Abstract A novel hydrazine and primary amine labile (1,3-dimethyl-2,4,6 (1 H ,3 H ,5 H )-trioxopyrimidine-5-ylidene)methyl (DTPM) vinylogous amide type protecting group was developed to protect aminosugars and other primary amines in a facile and cost effective manner.

The macrolide antibiotic primycin is shown to consist of more than twenty compounds. Nine components, representing about 90 % of the total, have been isolated and their structures elucidated by spectroscopic methods. Molecular weights were determined by FAB mass spectrometry. The components studied belong to three groups (see Fig.), depending on substituent R2 and each set represents homologous series, differing in substituent R1. The structure proposed previously for primycin was unambiguously proved and assigned as that of the A1 component.

Abstract Primycin, a mixture of macrolide antibiotic agents, on treatment with nucleophiles is transformed in a reversible reaction into water-soluble compounds with the same molecular weights as that of the starting materials. Detailed 2D NMR spectroscopic studies of the separated components revealed that the new compounds, named “pseudo-primycins”, are formed by a unique C35 → C37 ring expansion via translactonization.
