Recombinant Human Epidermal Growth Factor Accelerates Recovery of Mouse Small Intestinal Mucosa After Radiation Damage

PURPOSE To determine whether systemically administered recombinant human epidermal growth factor (rhEGF) accelerates the recovery of mouse small intestinal mucosa after irradiation. METHODS AND MATERIALS A mouse mucosal damage model was established by administering radiation to male BALB/c mice with a single dose of 15 Gy applied to the abdomen. After irradiation, rhEGF was administered subcutaneously at various doses (0.04, 0.2, 1.0, and 5.0 mg/kg/day) eight times at 2- to 3-day intervals. The evaluation methods included histologic changes of small intestinal mucosa, change in body weight, frequency of diarrhea, and survival rate. RESULTS The recovery of small intestinal mucosa after irradiation was significantly improved in the mice treated with a high dose of rhEGF. In the mice that underwent irradiation without rhEGF treatment, intestinal mucosal ulceration, mucosal layer damage, and severe inflammation occurred. The regeneration of villi was noticeable in mice treated with more than 0.2 mg/kg rhEGF, and the villi recovered fully in mice given more than 1 mg/kg rhEGF. The frequency of diarrhea persisting for more than 3 days was significantly greater in the radiation control group than in the rhEGF-treated groups. CONCLUSIONS Systemic administration of rhEGF accelerates recovery from mucosal damage induced by irradiation. We suggest that rhEGF treatment shows promise for the reduction of small intestinal damage after irradiation.

Mucoepidermoid Carcinoma of the Thymus: A Case Report

Mucoepidermoid carcinoma of the thymus is an extremely rare malignant mediastinal neoplasm, and to our knowledge, only 13 cases have been reported. We report a case of mucoepidermoid carcinoma of the thymus that was seen in a 53-yr-old man with right chest pain. Chest CT scan showed a huge, cystic mass having a focal solid portion with direct invasion of the adjacent anterior chest wall and pericardium in the anterior mediastinum. Mucoepidermoid carcinoma of the thymus should be included in the differential diagnosis for masses of the anterior mediastinum associated with extensive cystic changes, although the carcinoma is exceedingly rare.

Acute cholecystitis associated with Clonorchis sinensis infection

Clonorchis sinensis is one of the most common causes of trematodiasis that is caused by the ingestion of raw fish contaminated with infective cysts. The adult flukes are predominantly present in the intrahepatic bile ducts, but occasionally they may be found in the pancreatic duct and extrahepatic bile ducts. The clinical manifestations depend on the number of flukes, the period of infestation, and complications such as pericholangitic abscess, cholangitis, bile duct stones, and cholangiocarcinoma. However, primary acute cholecystitis associated with C. sinensis infection is extremely rare. Herein, we report on a case of primary acute cholecystitis associated with C. sinensis infection.

Histological Assessment of Mechanoreceptors in Achilles Allografts After Anterior Cruciate Ligament Reconstruction

Background: There is a lack of histological studies investigating the presence of mechanoreceptors in anterior cruciate ligament (ACL) allografts. Hypothesis: Mechanoreceptors would not grow in Achilles allografts after ACL reconstruction. Study Design: Case series study; Level of evidence, 4. Methods: Tissue samples were obtained from 11 patients who underwent ACL reconstruction using Achilles tendon allografts. They underwent biopsies during second-look arthroscopies. The mean period from ACL reconstruction to harvesting tissue was 26.63 months (range, 12-120 months). The control group consisted of 2 normal ACLs procured from 42- and 45-year-old men who underwent amputation above the knee due to trauma. Results: Ruffini corpuscles and free nerve endings were shown to be present in the specimens of the control group by processing hematoxylin-eosin stains and immunohistochemical stains with monoclonal antibodies against S-100. In the Achilles allografts, mechanoreceptors were not observed. However, fibroblasts, collagen fibers, and vessels that were not present in fresh-frozen Achilles allografts before surgery were observed. Conclusion: The results demonstrate that Achilles tendon allografts appeared similar to normal ligaments except for the lack of histological evidence of mechanoreceptors. In other words, there are no newly ingrown mechanoreceptors in ACL allografts.

Primary adenosquamous carcinoma of the colon.

Adenosquamous cell carcinoma (Ad-SCC) of the colon is rare. The pathogenesis of Ad-SCC is unclear, however, several hypotheses have been suggested. The clinical presentation and gross findings of Ad-SCC of the colon are similar to those of adenocarcinoma of the colon, but Ad-SCC has a more aggressive clinical course and a poorer prognosis. We report on two cases of Ad-SCC of the colon with obstruction; a collision-type Ad-SCC that has not only obstruction but also numerous hepatic metastases, and a composite-type Ad-SCC treated with left hemicolectomy followed by an adjuvant chemotherapy.

Synchronous granular cell tumors in the perianus and chest wall

Granular cell tumor (GCT) is a rare tumor that originates from the Schwann cells in the skin, soft tissues, and internal organs. Usually, GCTs are clinically benign, although malignant and multifocal forms are very rarely known to occur. Cases of GCT of the perianus are rare, and thus far, no study has reported synchronous GCTs of the perianus and the chest wall. We report a case of a 31-year-old woman with synchronous GCTs of the perianus and the chest wall to have a mind of consideration of the possibility of GCT in the differential diagnosis of perianal tumor.

Intramuscular ganglion of the quadriceps femoris.

Ganglion cysts are common lesions that are most often found around the joints of the hands and feet. Ganglia around the distal femur usually occur within the synovial membrane or tendon sheath, but rarely within muscles. Several cases of intramuscular ganglions in the hand and wrist have been reported, but a ganglion cyst in the quadriceps muscle has rarely been addressed in studies. In this report, we present a 17-year-old patient with a painful movable mass in the intramuscular area of the quadriceps femoris that was diagnosed by ultrasound and treated by excision and biopsy.

Superficial Fibromatosis Mimicking Glomus Tumor of the Second Toe

Various types of tumor can occur in the subungual space, including glomus tumors, subungual exostosis, hemangioma, epidermal cysts, and malignant tumors. While fibromatosis can occur at various sites throughout the body, it is very rarely seen in the toe. Here, we are the first to report a case of superficial fibromatosis mimicking a glomus tumor in the subungual space of the second toe. The presentation of this condition shows the possibility of encountering uncommon superficial fibromatosis in the distal phalanx of the toe, and suggests that superficial fibromatosis should be included in the differential diagnosis of a glomus tumor in the toe.

Abstract 1043: Autophagy induces cytoprotection in cisplatin-treated lung cancer cells

Proceedings: AACR 101st Annual Meeting 2010‐‐ Apr 17‐21, 2010; Washington, DC Purpose: Most of lung cancer patients had received the systemic chemotherapy because they had diagnosed the advanced stage at presentation. Cislplatin-based chemotherapy is main regimen for advanced lung cancer. But serious adverse reaction (SAE) had occurred after chemotherapy. This is main reason for chemotherapy withdrawal. If we will use the reduced dose of cispaltin, cisplatin-based the chemotheray will be continued. To figure out whether the effect of low dose of cisplatin will be synergistically enhance by autophagy inhibiton, even though the reduced dose of cisplatin less effect for treatment response, We investigated that role of autophagy by 3-methyladenine, class III PI3kinase inhibitor in cisplatin-treated lung cancer cells. Methods: Lung cancer cells were incubated with normal media and treated the 5uM (LD20) and 20uM(LD50) of cislpaltin according to time dependent. We examined the sensitivity to low or high doses of cisplatin treatment using the MTT cell viability assay and compared apoptosis and autophagy by nuclear stianing, apoptotic or autophagic related proteins or autophagic vacuoles. autophagy is also displayed the development of acidic vascular organelles by acridine orange and the fluorescent expression of GFP-LC3 protein in the GFP-LC3 transfected cells. Result: Low dose of cisplatin-treated lung cancer cells is relatively less sensitive to cell death rather than high dose treated cells in a time-dependent manners. We didnot find out the nuclear fragmentation in lower dose even though detected its in high dose. PARP cleavages also were not detected in low dose of cisplatin exception 24H. We observed that massive vacuolization in the cytoplasm were seen at lower dose treated cells. Acridine orange stain-positive cells also were increased according to time dependent. We figured out that the autophagosome-incorporated LC3 II protein expression more increased in low dose treated cells (24H to 48H) in spite of no detection of LC3 II in high dose treated cells. The expression of GFP-LC3 were increased in 5uM treated cells by time-dependent manner. When 3-methyladenine, class III PI3kinase inhibitor, was pretreated in 5uM cisplatin-treated lung cancer cells, cell survival significantly were decreased. Conclusion: Autophagy induces cytoprotection in lower dose of cisplatin-treated lung cancer cells. Citation Format: {Authors}. {Abstract title} [abstract].
