Comparison of taurine biosynthesis ability between juveniles of Japanese flounder and common carp

Summary.This study was conducted to investigate taurine deficiency and the ability of taurine biosynthesis in both juvenile Japanese flounder (JF) and juvenile common carp (CC) in vivo using low taurine level diets. Three different taurine level diets were prepared by the supplementation of taurine to the basal composition (JF – 0, 0.5 and 1.5% in JF; CC – 0, 1, 3% in CC). The final average body weight and feed efficiency of JF fed the JF – 1.5% was significantly higher than those of fish fed on the JF – 0%. On the other hand, no significant difference was observed in CC fed with CC – 0, 1, and 3% diets. The taurine retention rate was negative in the case of JF-fed with the taurine-free supplement (JF – 0%). On the other hand, the taurine retention rate was about 280% in the case of CC-fed with the taurine-free supplement (CC – 0%). These findings indicate that while taurine is essential for growth of JF, it is not essential for the growth of CC.

Optimum dietary taurine level in casein-based diet for juvenile red sea bream Pagrus major

This study was conducted to investigate the effect of dietary taurine and cholyltaurine (C-tau) on growth and body composition of juvenile red sea Bream Pagrus major. Semi-purified casein-based diets supplemented with 0 (control diet), 0.1, 0.3, 0.5 and 0.7% taurine and 0.5% C-tau were fed to red sea bream (average body weight 4.7g) for 6 weeks at 20°C. The growth and feed efficiency were the lowest in fish fed the control diet. Taurine supplementation improved the growth and feed efficiency of fish dose-dependently, and the taurine requirement was estimated as 0.52% in terms of optimizing growth and 0.48% in terms of optimizing feed efficiency. Taurine content in the whole body and liver increased with the dietary taurine level. Supplemental C-tau at the 0.5% level had limited effects on the growth and no effect on body taurine, biliary bile salt and liver fat contents. From these results it can be inferred that the optimal dietary taurine requirement of juvenile red sea bream is 0.5% on a dry weight basis, and that the supplementation of taurine in the diet not only improves the growth but also increases hepatic lipid levels of red sea bream juveniles.

Effects of taurine levels in broodstock diet on reproductive performance of yellowtail Seriola quinqueradiata

The effect of dietary taurine was investigated on reproductive performance in yellowtail Seriola quinqueradiata. Two-year-old fish of average body weight 6.1kg were fed on diets containing three levels of taurine (T-0, T-0.5 or T-1.0%) for 5 months prior to spawning. For spawning investigations, fish were induced to maturity by human chorionic gonadotrop in injection (600 IU/kg-fish) and artificially inseminated. Oocyte growth improved significantly (P<0.05) with the increase of dietary taurine. The collection of eggs from females reared on the T-0% diet was not successful. The success rate of spawning for females fed on T-0.5 and 1.0% diets was one out of six, and six out of seven, respectively. The taurine levels of the liver and serum in the T-0% diet group were much lower than that in the T-0.5 and T-1.0% diet groups (P<0.05). Fish fed the T-0% diet showed higher contents of serine in the liver and serum. The taurine content of the ovary was not significantly different among the different dietary treatments. These results indicate that taurine has a positive effect on the improvement of spawning performance of yellowtail.

Effect of dietary taurine and lipid contents on conjugated bile acid composition and growth performance of juvenile Japanese flounder Paralichthys olivaceus

The effects of dietary taurine levels and lipid contents on the conjugated bile acid composition and growth performance of juvenile Japanese flounder Paralichthys olivaceus were investigated. Six types of diet (three different levels of taurine at two different levels of lipid) were fed to juveniles (average body weight, 0.04g). Fishmeal that was washed with 70% ethanol to remove taurine was used as the sole protein source. Feeding experiments were carried out at 20°C for 6 weeks. At the end of the experiments, fish were weighed and analyzed for free amino acids in the body and the composition of the conjugated bile acids. The body weight and percent weight gain of the juveniles were improved by the dietary taurine supplementation. The taurine contents of the whole body and tissues increased with the increase of the dietary taurine contents. The conjugated bile acids in the gall bladder consisted of taurocholic acid and taurochenodeoxycholic acid, which increased with the increase of the dietary taurine level. Taurocholic acid accounted for more than 95% of the total conjugated bile acids. This indicates that taurine is the sole amino acid to conjugate bile acid in Japanese flounder.

Nutritional evaluation of rotifers in rearing tanks without water exchange during seed production of amberjack Seriola dumerili

We evaluated the nutrient contents of rotifers sampled from larval-rearing tanks (tank rotifers) without water exchange during the seed production of amberjack Seriola dumerili at three facilities (Kamiura, Kagoshima, and Miyazaki) and compared them with the nutrient contents of freshly enriched rotifers. Compared to the enriched rotifers, the lipid contents, especially neutral lipids and proportion of 22:6n-3, tended to decrease in the tank rotifers. These trends were clearer at Miyazaki where the tank rotifers were sampled before daily supplementation of microalgae (Nannochloropsis). Crude protein content of the tank rotifers did not decrease markedly although the proportion of lysine tended to decrease. Vitamin C and E contents of the tank rotifers decreased significantly only at Miyazaki. Calcium content of the tank rotifers increased at Kamiura and Miyazaki, and the increases in iron and manganese contents of the tank rotifers at Miyazaki and zinc content at Kagoshima were pronounced. These results suggest that the nutritional value of rotifers in larval-rearing tanks without water exchange can be maintained by appropriate supplementation of microalgae. The effect of certain minerals that became high in tank rotifers on subsequent larval development requires further investigation.

Nutritional evaluation of live food organisms and commercial dry feeds used for seed production of amberjack Seriola dumerili

To improve the nutritional quality of live foods and dry feeds ordinarily used for the seed production of amberjack Seriola dumerili, the nutrient contents of rotifers, Artemia nauplii and commercial feeds used in two larval production stations were evaluated. For comparison of the nutrient contents, artificially produced larvae, wild-caught juveniles and wild zooplankton samples were also analyzed. The proportions of 22∶6n-3 in the polar lipid of the cultured larvae increased by feeding the dry feeds. The taurine contents of the cultured larvae reflected the contents of their foods (rotifers<dry feed<Artemia nauplii). The taurine content and the proportion of 22∶6n-3 in Acartia spp. were higher than in foods fed to the larvae. These parameters in the wild juveniles were higher than the cultured ones. The A/E ratios [(each essential amino acid/total essential amino acids)×1000] of the total amino acids of the live foods and dry feeds were similar to those of the cultured larvae, except for the lower ratios of histidine, arginine, threonine and lysine in the live foods. The mucosal folds of the intestine of the cultured larvae did not show typical signs of dietary phospholipid deficiency. These results suggest that requirements of nutrients such as 22∶6n-3 and taurine should be determined for mass production of amberjack seeds.

Effects of biotechnologically processed soybean meals in a nonfishmeal diet on growth performance, bile acid status, and morphological condition of the distal intestine and liver of rainbow trout Oncorhynchus mykiss

This study evaluated four commercially available biotechnologically processed soybean meals (SBMs) that were included as the primary protein sources in a nonfishmeal diet for juvenile rainbow trout Oncorhynchus mykiss. The processed SBMs were Afcep SBM fermented with mixed bacteria predominantly Bacillus spp., DaBomb SBM fermented with Lactobacillus acidophilus, PepSoyGen SBM fermented with Aspergillus oryzae and Bacillus subtilis, and HP300 SBM treated enzymatically. Fishmeal (FM) and common heat-treated and hulled SBM were used in the reference diets. Sodium dodecyl sulfate (SDS) polyacrylamide gel electrophoresis (PAGE) analysis revealed that the protein components of Afcep SBM appeared to have relatively small molecular sizes compared with the other SBM products. After a 10-week feeding trial, growth of fish fed the common SBM diet was depressed, and abnormal liver and distal intestinal morphologies and bile acid characteristics were observed. The Afcep SBM diet showed approximated growth performance and comparable physiological conditions relative to the FM diet, while only limited improvements were observed in fish fed the other processed SBMs. These findings suggest that Afcep SBM, with its smaller molecular mass proteins, is the most promising processed SBM in nonfishmeal diets for rainbow trout.

Postprandial response and tissue distribution of the bile acid synthesis-related genes, cyp7a1, cyp8b1 and shp, in rainbow trout Oncorhynchus mykiss

In mammals, cholesterol 7α-hydroxylase (CYP7A1) and sterol 12α-hydroxylase (CYP8B1) are rate-limiting enzymes in bile acid synthesis. In addition, a small heterodimer partner (SHP) is also known to inhibit bile acid synthesis via the suppression of CYP7A1 and CYP8B1 expression. However, little information is currently available regarding primary structure of the genes involved in bile acid synthesis in fish. We therefore cloned cyp7a1, cyp8b1 and shp genes from rainbow trout and obtained cDNAs encoding two isoforms each of Cyp7a1 (-1 and -2), Cyp8b1 (-1 and -2) and Shp (-1 and -2). Both cyp7a1-1 and -2 encoded proteins of 512 amino acids. Trout cyp7a1-1 was expressed not only primarily in the kidney, pyloric caecum and mid-gut, but also weakly in the liver, eye, gill and ovary. cyp7a1-2 was highly expressed in the liver, pyloric caecum and mid-gut. cyp8b1-1 and -2, which encoded proteins of 512 and 509 amino acids, respectively, were principally expressed in the liver. Both shp-1 and -2, which encoded proteins of 288 and 290 amino acids, respectively, were strongly expressed in the liver, but shp-2 was also highly expressed in the gallbladder and digestive tract. The temporal changes in the expression of cyp7a1-1/-2, cyp8b1-1/-2 and shp-1/-2 in the liver were assessed after consumption of a single meal. Expression of cyp7a1-1/-2 and cyp8b1-1/-2 increased within 3h post feeding (hpf) when the stomach was still approximately 84% full and the gallbladder was almost completely empty. Although the expression of shp-1 did not change after feeding, the expression pattern of shp-2 was inversely related to the expression patterns of cyp7a1-1/-2 and cyp8b1-1/-2. Specifically, shp-2 expression decreased until 3 hpf before returning to initial levels at 24 hpf. These findings suggest that Cyp7a1s/8b1s and Shp-2 function antagonistically in bile acid synthesis in rainbow trout.
