Evaluation of extenders and cryoprotectants on motility and morphology of longtooth grouper (Epinephelus bruneus) sperm.

The objective was to evaluate various extenders and cryoprotectants on postthaw motility of longtooth grouper (Epinephelus bruneus) sperm, based on sperm motility ratio (SMR), sperm velocity (SV), and morphological damage after thawing. To evaluate the optimal cryoprotectant for cryopreservation of longtooth grouper sperm, semen was diluted in 0.3 M glucose extender containing one of four cryoprotectants (dimethyl sulfoxide, methanol, ethylene glycol, and glycerol [Gly]) at a final concentration of 10% or 20%, and then frozen in liquid nitrogen vapor (-76 °C for 3 minutes) before storage in liquid nitrogen. Semen diluted in 0.3 M glucose containing 10% Gly had the highest postthaw SMR (57.5 ± 2.5%, mean ± SEM). To identify the optimal extender, semen was diluted in one of two longtooth grouper artificial seminal plasma (LG-ASP; LG-ASP1 and LG-ASP2) extenders containing Gly at concentrations of 0%, 5%, 10%, or 20%, then cryopreserved using the described procedure. Semen diluted with LG-ASP2 extender containing 10% Gly cryoprotectant had the best postthaw SMR (66.3 ± 2.0%) and SV (135.9 ± 4.5 μm/s). Compared with fresh sperm, some structural damage was observed in cryopreserved sperm. We concluded that the combination of LG-ASP2 and 10% Gly (as extender and cryoprotectant, respectively), resulted in high postthaw SMR and SV for cryopreservation of longtooth grouper sperm.

Cryopreservation of Common Korean Bitterling Acheilognathus signifer Sperm

묵납자루(Acheilognathus signifer)는 한국 고유어종으로 잉 어과(Cyprinidae)의 납자루아과(Acheilognathinae)에 속하며, 한강 수계를 포함하여 그 이북지역인 임진강, 대동강, 압록강, 성천 및 회양 등에 분포 하는 것으로 보고되어 있다(Uchida, 1939). 묵납자루의 체색은 암녹색으로 등쪽이 짙고 배쪽은 연 한 노란색을 띠고 있으며, 등지느러미와 뒷지느러미의 기부는 회갈색이지만 중앙부는 노란색의 넓은 띠가 현저하고 가장자 리는 흑갈색을 띠고 있어 칼납자루(Acheilognathus koreensis) 와 함께 담수관상어로서 가치가 매우 높은 어종이다. 그러나 묵 납자루는 포획, 반출 및 유통 등이 야생동물보호법으로 금지되 어 있는 멸종위기 야생동식물 2급으로(MEV, 2012), 일반인이 쉽게 취급할 수 없는 어종이다. 묵납자루의 서식지는 하천 중· 상류 지역에 바닥이 뻘과 모래의 비율이 높고, 하천 변에 수초 가 무성한 정수역을 선호하며, 담수산 이매패류인 작은말조개 (Unio douglasiae sinuolatus)에 산란하는 독특한 산란 특성을 가지고 있다(Beak and Song, 2005a, b). 이로 인해 잉어(Cyprinus carpio) 및 붕어(Carassius carassius)와 같은 일반 담수어류 와 달리 채란·채정이 용이하지 못하다. 또한 채란·채정이 이루 어졌다 하더라도 그 양이 매우 적어 인공종묘생산 기술개발 및 대량생산에 어려움이 있다. 따라서 한국 고유어종의 종 보존 및 담수관상어 시장 활성화를 위해서는 묵납자루의 증식·복원 연 구 및 인공종묘 대량생산 기술개발이 필요하다. 어류 정자의 냉동보존에 관한 연구는 Blaxter (1953)가 청어 (Clupea pallasii) 정자의 냉동보존을 성공적으로 이룬 이후, 유 용 어류의 정자냉동보존에 관한 많은 연구결과가 보고되고 있 다(Chang and Chang, 2002; Billard et al., 2004). 정자의 냉동 보존 기술은 양식 종의 종묘생산에 있어 어미의 방란·방정 시기 가 불일치하거나, 성비가 고르지 못할 때 발생하는 채란·채정의

Survival Rates with Time Course of Frozen-thawed Pacific Oyster Larvae in Indoor Rearing System.

Post-thawed larval rearing in Pacific oyster Crassostrea gigas was performed to investigate the survival rate with time course in three kinds of larvae cryopreserved. The highest survival rate and larval activity index (LAI) of post-thawed larvae were obtained from the permeation in 0.2 M sucrose and 2.0 M ethylene glycol (EG) at -1°C/min in freezing speed showing the survival rates just after thawing of 63.8% in trochophore, 84.1% in D-shaped veliger and 56.3% in early umbo veliger. In post-thawed larval rearing with food supply, the larvae lasted their lives until 24 hours in trochophore, 75 hours in D-shaped veliger and 57 hours in early umbo veliger. The results suggested that each larval stage post-thawed revealed no more further development to subsequent respective stage.
