Synthesis of a new Photo-Cross-Linking Nucleoside Analogue Containing an Aryl(Trifluoromethyl)Diazirine Group: Application for Eco RII and Mva I Restriction-Modification Enzymes

Abstract A new photo-cross-linking dU analog, 5-[4-(3-(trifluoromethyl)-3H-diazirin-3-yl)phenyl]-2′-deoxyuridine, was synthesized and incorporated into the recognition site of Eco RII and Mva I restriction-modification enzymes. The resulting base-modified 14-mer substrate was tested for cross-linking to these enzymes. Cross-linking is effected by irradiation of the enzyme-substrate complexs at 366 nm.

Probing the MvaI Methyltransferase Region that Interacts with DNA: Affinity Labeling with the Dialdehyde-Containing DNA Duplexes

Abstract Affinity labeling of methyltransferase MvaI by DNA duplexes containing oxidized 2′-O-β-D-ribofuranosylcytidine or 1-(β-D-galactopyranosyl)thymine residues was performed. Partial chemical hydrolysis of the covalently bound methylase in the conjugates with the dialdehyde-containing DNA allowed us to determine the amino acid region in the C terminus of methylase MvaI that interacts with DNA.

Abstract To study the interaction of the restriction endonucleases Mval and Eco RII with DNA we have synthesized some modified oligonucleotides. The results of hydrolysis demonstrate that both enzymes cleave their substrate by different mechanism.
