Ecological, biological and social dimensions of dengue vector breeding in five urban settings of Latin America: a multi-country study

BackgroundDengue is an increasingly important public health problem in most Latin American countries and more cost-effective ways of reducing dengue vector densities to prevent transmission are in demand by vector control programs. This multi-centre study attempted to identify key factors associated with vector breeding and development as a basis for improving targeted intervention strategies.MethodsIn each of 5 participant cities in Mexico, Colombia, Ecuador, Brazil and Uruguay, 20 clusters were randomly selected by grid sampling to incorporate 100 contiguous households, non-residential private buildings (businesses) and public spaces. Standardized household surveys, cluster background surveys and entomological surveys specifically targeted to obtain pupal indices for Aedes aegypti, were conducted in the dry and wet seasons.ResultsThe study clusters included mainly urban low-middle class populations with satisfactory infrastructure and –except for Uruguay- favourable climatic conditions for dengue vector development. Household knowledge about dengue and “dengue mosquitoes” was widespread, mainly through mass media, but there was less awareness around interventions to reduce vector densities. Vector production (measured through pupal indices) was favoured when water containers were outdoor, uncovered, unused (even in Colombia and Ecuador where the large tanks used for household water storage and washing were predominantly productive) and –particularly during the dry season- rainwater filled. Larval infestation did not reflect productive container types. All productive container types, including those important in the dry season, were identified by pupal surveys executed during the rainy season.ConclusionsA number of findings are relevant for improving vector control: 1) there is a need for complementing larval surveys with occasional pupal surveys (to be conducted during the wet season) for identifying and subsequently targeting productive container types; 2) the need to raise public awareness about useful and effective interventions in productive container types specific to their area; and 3) the motivation for control services that-according to this and similar studies in Asia- dedicated, targeted vector management can make a difference in terms of reducing vector abundance.

BITING ACTIVITY AND BREEDING SITES OF ANOPHELES SPECIES IN THE MUNICIPALITY VILLAVICENCIO, META, COLOMBIA

ABSTRACT Villavicencio, the capital city of the Department of Meta, Colombia, is at high risk for the urbanization of malaria because of the regions ecological conditions, as well as the permanent presence of infected human populations arriving from rural areas. From August to November 2002 and in April 2003, anopheline collections were undertaken in the area. Isofamilies were obtained from 331 wild females, which were then recorded according to their abundance as follows: Anopheles marajoara, Anopheles rangeli, Anopheles braziliensis, Anopheles darlingi, and Anopheles apicimacula. Anopoheles darlingi showed the highest biting activity (3.0) between 1800 and 1900 h. Forty-five breeding places were sampled, 64% of which were fish ponds, 6.7% flooded meadows, and 6.7% drainpipes, with these being the most representative locations. All sampled breeding sites were positive for anophelines. Anopheles marajoara could play an important role as an auxiliary vector in Villavicencios urban area. Control measures should be aimed at weeding the marginal areas around fish ponds and at evaluating the use of impregnated bed-nets.

Susceptibilidad de Aedes aegypti a DDT, deltametrina y lambdacialotrina en Colombia

OBJETIVOS: Evaluar el estado de susceptibilidad a insecticidas piretroides deltametrina y lambdacialotrina y al organoclorado DDT, e identificar los mecanismos bioquimicos asociados con resistencia en 13 poblaciones naturales de Aedes aegypti recolectadas en localidades de Colombia donde el dengue es un grave problema de salud publica. METODOS: Se recolectaron y criaron en condiciones controladas formas inmaduras de diferentes criaderos naturales del vector para cada localidad. Con la generacion F2 se realizaron bioensayos utilizando las metodologias OMS 1981 (papeles impregnados) y CDC 1998 (botellas impregnadas). En las poblaciones con mortalidades compatibles con disminucion de la susceptibilidad, se midieron los niveles de esterasas no especificas (ENE), oxidasas de funcion mixta (OFM) y acetilcolinesterasa modificada (ACEM) mediante pruebas colorimetricas. RESULTADOS: Todas las poblaciones del mosquito evaluadas evidenciaron resistencia al organoclorado DDT. En cuanto a los piretroides, se encontro resistencia generalizada a lambdacialotrina pero no a deltametrina. Los mecanismos bioquimicos de resistencia evaluados permitieron encontrar 7 de 11 poblaciones con ENE elevadas y una poblacion con OFM incrementadas. CONCLUSIONES: Se descarta la resistencia cruzada de tipo fisiologico entre el DDT y lambdacialotrina en las poblaciones de A. aegypti evaluadas. La resistencia fisiologica a lambdacialotrina parece asociarse con el incremento de las ENE. El comportamiento diferencial en los niveles de susceptibilidad y los valores enzimaticos entre poblaciones se asociaron con la variabilidad genetica y presion de seleccion quimica a nivel local.

Effectiveness and feasibility of long-lasting insecticide-treated curtains and water container covers for dengue vector control in Colombia : a cluster randomised trial

Background Long-lasting insecticide-treated net (LLIN) window and door curtains alone or in combination with LLIN water container covers were analysed regarding effectiveness in reducing dengue vector density, and feasibility of the intervention. Methods A cluster randomised trial was conducted in an urban area of Colombia comparing 10 randomly selected control and 10 intervention clusters. In control clusters, routine vector control activities were performed. The intervention delivered first, LLIN curtains (from July to August 2013) and secondly, water container covers (from October to March 2014). Cross-sectional entomological surveys were carried out at baseline (February 2013 to June 2013), 9 weeks after the first intervention (August to October 2013), and 4–6 weeks after the second intervention (March to April 2014). Results Curtains were installed in 922 households and water container covers in 303 households. The Breteau index (BI) fell from 14 to 6 in the intervention group and from 8 to 5 in the control group. The additional intervention with LLIN covers for water containers showed a significant reduction in pupae per person index (PPI) (p=0.01). In the intervention group, the PPI index showed a clear decline of 71% compared with 25% in the control group. Costs were high but options for cost savings were identified. Conclusions Short term impact evaluation indicates that the intervention package can reduce dengue vector density but sustained effect will depend on multiple factors.

Genetic structure of Anopheles (Nyssorhynchus) marajoara (Diptera: Culicidae) in Colombia.

Five Anopheles marajoara Galvão and Damasceno populations, representing diverse ecological conditions, were sampled throughout Colombia and analyzed using nine hypervariable DNA microsatellite loci. The overall genetic diversity (H = 0.58) was lower than that determined for some Brazilian populations using the same markers. The Caquetá population (Colombia) had the lowest gene diversity (H = 0.48), and it was the only population at Hardy-Weinberg equilibrium. Hardy-Weinberg disequilibrium in the remaining four populations was probably caused by the Wahlund effect. The assignment analyses showed two incompletely isolated gene pools separated by the Eastern Andean cordillera. However, other possible geographical barriers (rivers and other mountains) did not play any role in the moderate genetic heterogeneity found among these populations (F(ST) = 0.069). These results are noteworthy, because this species is a putative malaria vector in Colombia.

ESTABLISHMENT AND CHARACTERIZATION OF A CELL LINE FROM THE MOSQUITO ANOPHELES ALBIMANUS (DIPTERA : CULICIDAE)

A new cell line designated LSB-AA695BB, was established from embryos of the mosquito Anopheles albimanus. The primary culture was initiated in April, 1995, and the first passage was made 48 days later. Serial subcultures of the cells have been carried through 90 passage from Abril 1995 to February 1996. The cells were grown at 28 degrees C in MK/VP12 medium, supplemented with 20% fetal bovine serum: the pH tolerance ranged between 6.8 to 7.0. The cells have also been adapted to MM/VP12 medium under the same pH, temperature and serum concentration. The majority of the cells were a fibroblast-type. Isozyme characterization showed a pattern similar to that of An. albimanus pupae and adults but distinct from Ae. taeniorhynchus and Ae. albopictus (C6/36) mosquito cell lines. The culture was shown to be free of mycoplasma, bacteria and fungi. Microsporidia contamination of transovarial transmission was controlled with 6.0 micrograms/ml of albendazole.

Untangling the transmission dynamics of primary and secondary vectors of Trypanosoma cruzi in Colombia: parasite infection, feeding sources and discrete typing units

BackgroundTrypanosoma cruzi is the causative agent of Chagas disease. Due to its genetic diversity has been classified into six Discrete Typing Units (DTUs) in association with transmission cycles. In Colombia, natural T. cruzi infection has been detected in 15 triatomine species. There is scarce information regarding the infection rates, DTUs and feeding preferences of secondary vectors. Therefore, the aim of this study was to determine T. cruzi infection rates, parasite DTU, ecotopes, insect stages, geographical location and bug feeding preferences across six different triatomine species.MethodsA total of 245 insects were collected in seven departments of Colombia. We conducted molecular detection and genotyping of T. cruzi with subsequent identification of food sources. The frequency of infection, DTUs, TcI genotypes and feeding sources were plotted across the six species studied. A logistic regression model risk was estimated with insects positive for T. cruzi according to demographic and eco-epidemiological characteristics.ResultsWe collected 85 specimens of Panstrongylus geniculatus, 77 Rhodnius prolixus, 37 R. pallescens, 34 Triatoma maculata, 8 R. pictipes and 4 T. dimidiata. The overall T. cruzi infection rate was 61.2% and presented statistical associations with the departments Meta (OR: 2.65; 95% CI: 1.69–4.17) and Guajira (OR: 2.13; 95% CI: 1.16–3.94); peridomestic ecotope (OR: 2.52: 95% CI: 1.62–3.93); the vector species P. geniculatus (OR: 2.40; 95% CI: 1.51–3.82) and T. maculata (OR: 2.09; 95% CI: 1.02–4.29); females (OR: 2.05; 95% CI: 1.39–3.04) and feeding on opossum (OR: 3.15; 95% CI: 1.85–11.69) and human blood (OR: 1.55; 95% CI: 1.07–2.24). Regarding the DTUs, we observed TcI (67.3%), TcII (6.7%), TcIII (8.7%), TcIV (4.0%) and TcV (6.0%). Across the samples typed as TcI, we detected TcIDom (19%) and sylvatic TcI (75%). The frequencies of feeding sources were 59.4% (human blood); 11.2% (hen); 9.6% (bat); 5.6% (opossum); 5.1% (mouse); 4.1% (dog); 3.0% (rodent); 1.0% (armadillo); and 1.0% (cow).ConclusionsNew scenarios of T. cruzi transmission caused by secondary and sylvatic vectors are considered. The findings of sylvatic DTUs from bugs collected in domestic and peridomestic ecotopes confirms the emerging transmission scenarios in Colombia.

Preliminary Biological Studies on Larvae and Adult Anopheles Mosquitoes (Diptera: Culicidae) in Miraflores, a Malaria Endemic Locality in Guaviare Department, Amazonian Colombia

ABSTRACT In the malaria endemic municipality of Miraflores in southeastern Amazonian Colombia, several aspects of the biology of local Anopheles species were investigated to supplement the limited entomological surveillance information available and to provide baseline data for malaria prevention and vector control. Anopheles darlingi Root, 1926 was the most abundant species (95.6%), followed by Anopheles braziliensis (Chagas) (3.6%) and Anopheles oswaldoi s.l. (Peryassu) (0.7%). During the dry season, exophagic activity was prevalent only between 1800–2100 hours; after this (2100-0600 hours) only endophagy was encountered. In contrast, during the rainy season, both endophagy and exophagy occurred throughout the collection period. The human biting rate for An. darlingi was 8.6. This species was positive for Plasmodium vivax VK210 with a sporozoite rate = 0.13 (1/788). Breeding sites corresponded to stream (n = 7), flooded excavations (n = 4), flooded forest (n = 1), wetlands (n = 2), and an abandoned water reservoir (n = 1). An. darlingi predominated in these sites in both seasons. Based on these data, An. darlingi is the main local malaria vector, and we recommend that local prevention and control efforts focus on strengthening entomological surveillance to determine potential changes of species biting behavior and time to reduce human-vector interactions.

Malaria vectors in San José del Guaviare, Orinoquia, Colombia.

Abstract This study was conducted to determine Anopheles species composition and their natural infectivity by human Plasmodium in 2 localities with the highest malaria transmission in San Jose del Guaviare, Guaviare, Colombia. A total of 1,009 Anopheles mosquitoes were collected using human landing catches during 8 months in 2010. Anopheles darlingi was the most abundant (83.2%) followed by An. albitarsis s.l. (8.6%), Anopheles braziliensis (3.8%), An. oswaldoi s.l. (1%), and An. rangeli (0.3%). Anopheles darlingi showed the highest human biting rate, and it was found naturally infected with Plasmodium vivax VK210 (0.119%) using enzyme-linked immunosorbent assays. All species were collected biting both indoors and outdoors. Anopheles darlingi showed biting activity overnight with an indoor peak between 1200–0100 h. Therefore, we recommend that malaria prevention strategies focus on 1) insecticide-treated nets to reduce human–vector contact when people are most exposed and unprotected; 2) accurate diagnoses; 3) adequate treatment for patients; 4) more timely epidemiological notification; and 5) improved entomological surveillance.

Anopheles (Díptera: Culicidae) vectors of malaria in Puerto Carreño municipality, Vichada, Colombia

INTRODUCTION The study of the biological aspects of Anopheles spp., strengthens the entomological surveillance. OBJECTIVE To determine biological aspects and behavior of adult Anopheles mosquitoes in the urban area of Puerto Carreño municipality, Vichada, Colombia. MATERIALS AND METHODS Wild anophelines were collected landing on humans both indoors and outdoors between 18:00h and 06:00h for 50 min/h during two consecutive nights/month for eight months in the urban area of Puerto Carreño. The biting rate activity, the natural infection by Plasmodium falciparum and P. vivax VK247 and VK210 using ELISA, and the annual entomological inoculation rate were determined for each species. The members of the Albitarsis complex were determined by amplificacion of the white gene by polymerase chain reaction. RESULTS In order of abundance the species found were An. darlingi (n=1,166), An. marajoara sensu stricto (n=152), An. braziliensis (n=59), An. albitarsis F (n=25), An. albitarsis sensu lato (n=16), An. argyritarsis (n=3) and An. oswaldoi sensu lato (n=2). An. darlingi showed two activity peaks between 21:00 to 22:00 and 05:00 to 06:00 hours outdoors and between 21:00 to 22:00 and 04:00 to 05:00 indoors. Natural infection of this species was found with P. vivax VK210 and its annual entomological inoculation rate was 2. Natural infection of An marajoara sensu stricto with P. falciparum was found, with an annual entomological inoculation rate of 5 and a peak biting activity between 18:00 to 19:00 hrs both indoors and outdoors. CONCLUSION Transmission of malaria in the urban area of Puerto Carreño, Vichada, can occur by An. darlingi and An. marajoara s.s.
