Elemental content of some anti-diabetic ethnomedicinal species of genus Ficus Linn. using atomic absorption spectrophotometry technique

Elemental investigation of some selected species of genusxa0Ficusxa0(Moraceae) having anti-diabetic property was conducted using atomic absorption spectrophotometry technique. The flame atomic absorption spectroscopy was employed for the estimation of element contents of different plant parts of seven different species includingxa0Ficus bengalensis,xa0Ficus religiosa,Ficus microcarpa,xa0Ficus racemosa,xa0Ficus hispida,xa0Ficus caricaxa0andxa0Ficus lacorxa0content for clinical monitoring. Na, K, Cu, Zn, Cr, Ca, Mn, Fe, Ni, Mg, Cd, Co and Pb were detected and their concentration determined. The results of the present work give the proper reasonable information for the utilization of following plants in the treatment of diabetes through ethnomedicinal as well as AAS studies. These medicinal plants were can be considered as potential sources for providing a reasonable amount of the required elements to the diabetic patients in a particular way, as they contain appreciable amounts of the certain elements which are capable for influence of insulin. Effect of seasonal variations on the haematochemical profile of Cholistani service bulls

ABSTRACT This reported study is of Pakistans indigenous Cholistani cattle breed, a native of the Cholistan desert where it has been reported as having high production potential for nomadic herders. The aim of the present study was to assess haematochemical profile of adult Cholistani service bulls (nu2009=u200916), being reared at the Semen Production Unit Karaniwala, Bahawalpur, Cholistan desert, Pakistan, during various seasons as an indicator of their health status. Keeping in view the prevailing climatic conditions of the study area, four seasons of three months duration each were defined as (i) cool dry winter (November, December, January) (ii) temperate spring (February, March, April) (iii) hot dry summer (May, June, July) and (iv) hot humid summer (August, September, October). Blood specimens were collected fortnightly during the study period for haematochemical analyses (automated). The present study revealed that Cholistani service bulls have a tendency to maintain most of their haematochemical parameters more or less at the same level during various seasons which is suggestive of their adaptability under stressful climates without showing any signs of stress.

Serum trace mineral variations in Nili-Ravi buffaloes suffering with prepartum vaginal prolapse in two different agro-ecological zones of Punjab, Pakistan

The present study was conducted during 2005 and 2006 on 200 Nili-Ravi buffaloes kept in two agroecological zones (irrigated [zone 1] and rain-fed [zone-2]) of Punjab, Pakistan, with the objective to determine the level of trace minerals (Cu, Fe, Zn, Se) in serum of the buffaloes suffering from vaginal prolapse and to compare them with their healthy counterparts. In each zone 50 buffaloes suffering from prepartum vaginal prolapse during their seventh month of gestation were identified through survey. Vaginal prolapse-affected buffaloes belonging to zone 1 were identified as group VPB1 (N = 50), whereas buffaloes belonging to zone 2 were recognized as VPB2 (N = 50). The buffaloes of control group in zone 1 and zone 2 were identified as NCB1 and NCB2, respectively. The blood samples in all four groups of buffaloes were collected three times, i.e., first when these animals were in the eighth month of gestation, second during the eighth to ninth month of gestation, and finally when these animals were in the ninth or later month of gestation. The mean serum copper concentrations in buffaloes of group VPB1 were significantly lower (P < 0.05) in comparison with NCB1 and NCB2, whereas there were nonsignificant differences (P > 0.05) in copper concentrations between VPB1 and VPB2. There was a significant difference (P < 0.05) of iron concentration in VPB1 compared with NCB1 and NCB2. Similarly, VPB2 also had significantly lower (P < 0.05) iron concentrations compared with NCB1 and NCB2. Serum zinc concentrations were significantly lower (P < 0.05) in animals of the VPB1 group when compared with NCB1 and NCB2. Similarly, lower zinc concentrations were observed in VPB2 in comparison with NCB1 and NCB2. There was significantly lower (P < 0.05) zinc concentration in affected buffaloes (VPB1 and VPB2) from the ninth month of gestation to term when compared with those in the eighth to ninth mo of gestation, and with those not yet in the eighth month of gestation. Serum selenium concentration were significantly higher (P < 0.05) in control group buffaloes (NCB1 and NCB2) in comparison with vaginal prolapse-affected buffaloes (VPB1 and VPB2). During different stages of gestation, mean serum selenium concentrations varied nonsignificantly (P > 0.05) within each group of buffalo. Based on information obtained from this study, it was concluded that the low serum concentration of copper and selenium are linked to increased incidence of vaginal prolapse in buffaloes during the last trimester of gestation.

Pathological effects of concurrent administration of aflatoxin B 1 and fowl adenovirus-4 in broiler chicks

The current study was designed to investigate pathological effects of fowl adenovirus in broilers exposed to aflatoxin B1. Fowl Adenovirus-4 (FAdV-4) infection is remerging in all types of poultry birds in Pakistan. Poultry feed contamination with mycotoxin (aflatoxin) is another important global issue. A total of 125-day old broiler birds were divided into six equal groups. Group A served as control. B and C groups were administered with aflatoxin B1 (AFB1) 100 and 200u202fμg/kg feed. Group D was infected with FAdV-4, while groups E and F administered with both AFB1 (100 & 200xa0μg/kg) along with FAdV-4. These birds were monitored for clinical signs and mortality. Feed intake, body weight (BW), relative organ weights and gross & histopathological lesions were recorded. The highest mortality was observed in group F (FAdV-4xa0+xa0AFB1 200u202fμg/kg feed) and the lowest mortality was observed in group B (AFB1 100u202fμg/kg feed). Body weights of all the groups were significantly (pu202f<u202f0.05) lower as compared with control group. Relative weight of liver and kidneys in groups E and F were significantly higher as compared with control. Grossly, liver was swollen, anemic with round margins in groups D, E and F. Kidneys were also swollen with whitish areas indicating dead tissue. Microscopically intranuclear inclusion bodies were observed in group D-F. The hepatic parenchyma was also indicating necrotic changes along with vacuolar degeneration. In renal parenchyma, acute tubular necrosis was observed in groups C, E and F. It was concluded that AFB1 intoxication lead to dose dependent changes in liver and kidneys. Severity of the changes was increased in interactive groups of AFB1 with FAdV-4. Therefore, feed should be regularly monitored for AFB1 levels and day old chicks for vertically transmitted FAdV-4 to prevent losses.

Correlation of biochemical constituents of seminal plasma with semen quality in Teddy goat (Capra hircus) bucks

This study was planned to determine the relationship between semen quality parameters and the levels of biochemical constituents of seminal plasma of Teddy (Capra hircus) buck semen. For this purpose, semen ejaculates were collected from five mature healthy Teddy bucks. All the experimental bucks were kept under natural environmental conditions. Semen was collected twice in a week for the duration of 6 weeks by Artificial Vagina (AV) in the breeding season (February‐April). Two successive ejaculates of single buck were pooled at time of collection, and a total of 60 semen samples were processed for semen analysis. Sperm per cent motility, sperm concentration, dead sperm percentage, morphological abnormal spermatozoa, plasma membrane integrity were correlated with biochemical constituents of seminal plasma. The mean per cent motility (89.18% ± 0.37%), sperm concentration (1.86 ± 0.04 × 109/ml), dead sperm percentage (8.08% ± 0.29%), morphological abnormal spermatozoa (6.05% ± 0.29%) and plasma membrane integrity (88.22% ± 0.34%) were recorded. The seminal plasma contained Na+ (144.12 ± 1.59 mEq/L), K+ (27.38 ± 0.49 mEq/L), Cl− (65.73 ± 0.45 mEq/L), Ca++ (9.34 ± 0.22 mg/dl), P (19.32 ± 0.97 mg/dl), aspartate aminotransferase (AST; 26.48 ± 1.30 IU/L), alanine aminotransferase (ALT; 168.47 ± 5.18 IU/L), lactate dehydrogenase (LDH; 215.98 ± 6.06 IU/L), albumin (1.90 ± 0.10 g/dl), globulins (2.08 ± 0.11 g/dl) and total protein (3.98 ± 0.20 g/dl). The collected data were analysed by applying Pearsons correlation coefficients. Dead sperm percentage had negative correlation with sodium (r = −.278, p < .05), albumin (r = −.294, p < .05), globulin (r = −.266, p < .05) and total protein (r = −.295, p < .05). Phosphorus was negatively associated with sperm concentration (r = −.262, p < .05). AST was negatively correlated with plasma membrane integrity (r = −.292, p < .05). It was concluded that most of the semen quality parameters of Teddy bucks were positively correlated with biochemical constituents, but opposite trends were found in case of dead sperm percentage. The seminal biochemical constituents dynamically interact with each other.

Effects of FSH extracted from in vitro cultured anterior pituitary cells of male buffalo calves on body and testes weight, serum FSH and total cholesterol and hematological variables in male rabbits

In this study, anterior pituitary glands were collected from 12 young male buffalo calves after slaughter, cultured with gonadotropin releasing hormone (GnRH) and estrogen stimulus and the extract obtained. Adult male rabbits (n = 15) were divided into three equal groups. Rabbits of Group A served as control; those of Groups B and C were given extract containing 4 and 8 mIU of follicle stimulating hormone (FSH), respectively twice daily for 3 weeks. Body weight of rabbits was recorded before and after treatment; blood samples were collected after treatment and analyzed for hemoglobin (Hb), red blood cell (RBC) count, white blood cell (WBC) count, packed cell volume (PCV), platelet counts, mean corpuscular volume (MCV) and mean corpuscular hemoglobin (MCH), while serum samples were analyzed for FSH and total cholesterol. Then, all rabbits were slaughtered, and weight of paired testes was recorded. Results showed that the values for weight gain, RBC count, WBC count, PCV and MCH did not differ among rabbits of three groups. Blood Hb was greater (P < 0.05) in rabbits of Group B than Group C. Testis weight, serum FSH, total cholesterol and blood platelets count were greater in rabbits of Groups B and C, while MCV was less in rabbits of Group C, compared to the control (P < 0.05). In conclusion, in vitro cultured cells of adenohypophysis from male buffalo calves showed FSH activity. This FSH increased testes size, serum FSH, total cholesterol and blood platelets counts and decreased MCV in rabbits. However, it had no effect on weight gain, RBC counts, WBC counts, PCV and MCH.

Genetic assessment of the genus Pisum L. based on sequence specific amplification polymorphism data

The present study was conducted to analyze genetic diversity among 56 accessions of Pisum sativum, Pisum elatius, Pisum pumilio, Pisum fulvum and Podandrogyne brevipedunculata from Syria (42) and a set of 16 reference accessions from Israel (3), Ethiopia (2), Greece (2), Italy (2), Turkey (3), Tunisia (1) and Georgia (1). The study was based on the analysis of multiple data sets generated using sequence specific amplification polymorphism (SSAP) and associated morphological and ecogeographic data. The diversity analysis based on SSAP markers resulted in 83 scorable fragments in P. sativum, P. elatius (63), P. fulvum (62), P. pumilio (53), P. brevipedunculata (31) and P. abyssinicum (29). Genetic variability was measured as Nei’s gene diversity and maximum polymorphism was found in P. elatius (33.87%) and P. fulvum (32.74%) whereas minimum diversity was observed in P. brevipedunculata (12.48%). Pair wise band sharing among Pisum species was also calculated, significant band sharing was observed between P. fulvum and P. elatius, P. elatius and P. sativum and P. pumilio and P. elatius. The relationship among species as revealed by SSAP data could not significantly be correlated with those based on the agro-morphologic characters, suggesting that the two inferences are independent and estimates of genetic relations differently among Pisum taxa. The ecogeographic data associated with Pisum species (latitude, longitude) were also plotted using Arc View GIS 3.2 in order to show the distribution of species in Syria.
