Xiao-Long Yuan; Min Cao; Guiqi Bi

Abstract Pseudo-nitzschia multiseries belongs to Baciuariophyta, which can produce the neurotoxin known as domoic acid, a toxin that can cause amnesic shellfish poisoning in human beings. The research aspects mainly focus on its toxicity and diversity. However, there is no mitochondrial genome information about P. multiseries. The complete mitochondrial genome of P. multiseries was obtained through amplifying. The mitogenome is 46 283 bp in length with 31.04% GC content. It has a typical cyclic structure. The genome contains 37 protein-coding genes, including unidentified open reading frames, 23 tRNAs and 2rRNAs. Four group II introns were found in rps5, cob, cox1 and cox2. The co-linear analysis result exhibited existence of gene modules re-arrangements and inversions among P. multiseries, Synedra acus and Thalassiosira pseudonana. The gene contents comparison showed they have high similarities. Phylogeny results suggest P. multiseries has a close evolutionary relationship with T. pseudonana.

Guiqi Bi; Guoqiang Liu; Ezi Zhao; Qingwei Du

Abstract 454 GS FLX Titanium sequencing data were used to obtain the complete mitochondrial genome of Calliarthron tuberculosum (26,469 bp). The mitogenome contains 44 genes, including 2 ribosomal RNA, 19 transfer RNA, 5 ribosomal proteins, 1 ymf, 2 open reading frames (ORFs) and 16 genes involved in cellular respiration. The secY and rps12 are overlapped by 69 bp. rps3 and rpl16 also have a 45 bp overlapped region. Except for open reading frames near stem-loop region, the results show Calliarthron tuberculosum mtDNA has a high similarity with other Florideophyceae algae mitogenomes in gene synteny, structure and gene content. Phylogeny analysis indicates a close genetic relationship of Calliarthron tuberculosum with Sporolithon durum.

Complete sequence and analysis of plastid genomes of Pseudo-nitzschia multiseries (Bacillariophyta)

Min Cao; Xiao-Long Yuan; Guiqi Bi

Abstract Pseudo-nitzschia multiseries are common members of phytoplankton communities, which can bring about blooms in coastal and ocean. Meanwhile, the neurotoxin was produced by this species can cause amnesic shellfish poisoning in both marine mammals and human beings. Whereas the lacking of plastid genome information on this species obstacles the study of its evolutionary position. Here, the complete plastid genome sequence of Pseudo-nitzschia multiseries is presented. The genome is a circular DNA composed 111 539 bp of with one rRNA operon. The average G + C content of this plastid genome is 31.37%. This genome includes 96 protein coding genes (containing four unidentified open reading frames), 27 transfer RNA genes, three ribosomal RNA genes, and one tRMA. The comparison between P. multiseries and Thalassiosira pseudonana has a high similarity in gene content and structure. In addition, our phylogenetic result suggests that the P. multiseries is the sister group of T. pseudonana.

Complete mitochondrial genome of Fistulifera solaris (Bacillariophycidae).

Xianghai Tang; Guiqi Bi

Abstract The complete mitochondrial DNA of pennate diatom Fistulifera solaris was determined. The circular mitochondrial genome is 39 648 bp in length with 28.1% GC content and contains 63 genes, including 33 protein-coding genes, three conserved open reading frame (ORF154, ORF192 and ORF251), 25 tRNA and 2 rRNA genes. All protein-coding genes have AUG as start codon. A group I intron was found in nad11 gene. Both the gene content and the structure of Fistulifera solaris mitochondrial genome are very similar to Berkeleya fennica mitogenome (KM886611). Phylogeny analysis indicate F. solaris a close genetic relationship with Berkeleya fennica. This new mitogenome will provide more useful information for exploration in diatoms diversity and evolution.

The complete chloroplast genome of Guillardia theta strain CCMP2712.

Xianghai Tang; Guiqi Bi

Abstract The complete chloroplast sequence of Cryptophyceae algae Guillardia theta strain CCMP2712 was determined in this study. The genome consists of 138 455 bp containing a pair of rRNA-coding invert repeats of 4973 bp. The overall GC contents of the chloroplast genome were 32.9%. A total of 159 functional genes were annotated, which included 124 protein-coding genes, 30 tRNAs, 1tmRNA and 4 rRNA genes. Short intergenic regions, no introns and some overlapping genes make this cp genome compact. Phylogenetic analysis with the related algae cp genomes revealed that G. theta strain CCMP2712 a typical Cryptophyta algae and had a close genetic relationship with Rhodomonas salina (EF508371) and another previously reported G. theta (AF041468).

Qi Mao; Guiqi Bi

Abstract Ficus racemosa, with immense medicinal value, and known as Cluster Fig Tree, Indian Fig Tree or Goolar (Gular) Figis, is a species of plant which belongs to family Moraceae. The complete chloroplast genome of Ficus racemosa was obtained by de novo assembly using next-generation sequencing data. The chloroplast genome of F. racemosa was 159 473 bp in length, which consisted of a large single region (88 110 bp), a small single copy region (20 007 bp) and a pair of invert repeat regions (25 678 bp). The overall GC content of this chloroplast genome was 36.0%. The chloroplast genome harbored 117 genes, including 84 protein-coding genes, 27 tRNA, and eight rRNA genes (4.5S rRNA, 5S rRNA, 16s rRNA and 23s rRNA) that were two copied. Phylogenetic analysis of the complete chloroplast genome sequences with the report-related chloroplast genomes revealed that Ficus racemosa is most closely related to Morus indica, a typical higher plant in fiamly Moraceae.
