Petunia has a monofactorial gametophytic self-incompatibility system which is characterized by a mechanism for pollen-tube rejection after incompatible pollination about half-way between stigma and ovule. From in vitro experiments it is known that pollen tube extension requires de novo synthesis of proteins on 80 S ribosomes. The synthesis of these structural proteins has no need of new mRNA formation. The situation in the style is quite different: both RNA and protein synthesis are positively affected by pollination. There is a qualitative difference in synthesis between self- and cross-pollinated styles. These findings suggest that inhibition or rejection of pollen tubes is ‘the normal reaction’, whereas unaffected penetration is based on the synthesis of special enzymes after compatible pollination which break down the existing incompatibility barrier.

Abstract Phytic acid has been detected in the anthers of young flower buds of Petunia hybrida , the amount increasing slowly as the flower develops until anther dehydration, when there was a more rapid increase in phytic acid content. In mature pollen, the phytic acid content was found to be 2.0 % by weight, of which 90 % was water soluble, while free myo -inositol was a relatively low 0.06 % by weight. Breakdown of phytic acid was initiated soon after pollen germination began, and its degradation products, myo -inositol and inorganic phosphate, were rapidly mobilized for phospholipid and pectin biosynthesis. Both are in high demand during pollen tube elongation. Utilization of myo -[2- 3 H]inositol for phospholipid biosynthesis was about five times that for pectin synthesis during the first few hours of pollen germination. The label in the phospholipid was identified as the myo -inositol moiety of phosphaltidylinositol, while the pectin material contained predominantly labelled arabinose, with smaller amounts of label in galacturonic acid, glucose and xylose. A chase experiment showed that the myo -inositol moiety of phosphatidylinositol was subject to a relatively rapid turnover, while the label in pectin was not. Labelling germinating pollen with [ 32 P]orthophosphate gave label in phosphatidic acid, phosphatidylinositol, phosphatidylethanolamine and phosphatidylcholine of the phospholipids. Phosphatidylinositol contained 30 % of this label initially, a proportion which declined to 10 % over longer periods of germination.

Abstract Homogenates from pollen of Lilium longiflorum (cv. Arai) contain phosphatidylinositol phospholipase activity, which increased during germination. Analysis of radioactive products resulting from incubation of phosphatidylinositol, labelled in the myo -inositol, phosphate or glycerol moiety, showed that cleavage had only taken place between glycerol and phosphate. The activity is therefore of a phospholipase C-type (EC 3.1.4.10). It was stimulated by several divalent cations; Ca 2+ was the most effective. Zn 2+ , Cu 2+ and EDTA are inhibitory. The activity was found in both the cytosol and particulate fractions.

Abstract A considerable incorporation of [ 32 P]orthophosphate into phospholipids was observed during germination of pollen of Lilium longiflorum , particularly during the period of most rapid pollen tube elongation. The major phospholipids were phosphatidylethanolamine, phosphatidylcholine, phosphatidylinositol, phosphatidylglycerol and phosphatidic acid. The amount of lipid-bound phosphorus (0.05 μmol/mg pollen) did not change significantly during germination, suggesting that the incorporation was mainly due to turnover. myo -[2- 3 H]Inositol was incorporated into phospholipids as well as into pectic cell wall polysaccharides. The phospholipids labelled with both 3 H and 32 P were found to be phosphatidylinositol (90% of 3 H-label) and phosphatidylinositol monophosphate. The identity of the latter was established by thin layer chromatography, by anion exchange column chromatography (with immobilized neomycin sulphate as the stationary phase), and by the formation of glycerylphosphoryl inositol phosphate and inositol bisphosphate during alkaline hydrolysis. Pulse-chase experiments suggested that about 40% of the myo -inositol moieties in phosphatidylinositol is exchanged during the 4 hr-period of most rapid pollen tube extension. This high figure may be a consequence of membrane flow known to take place during pollen tube elongation, proceeding at the high rate of approximately 1 mm/4 hr. Phosphatidylinositol phospholipase C activity, which could play a role in this turnover, was detected in pollen tube homogenates. Phospholipase activities towards the other major phospholipids were also observed.

The activation of the ovary after pollination occurs before fertilization takes place and starts with the carpels and the placenta.
