Zur Ultrastruktur der fleckförmigen Hornhautdystrophie (Groenouw II): Untersuchung der parazentralen tiefen Hornhautschichten

Electron microscopic studies proved, that in cases of macular corneal dystrophy (Groenouw II) abnormal deposits of acid mucopolysaccharide are demonstrable in the different layers of the cornea in a polymorphic form. Because of the investigations now performed it can be established, that the opacities of the paracentral, deep corneal layers result from both dispersed vesicular and plaque-like deposits of storage material in the border area of the stroma and Descemets membrane.Electron microscopic studies proved, that in cases of macular corneal dystrophy (Groenouw II) abnormal deposits of acid mucopolysaccharide are demonstrable in the different layers of the cornea in a polymorphic form. Elektronenmikroskopische Untersuchungen haben ergeben, daß in Fällen von fleckförmiger Hornhautdystrophie (Groenouw II) Ablagerungen von sauren Mucopolysacchariden in den verschiedenen Schichten der Hornhaut in vielgestaltiger Form vorkommen können. Auf Grund der jetzt durchgeführten Untersuchungen kann festgestellt werden, daß die massiven Trübungen der parazentralen und peripheren Bereiche in den tiefen Schichten teils durch verstreute vesikuläre Anordnung, vor allem aber durch plaqueartige Ablagerungen von Speicherstoffen im Grenzbereich von Stroma und Descemetscher Membran entstehen.

Der Verlauf der Zonulafasern

Zonular fibers could be shown between the basement membrane of the pigmented epithelium of pars plana and the elastic tissue of Bruchs membrane. A connection of zonular fibrils between the unpigmented epithelium and the basement membrane of pigmented epithelium of the pars plana could not be observed. It is possible to discuss two independent courses of zonular fibers.Zonular fibers could be shown between the basement membrane of the pigmented epithelium of pars plana and the elastic tissue of Bruchs membrane. A connection of zonular fibrils between the unpigmented epithelium and the basement membrane of pigmented epithelium of the pars plana could not be observed. It is possible to discuss two independent courses of zonular fibers. Zonulafasern konnten zwischen der Basalmembran des pigmentierten Epithels der Pars plana und dem darunterliegenden elastischen Gewebe der Bruchschen Membran nachgewiesen werden. Eine Verbindung von Zonulafibrillen zwischen dem unpigmentierten Epithel und der Basalmembran des pigmentierten Epithels der Pars plana konnte nicht beobachtet werden. Man kann daher von zwei getrennt verlaufenden Zonulafasersträngen sprechen.

Mit Hilfe der Rasterelektronenmikroskopie wurden 3 Einstrahlungszonen von Zonulafasern dargestellt. 1. Die Zone, die sich auf den Linsenrand bezieht. 2. Die Zone der Pars plicata corporis ciliaris und 3. die Zone der Pars plana. Die Zone, die sich auf den Linsenrand bezieht. Die Zone der Pars plicata corporis ciliaris und die Zone der Pars plana. Außerdem werden Verbindungen von Zonulafasern mit der Glaskörpergrenzmembran aufgezeigt.

Isolation of different hydroxyproline containing proteins from bovine vitreous body collagen

From pepsin-solubilized vitreous body collagen three different precipitates were collected by differential salt precipitation. These three different protein fractions contain hydroxyproline and show different patterns in poly-acrylamide gel electrophoresis suggesting different collagen types. Aus pepsin-gelöstem Glaskörperkollagen konnten mit Präzipitation aus Salzlösungen verschiedener Molarität drei verschiedene Präzipitate erhalten werden. Diese drei Fraktionen enthalten Hydroxyprolin und zeigen in der Polyacrylamidgel Elektrophorese verschiedene Bandenspektren. Daher wird vermutet, daß es sich bei den drei Präzipitaten um verschiedene Kollagentypen handelt.

Der Angriff verschiedener Proteasen auf isolierte Zonulafasern

Some proteases, i.e. trypsin, α-chymotrypsin, thermolysin, proteinase K, α-amylase, collagenase, and papain were investigated on their effect on isolated zonular fibers. All these enzymes but collagenase were zonulolytic active. An attack on the ground substance of the fibers by substances solving glycosaminoglycans and proteoglycans (hyaluronidase, EDTA, guanidinium chloride, H2O2) showed an increased effect of the enzymes used. These results suggest that the interfibrillar matrix has a protective function on the zonular fibers. Einige Proteasen, wie Trypsin, α-Chymotrypsin, Thermolysin, Proteinase K, α-Amylase, Kollagenase und Papain wurden auf ihre Wirksamkeit auf isolierte Zonulafasern untersucht. Bis auf Kollagenase waren alle diese Enzyme zonulolytisch aktiv. Ein Angriff auf die Grundsubstanz der Fasern durch Substanzen, welche die Glycosaminoglycane und Proteoglycane lösen (Hyaluronidase, EDTA, Guanidiniumchlorid, H2O2), zeigte einen verstärkten Effekt der verwendeten Enzyme. [The course of zonular fibers in the area of pars plicata corporis cilaris pars plana, ora serrata and retina. Eectron microsocpic study].

The course of zonular fibers in the area of pars plicata corporis ciliaris could be shown down to the pigmented epithelial layer and its basement membrane. Single fibrils enter in a connection with the plasma membrane in all investigated parts. Fibrils could be observed in the cytoplasm of the ora serrata. For the other areas (pars plicata corporis ciliaris, pars plana) this can be suggested too. Der Verlauf der Zonulafasern in der Pars plicata corporis ciliaris konnte bis zum pigmentierten Epithel und dessen Basalmembran verfolgt werden. Einzelfibrillen treten in allen untersuchten Abschnitten mit der Zellmembran in Verbindung. Im Cytoplasma der Ora serrata konnten deutlich Fibrillen beobachtet werden. Für die übrigen Abschnitte (Pars plicata corporis ciliaris, Pars plana) kann dies ebenfalls angenommen werden.The course of zonular fibers in the area of pars plicata corporis ciliaris could be shown down to the pigmented epithelial layer and its basement membrane. Die Wirkung von Äthylendiamintetraessigsäure auf die Retina

Electron microscopic investigations show that intravitreal application of EDTA causes destruction of the viteous body and damage in all layers of the retina. Therefore, it is concluded that it is useless to try prevention of metallosis of the eye by EDTA. Die elektronenoptischen Untersuchungen zeigen, daß durch intravitreale Gabe von EDTA Veränderungen des Glaskörpers und Schädigungen aller Schichten der Retina verursacht werden. Daher kann geschlossen werden, daß es nicht sinnvoll ist, mit EDTA die Verhinderung einer Metallose zu versuchen.Electron microscopic investigations show that intravitreal application of EDTA causes destruction of the viteous body and damage in all layers of the retina. [Electronmicroscopic investigation about the course of zonular fiber in the area of ora serrata and the peripheral retina (author's transl)].

Single zonular fibers could be shown in the area of ora serrata and the peripheral parts of retina between the basement membrane of pigmented epithelium and the elastic tissue of Bruchsmembran. The occurrence of fibroblasts in the proximity of zonular fibers was noticeable. Einzelne Zonulafasern konnten im Bereich der Ora serrata und den periphersten Netzhautanteilen zwischen der Basalmembran des pigmentierten Epithels und dem elastischen Gewebe der Bruchschen Membran nachgewiesen werden. Auffällig war das Auftreten von Fibroblasten in nächster Nähe der Zonulafasern.Single zonular fibers could be shown in the area of ora serrata and the peripheral parts of retina between the basement membrane of pigmented epithelium and the elastic tissue of Bruchsmembran.
