Publisher Summary This chapter discusses the present state of knowledge of cloning and expression vectors for A. nidulans and N. crassa . It provides an overview on the basic biology and genetic systems of A. nidulans and N. crassa to provide sufficient background information to understand the fungal transformation systems. Both of these fungi possess haploid vegetative mycelia, which under appropriate cultural conditions produce abundant uninucleate or multinucleate conidia. Under normal conditions, the diploid stage of the life cycle is transient, occurring just before meiosis. The sexual fruiting bodies contain many haploid ascospores. In N. crassa , the ascospores are contained within ordered tetrads. Mating in N. crassa is regulated by a mating-type locus with two alleles, whereas A. nidulans is essentially homothallic; that is, a single haploid conidium gives rise to a fertile ascocarp by self-fertilization. Extensive genetic maps are available for both organisms, and hundreds of genetically defined strains are available from culture collections.
