Selective Eosinophil Adhesion to Fibroblast Via IFN-γ-Induced Galectin-9

Among galectin family members, galectin-9 was first described as a potent eosinophil chemoattractant derived from Ag-stimulated T cells. In the present study a role of galectin-9 in the interaction between eosinophils and fibroblasts was investigated using a human lung fibroblast cell line, HFL-1. RT-PCR, real-time PCR, and Western blot analyses revealed that both galectin-9 mRNA and protein in HFL-1 cells were up-regulated by IFN-γ stimulation. On the one hand, IL-4, known as a Th2 cytokine, did not affect the galectin-9 expression in HFL-1 cells. We further confirmed that IFN-γ up-regulated the expression of galectin-9 in primary human dermal fibroblasts. Flow cytometric analysis revealed that IFN-γ up-regulated surface galectin-9 expression on HFL-1 cells. Stimulation of HFL-1 cells with IFN-γ up-regulated adhesion of eosinophils, but not neutrophils, to HFL-1 cells. This adherence of eosinophils to HFL-1 cells was inhibited by both lactose and anti-galectin-9 Ab. These findings demonstrate that IFN-γ-induced galectin-9 expression in fibroblasts mediates eosinophil adhesion to the cells, suggesting a crucial role of galectin-9 in IFN-γ-stimulated fibroblasts as a physiological modulator at the inflammatory sites.

Benign osteoblastoma of the temporal bone.

We investigated a 27-year-old woman with a retroauricular tumour. MRI revealed a signal void on T1- and T2-weighted images, and irregular enhancement with gadolinium-DTPA. The underlying dura mater also enhanced. The tumour was resected completely, and histological examination showed a benign osteoblastoma.

Impact of Incidental Irradiation on Clinically Uninvolved Nodal Regions in Patients With Advanced Non-Small-Cell Lung Cancer Treated With Involved-Field Radiation Therapy: Does Incidental Irradiation Contribute to the Low Incidence of Elective Nodal Failure?

PURPOSE To evaluate the incidental irradiation dose to elective nodal regions in the treatment of advanced non-small-cell lung cancer with involved-field radiation therapy (IF-RT) and the pattern of elective nodal failure (ENF). METHODS AND MATERIALS Fifty patients with advanced non-small-cell lung cancer, who received IF-RT at Kagawa University were enrolled. To evaluate the dose of incidental irradiation, we delineated nodal regions with a Japanese map and the American Thoracic Society map (levels 1-11) in each patient retrospectively and calculated the dose parameters such as mean dose, D95, and V95 (40 Gy as the prescribed dose of elective nodal irradiation). RESULTS Using the Japanese map, the median mean dose was more than 40 Gy in most of the nodal regions, except at levels 1, 3, and 7. In particular, each dosimetric parameter of level 1 was significantly lower than those at other levels, and each dosimetric parameter of levels 10 to 11 ipsilateral (11I) was significantly higher than those in other nodal regions. Using the American Thoracic Society map, basically, the results were similar to those of the Japanese map. ENF was observed in 4 patients (8%), five nodal regions, and no mean dose to the nodal region exceeded 40 Gy. On the Japanese map, each parameter of these five nodal region was significantly lower than those of the other nodal regions. CONCLUSIONS These results show that a high dose of incidental irradiation may contribute to the low incidence of ENF in patients who have received IF-RT.

Usefulness of FDG-PET imaging for the radiotherapy treatment planning of pyothorax-associated lymphoma

Pyothorax-associated lymphoma (PAL) is a non-Hodgkin’s lymphoma developing in the pleural cavity after a long-standing history of chronic pyothorax (CP). F-18 fluorodeoxyglucose positron emission tomography (FDG-PET) imaging is a useful modality for determination of disease extent of various malignant tumors, including malignant lymphoma, but there have been no reports describing the usefulness of FDG-PET imaging in PAL. Here we report a case of PAL that relapsed after chemotherapy and was successfully treated by radiotherapy. FDG-PET imaging revealed that the tumor was localized to a soft-tissue attenuation mass behind the CP cavity in the right thorax, but did not infiltrate the CP cavity. A total dose of 40 Gy was administered to the area that included the PET-positive lesion, instead of including the entire CP cavity in the radiation field. Although computed tomography (CT) showed a residual mass, no FDG uptake was indicated by FDG-PET imaging performed just after the end of radiotherapy, and additional irradiation was not performed. No sign of relapse was found by FDG-PET imaging 3 months later. FDG-PET imaging was useful for both the planning of radiotherapy and assessing the treatment response of PAL.

Development of Submillimeter Wave Catheter Transmitting a Gyrotron Output for Irradiation on Living Bodies

The development of a millimeter and submillimeter wave catheter for irradiation on living bodies using a gyrotron as the radiation power source is described. The GYROTRON FU-IV, optimized for such applications was used in the development. It was operated in both CW and pulsed regime at TE03 and TE32 modes with frequencies 302 GHz and 238 GHz respectively. Irradiation tests were made on thermal papers, beefs and liver of living rats.

Intraoperative radiotherapy for gliomas

SummaryIntraoperative radiotherapy (IORT) was performed in 20 of 36 patients with glioma; 11 glioblastomas, 7 malignant astrocytomas, 2 benign astrocytomas. Twenty or 25 Gy of irradiation was delivered in a single fraction intraoperatively, followed by external beam irradiation. The electron beam energy was selected so that the 80% isodose line fell at 2 or 3 cm below the residual tumor surface. Median survival time of IORT group was 14 months and that of the control group was 10 months. Difference of survival curve was significant. There were 6 incidences of complication caused by IORT; 1 radionecrosis, 1 convulsion, 1 abscess, and 3 severe brain edemas. IORT is suited for the treatment of malignant gliomas.

CT appearance of interstitial pulmonary emphysema.

Interstitial pulmonary emphysema (IPE) is the initial manifestation of ventilator-induced barotrauma. We report two patients in whom computed tomography was useful in showing IPE.

Evaluation of radiotherapeutic response in non-small cell lung cancer patients by technetium-99m MIBT and thallium-201 chloride SPET

Abstract.The purpose of this study was to investigate the relationship between technetium-99m hexakis-2-methoxyisobutylisonitrile (99mTc-MIBI) accumulation in tumours and response to radiotherapy in non-small cell lung cancer patients in comparison with the findings obtained using thallium-201 chloride (201Tl).Simultaneous dual single-photon emission tomography (SPET) imaging with 600 MBq 99mTc-MIBI and 111 MBq 201Tl was performed in 31 patients with biopsy- or sputum cytology-proven lung cancer. SPET images were acquired 15 min (early) and 2 h (delayed) after injection, and the early ratio, delayed ratio and retention index were measured. The tumours were classified into two groups on the basis of follow-up computed tomography (CT): responders (at least 50% reduction in tumour size) and non-responders (little or no change in tumour size).The mean (± SD) values of early ratio, delayed ratio and retention index using 99mTc-MIBI SPET were 3.0±1.1, 2.7±1.0 and –9.5±12.7, respectively, in responders and 2.4±0.7, 2.0±0.5 and –18.4±9.0, respectively, in non-responders. The corresponding values using 201Tl chloride SPET were 3.7±1.0, 4.7±1.5 and 24.2±22.1 in responders and 3.3±1.2, 4.0±1.3 and 20.4±20.5 in non-responders. Using 99mTc-MIBI, the delayed ratio and retention index in responders were significantly higher than those in non-responders (P<0.01 and P<0.05, respectively). The results of this study indicate that patients with higher delayed ratio and retention index values using 99mTc-MIBI SPET are likely to respond better to radiotherapy than those with lower values. 99mTc-MIBI SPET may give an indication of the short-term response to radiotherapy in patients with non-small cell lung cancer.

Submillimeter Wave Irradiation of Living Bodies using a Gyrotron and a Catheter

The development of a submillimeter wave catheter for irradiating on living bodies using a gyrotron as the radiation power source is described. The gyrotron FU-IV is optimized for development. The gyrotron is operated in the CW mode at a frequency of 302 GHz under a magnetic field of approximately 11 T. Irradiation tests using various antennas were performed on cow livers, living rats and malignant tumors implanted in living mice. Irradiation results were considered based on the microwave theory and ray optics.

Usefulness of Technetium-99m Human Serum Albumin Lymphoscintigraphy in Chyluria

Chyluria is a urologic manifestation of a lymphatic system disease abnormality and leads to nutritional deficiency. The case of a patient with chyluria is presented, and the results of lymphoscintigraphy with those of contrast lymphangiography are compared. Lymphoscintigraphy very clearly showed the site of the fistulae and was as precise as lymphangiography. Follow-up lymphoscintigraphy 2 months after therapy revealed no radionuclide accumulation. Lymphoscintigraphy is a useful, noninvasive, safe, and simple technique for the diagnosis and follow-up of chyluria.
