Spread of colorectal cancer micrometastases in regional lymph nodes by reverse transcriptase-polymerase chain reactions for carcinoembryonic antigen and cytokeratin 20

Background and Objectives: Lymph node metastasis is known as a significant predictor of prognosis in colorectal cancer patients. Recently, reverse transcriptase polymerase chain reaction (RT‐PCR) has been applied to detecting micrometastasis. To assess the risk of recurrence and accurately determine the spread of tumor cells, we examined lymph node micrometastases in a series of colorectal cancer patients.

Immunological response of regional lymph nodes after tumor cryosurgery: Experimental study in rats

Seven days after inoculation of metastasizing rat mammary tumor No. 1 into the thigh of 5-week-old female Sprague-Dawley rats, the tumor was treated cryosurgically by two-cycle freezing and by contact methods at -170 degrees C. Weights of the thymus and the spleen, histological findings of the lumbar lymph nodes, phytohemagglutinin (PHA)-induced blastogenesis of lymphocytes obtained from the lumbar lymph nodes and peripheral blood, and resistance rate to tumor rechallenge were examined 1, 3, 6, 10, and 17 week(s) after cryosurgery, with the following results: Thymus weight gradually decreased by 3 weeks after cryosurgery, while spleen weight increased by 1 week, recovering the preoperative level at 6 weeks. Paracortical hyperplasia of the lumbar lymph nodes markedly increased in 1 week and sinus histiocytosis increased after 3 weeks, both remaining at high values until 10 weeks, while germinal center hyperplasia showed a high value at 3 weeks and thereafter decreased gradually. PHA-induced blastogenesis of the lumbar lymph nodes significantly increased 1 week after cryosurgery and remained at its high value until 10 weeks. PHA-induced blastogenesis of peripheral lymphocytes showed the lowest value at 3 weeks and then significantly increased at 6 weeks. Resistance rate to rechallenge showed the lowest value at 3 weeks, reaching the highest level 10 weeks after cryosurgery. From the above results, it was suggested that anti-tumor immunity (resistance to tumor rechallenge) induced by cryosurgery was at the lowest level at 3 weeks after cryosurgery, and gradually increased starting at 6 weeks.(ABSTRACT TRUNCATED AT 250 WORDS)

VH and VL Gene Complexes Encoding an Anti-Spectrin Antibody are Defined by Nucleotide Sequencing of cDNA from a Hybridoma Generated from Hu-PBL-SCID Mouse

Previously we reported that human imunocompetent cells engrafted into scid mice mount a sustained and vigorous humoral immune response to murine erythrocytes. One of the dominant and consistently observed reactivity pattern of these antibodies in immunoblot analysis is with the alpha and beta isoforms of spectrin. In order to define the human xenoreactive response more completely, a hybridoma was generated (from a hu-PBL-scid mouse) whose antibody reacted with two high molecular weight species 225 to 250 kDa. We report here that this conserved antibody species reacts with both the murine and human erythrocyte proteins and cDNA nucleotide sequence analysis of the light and heavy chain genes encoding this antibody reveals that the light chain variable region gene has been previously observed in association with an autoreactive antibody. In addition to characterizing a conserved human B cell clonotype this is the first report of a human monoclonal antibody being generated from the hu-PBL-scid model using the standard hybridoma technology.

CLINICOPATHOLOGICAL STUDIES ON THE EARLY GASTRIC CANCERS : ESPECIALLY ON THE DEGREE OF THEIR SUBMUCOSAL INVASION

早期胃癌 (192例) のうち粘膜下浸潤 (sm) 胃癌 (68例) の粘膜下層 (sm層) への浸潤程度を新たにsm1, sm2, sm3の3段階に分類し, 早期胃癌の臨床病理学的検討を行い, 以下の結果を得た.(1) sm層への浸潤程度が強くなるほどリンパ節転移率, 脈管侵襲陽性率が高率となった.(2) sm2~3と判定された症例は組織型ではpor, tub2に, 肉眼病型では混合型 (IIc+IIa), 占居部位ではA領域のものに頻度が高い傾向がみられた.(3) sm胃癌再発死亡5例中4例は脈管侵襲陽性のsm2~3例であり, 再発様式は肝転移2例, 腹膜再発, リンパ節再発, 不明が各1例ずつみられた.早期胃癌の術式や術後補助療法の決定に当っては, sm層への浸潤程度をも考慮する必要性があると推察された.
