Proximate Content of “Klekap” (Microphytobenthos and Their Associated Meiofauna) from Milk-Fish Pond

Microphytobentos and their associated mieofauna (klekap) have important role in milkfish pond ecosystem expecially traditional fish pond. Microphytobenthos and their associated meiofauna not only consists of unicellular microalgae from class Bacillariophyceae, Cyanophyceae, Chlorophyceae but also a lot of meiofauna such as Nematoda, Annelida, Mollusc, Arthropoda, etc. Microphytobenthos and their associated meiofauna inhabit in the upper most surface sediment layer. They also have important role in the primary and secondary productivity. This research has purpose to investigate the composition of proximate in the microphytobenthos and their associated meiofauna. Microphytobenthos and their associated meiofauna were taken from three milkfish ponds in Pati area, Central of Jawa, Indonesia. Samples were taken directly by hand core from 3 stations with 3 replications. The research result showed that the value of protein content had range 6.9±0.33 - 7.73±0.37 %-dw, the value of carbohydrate content had range 10.44±1.2 - 12.59±0.15 %-dw, the value of lipid content had range 0.86±0.07 - 0.96±0.07 %-dw. It concluded that the third station has higher value of protein, lipid and carbohydrate compared than the first and the second station.

Magelang duck is a wild type of local duck from Indonesia. The advantagesof Magelangduckcompare tootherlocalduck from Indonesiaareabilityto livein the highlandsandlowlands and high production of egg and meat. Geneticcharacterization of Magelangduck still not available until now.The aim of the research is selectprimers forampliflying COIgeneof mitochondrialDNAof MagelangduckusingLCO-HCO, bird-f1 -HCO, andbcl-bch primers.The research methodwas DNAisolationfrom Magelangduck. Followed by, selection of primer in silico to find homologywithin COIsequenceusing ClustalX , Genedoc , and FastPCR programs. Amplification of COIgenewas performedusing PCRwith all primerpairs. Result showed partial homology with all primer in COI sequence. TheamplificationusingtheLCO-HCO primer producedxa0 primerdimer .Primerbirdf1-HCOand bch-bcl primers showed no amplification. Kloning Gen pcbC dari Penicillium chrysogenum ke dalam Plasmid pPICZA untuk Pengembangan Produksi Penisilin G

Availability of drugs in Indonesia is still limited by the high prices of drugs due to on the imported raw materials that reaches 95%. Developing antibiotic raw materials can be achieved by increasing of penicillin G production, which is the raw material for the formation of semisynthetic penicillin derivatives through the production of 6-aminopenisillanic acid (6-APA). One of the important enzyme in the penicillin G biosynthesis is Isopenisilin N Synthase (IPNS) that encodes by pcb C gene on Penicillium chrysogenum . This study aimed to obtain a recombinant of pcb C gene fragments that is inserted into pPICZA plasmid. Amplification of pcb C gene used pcbC-F and pcbC-R primers. The pcbC gene fragment was inserted into pPICZA vector and then transformed into TOP 10 F’. The results showed that the recombinant of the pcbC gene fragment from P. chrysogenum has been obtained. Analysis of DNA sequences using the BLAST program showed that the pcbC gene fragment has high homology (99%) with the pcb C gene from P. chrysogenum Wisconsin 54-1255 and P. chrysogenum AS-P-78 which encodes IPNS Keywords: pcbC Gene, Penicillium chrysogenum, cloning, penicillin G
