Influence of varied [Ca2+]o and [Na+]o on slow responses of newborn and adult rat hearts

The electrophysiological properties of slow responses of rat hearts were investigated in newborn (two-day-old) and adult (few-month-old) rats under conditions of high [K+]0 (25 mM) with isoproterenol (1 X 10(-7)M) using the standard techniques of intracellular microelectrode recording. The potentials of slow responses were measured in varied [Ca2+]0: in adults the peak potential varied linearly with a slope of about 31 mV for a 10-fold change in [Ca2+]0, while in newborns it varied, but not linearly, and showed a positive peak potential in low [Ca2+]0 (0.5 mM). In a low [Na+]0 solution (55% [Na+]0), with constant [Ca2+]0 (2 mM), the peak potential was reduced in newborn hearts, while it was little affected in adult hearts. These results suggest that the peak potential of the slow response depends mainly on [Ca2+]0 in adult rat hearts, while it depends on both [Ca2+]0 and [Na+]0 in newborns.

Training effect on the muscle fibre types in race horses.

Changes in the composition of the muscle fibre types by training of race horses were observed. The glycolytic muscle ratio, expressed as percentage of glycolytic muscle fibres against total number of muscle fibres, was increased with age and decreased by training. It is well known that the spinalization of skeletal muscle contraction is activated by training. Therefore, it is suggested that the training effect on skeletal muscles in the race horse is manifested as an increased tonic contractive activity.

Cardiopulmonary Function During Six Weeks Exposure to Simulated High Altitude in Growing Rats

慢性的な低酸素暴露が,成長期ラットの心肺機能に及ぼす影響を明らかににるために,コントロールチャンバーを用いて実験を行なった.5週令のWistar系雄ラット35匹を7群次分けた.5週令で観察する群をC0,標高2,500m相当の低圧低酸素環境に5週令から暴露し,8および11週令で観察する群をT3,T6,11週令まで暴露したのち海面相当の常圧常酸素環境にもどし,22週令で観察する群をT17とし,それぞれの群に対して海面相当で飼育にる対照群C3,C6,C17,を設けた.温度,湿度,および二酸化炭素濃度はそれぞれ常に25°C,60%,および500~1,500ppmに制御した.各群に対して,形態的,心電図的,血液学的検査を常圧常酸素条件下で実施した.暴露群では対照群と比べて,成長にともなう体重の増加が抑圧され,QT間隔は延長し,心臓重量も大きい傾向にあったが,暴露による副腎重量の増大は,常圧常酸素環境へ戻すことにより回復した.赤血球数,Hb濃度,Hct値,および右心室重量はT3群で大きく増加したが,その後の増加は抑圧された.T6群では,平均電気軸のバラツキが大きくなり,肺重量体重比ならびにMCV,MCHが大きくなった.高地環境下でみられる心肺系の典型的な変化が低酸素暴露のみによって生じたことから,心肺機能にとって酸素分圧の低下が多くの高地環境要因の中で最も重要な作用因子であり,高地環境への適応過程として生理状態の安定に向け段階的な変化が生ずる可能性が示唆された.

Cardiopulmonary Function of the Rat During Exposure to Simulated High Altitude

The animals transported to or born in highlands are influenced by the high altitude environment, which consists of many biometeorological factors such as barometric pressure (PB), partial pressures of atmospheric constituents, ambient temperature, humidity, solar radiation, etc.. It has been assumed that among these many environmental factors partial pressure of oxygen (PO2) plays the most important role upon the cardiopulmonary function of the animals1,2). In natural environment, however, an animal is to be exposed to hypobaric as well as hypoxic condition, as PO2 decreases together with PB proportionately to an increase in altitude. Therefore, in the present investigation, hypobaric and/or hypoxic conditions were produced by using the control chamber under the constant state of other environmental conditions in order to discriminate the effects of PB and PO2 on the cardiopulmonary function of the rat.

Electrocardiographical Studies on Cardiac Arrest in the Dog

雑種成犬16頭において, ジギタリス急性中毒, 電気ショック, 高温負荷, 低温負荷, 呼吸停止, アセチルコリン投与などをそれぞれ負荷条件として, 実験的に心停止を誘発した. その時点において, ただちに通常用いられている蘇生処置を試みた. それぞれの一連の経過をA-B誘導心電図によって追跡した結果, 蘇生し得たものと, 死の転帰をとったものは, つぎのとおり類型できるものと考えられた. 1. 期外収縮から, 心房または心室の粗動または細動に移行するもの, および突発的に細動が出現する経過をたどって心停止に至った場合は, 蘇生の可能性が低い. この場合の異所性刺激生成の発現機序は, 第二次以下のぺースメーカーの興奮水準が, 第一次ペースメーカーの興奮水準を凌駕するものと推測された. 2. 徐脈が次第に進行するもの, および徐脈から房室ブロックを経て心停止に至る, いわゆる伝導異常による不整脈を経て心停止に至った場合には, 蘇生の可能性が高い. このような経過を示すものの中にも, 異所性刺激生成による不整脈を経るものもある. この場合の異所性刺激生発現機序は, 上述のものと異なり, 第一次ぺースメーカーの興奮水準が著しく低下したものと推測された.

Acclimatization of Cardio-respiratory Function of Mice to High Altitude Environment

高海抜環境に順応したマウスを平地へ回収したのち,その生理状態がどのように変化していくかについて,血液検査と呼吸ガス代謝試験により追跡した.平地で生産したICR-JCL系マウス5腹58匹を3週令で離乳し,対照群(平地飼育)20匹(雌雄各10匹),高海抜環境曝露群24匹(雌雄各12匹)を無作為抽出した.曝露群は3週令から10週令までの7週間を海抜1,400mの高所で飼育し,11週令時に,現地で観察を行なったのちに平地へ回収し,13週令および16週令時に,対照群とともに観察をくりかえした.高海抜環境下ではRBCの増数,MCVの減少,酸素消費量とRQの増加が認められ,増体量は減少する.曝露群を平地へ回収後,16週令時の観察結果では,対照群に比べMCVが増加,酸素消費量が減少し,RQの値は小さくなるが,増体量は逆に増加する.これまでにえられた一連の実験成績からみれば,高海抜環境に順応した結果,曝露群では,酸素をより有効に利用できるような生理状態がととのえられたと考えられる.これらの結果にもとずいて,高海抜環境に対する生体の順応性を利用した家畜管理技術開発の可能性について考察した.
