moist heat pressure duration time on general nutritional properties, in vitro protein degradability, Maillard reaction product formation, and molecular and microscopic structural characteristics of canola meal were investigated. Increased dry heat temperature rapidly increased digestible protein and non-protein nitrogen content, and constricted amide II secondary structure. Increased moist heat pressure treatment duration promoted browning, and the conversion of protein to more intermediately and slowly degradable forms. Dry heat and moist heat pressure affected meal protein solubility and protein and lipid-related functional groups. Moist heat pressure fragmented canola meal into enzyme-resistant aggregates with crevices containing oil bodies. Induced changes may impact the supply of protein and amino acids and subsequently the yield and composition (protein and lipid) of milk produced by dairy cows. These findings benefit producers of canola meal by further describing the effects of processing and treatment conditions on protein characteristics, particularly those which affect the production potential of ruminants fed canola meal as a source of protein. Abstract To improve the protein nutritional quality of canola (Brassica napus L.) meal, further investigation of the effects of processing conditions and post-production treatments is desirable. The impact of barrel dry heat temperature (20 °C (cold press) and 100 °C (expeller)) and moist heat pressure (MHP) duration time on general nutritional properties, Maillard reaction product (MRP) formation, in vitro protein degradability, and molecular and microscopic structural characteristics of canola meals were investigated. Increased MHP duration reduced (p < 0.05) dry matter, soluble protein, rapidly degradable protein, yellowness (early MRP), whiteness (late MRPs), absorbance at 294 nm (intermediate MRPs), and amide I; and increased (p < 0.05) non-protein N, neutral detergent fibre, neutral detergent insoluble crude protein (CP), intermediately and slowly degradable protein, in vitro effective CP degradability, redness, degree of colour change, and browning. Increased dry heat temperature reduced (p < 0.01) CP and rapidly degradable protein, constricted amide II, reduced (p < 0.05) protein solubility in 0.5% KOH and increased (p < 0.05) acid-detergent fibre and intermediate MRPs. Browning index and redness exhibited potential as rapid indicators of effective CP degradability and soluble protein, respectively. Dry heat and MHP altered (p < 0.05) lipid-related functional groups. Dry heat affected napin solubility, and MHP altered cruciferin and napin solubility. Application of MHP induced the formation of proteolysis-resistant protein aggregates with crevices containing oil bodies. Induced changes may impact the supply of proteins and amino acids and subsequently the yield and composition (protein and lipid) of milk produced by dairy cows.
